Project description:Diabetic retinopathy is one of the leading causes of blindness in diabetic patients. Emerging evidence suggests that retinal neurodegeneration is an early event in the pathogenesis of diabetic retinopathy, but the underlying causes of neuronal loss are unknown. To unravel potential mechanisms underlying early retinal neurodegeneration in diabetic retinopathy, a gene expression profiling study was undertaken to compare the gene expression in retinas of 8-week db/db diabetic mice with that of lean non-diabetic littermates. Retinas were obtained from 8-week db/db diabetic mice and age-matched lean non-diabetic controls. Total RNA was extracted and processed for being hybridized onto affymetrix DNA microarrays.

Here, we found significant changes of lncRNAs in the livers of db/db mice. Among them, abnormal expression of ultraconserved elements was noticed.

Products differentially secreted from perigonadal adipose tissue of db/db mice.

Project description:A control group of male C57BL/6J mice were fed a 60% fat diet (Research Diets D12492i) for 8 weeks, resulting in “ad libitum obesity” (ALO). Leptin receptor-deficient B6.BKS(D)-Leprdb/J (“db/db”) male mice were fed a low-fat chow diet (PicoLab Rodent Diet 20; Purina Mills Inc). Once the average body weights of the two groups were equivalent, all mice were fasted for 24 hours, after which they were sacrificed and perigonadal adipose tissue (PGAT) was dissected out. PGAT was cultured ex vivo for 6 hours, after which conditioned media was collected and submitted for proteomic profiling. At the time of sacrifice, ALO mice were 17 weeks old and db/db mice were 10 weeks old. “HFD” is also used to refer to the ALO condition while “DB” is used to refer to the “db/db” condition.

Quantitative proteomic analysis of liver proteins from WT and db/db mice

Project description:Liver is a key organ which controls glucose and lipid homeostasis. Dysregulation of hepatic glucose and lipid metabolism causes metabolic diseases, including type 2 diabetes (T2D) and metabolic-associated liver diseases (MAFLD). However, how glucose and lipid homeostasis are integratively regulated is not fully understood. The key factors that drive the pathogenesis of metabolic diseases need to be identified. To identify these regulators, we performed quantitative proteomic analysis in the livers of wide type and db/db mice (n=4 for each group). The quantitative proteomic analysis showed that 1233 proteins were upregulated and 2724 proteins were downregulated in the livers of db/db mice. These differentially regulated proteins contribute to the pathogenesis of metabolic diseases.
