Project description:With the growing antibiotic resistance of Staphylococcus aureus, it is urgent to discover new antimicrobials to solve this problem. By bacterial growth based high-throughput screening, we identified two compounds, C218-0546 and STK848198, with effective antimicrobial activity against S. aureus. This study aims to explore the model of action of the compounds by transcriptomic analysis and other microbiological methods. The transcriptomic data would provide us information about the regulated genes, and hence help us further study the underlying antimicrobial mechanisms by the compounds.

Project description:Staphylococcus aureus is a leading cause of hospital-associated infections. In addition, highly virulent strains of methicillin-resistant S. aureus (MRSA) are currently spreading outside health care settings. Survival in the human host is largely defined by the ability of S. aureus to resist mechanisms of innate host defense, of which antimicrobial peptides form a key part especially on epithelia and in neutrophil phagosomes. Here we demonstrate that the antimicrobial-peptide sensing system aps of the standard community-associated MRSA strain MW2 controls resistance to cationic antimicrobial peptides. The core of aps-controlled resistance mechanisms comprised the D-alanylation of teichoic acids (dlt operon), the incorporation of cationic lysyl-phosphatidylglycerol (L-PG) in the bacterial membrane (mprF), and the vraF/vraG putative antimicrobial peptide transporter. Further, the observed increased production of L-PG under the influence of cationic antimicrobial peptides was accompanied by the up-regulation of lysine biosynthesis. In noticeable difference to the aps system of S. epidermidis, only selected antimicrobial peptides strongly induced the aps response. Heterologous complementation with the S. epidermidis apsS gene indicated that this is likely caused by differences in the short extracellular loop of ApsS that interacts with the inducing antimicrobial peptide. Our study shows that the antimicrobial peptide sensor system aps is functional in the important human pathogen S. aureus, significant interspecies differences exist in the induction of the aps gene regulatory response, and aps inducibility is clearly distinguishable from effectiveness towards a given antimicrobial peptide. Project description:Multi-drug resistant Staphylococcus aureus (S. aureus) infections continously threaten public health. The rapid escalation in morbidity and mortality rates associated with methicillin-resistant S. aureus (MRSA) infections necessitates the urgent development of novel antimicrobial agents. Our study reveals that the FDA-approved drug cinacalcet (CNA) effectively functions as an antibacterial and anti-biofilm agent against S. aureus without detectable resistance. It evidently improved survival rate of mice infected with clinical multi-resistant S. aureus in a pneumonia model. Subsequent proteomic and biochemical experiments suggest that the primary antibacterial mechanism involves membrane structure disruption, ATP content reduction, and reactive oxygen species (ROS) production. Concurrently, LiP-SMap combined with biochemical validation indicates that CNA inhibits biofilm formation by targeting IcaR, a negative regulator of icaADBC, thereby enhancing its binding capacity to the ica operator DNA and subsequently suppressing extracellular polysaccharide formation. Importantly, compared to vancomycin, CNA demonstrates stronger biofilm bacterial clearance in a mouse thigh infection model. Collectively, our findings propose that CNA can be repurposed as a potential therapeutic agent for treating multidrug-resistant S. aureus infections and their associated biofilms.

Project description:Mesenchymal Stromal Cells (MSCs) are widely studied in regenerative medicine because of their unique biological properties. The MSC-derived secretome is a cell-free product containing soluble factors and extracellular vesicles that mediate many of the biological properties of MSCs, including immunomodulatory and antimicrobial activities. This study aimed to evaluate the antimicrobial activity of a freeze-dried secretome derived from canine adipose tissue MSCs (canine lyosecretome, c-Lyo) against pathogens relevant to canine infections. The antimicrobial activity of c-Lyo was assessed using minimal inhibitory concentration (MIC) assays against six bacterial species (Escherichia coli, Salmonella enterica subsp. enterica serovar Typhimurium, Staphylococcus aureus, methicillin-resistant Staphylococcus aureus (MRSA), Staphylococcus pseudintermedius and Pseudomonas aeruginosa) and one yeast (Malassezia pachydermatis). Potential interactions between c-Lyo and commonly used veterinary antimicrobials (gentamicin, amoxicillin, enrofloxacin, and ketoconazole) were evaluated using checkerboard assays (CkA). Proteomic characterization of the c-Lyo and transcriptomic analysis of canine MSCs were also conducted to explore molecular mechanisms underlying antimicrobial activity. The results demonstrated that c-Lyo exerts antimicrobial activity against all tested microorganisms, with variable sensitivity among species. CkA exhibited predominantly additive interactions with conventional antimicrobials and occasional synergistic effects, with no evidence of antagonism. Proteomic and transcriptomic analyses supported the presence of antimicrobial peptides and immune-related pathways that potentially contribute to both direct antimicrobial activity and the modulation of host defense mechanisms. These findings indicate that c-Lyo may represent a promising cell-free biologic approach for antimicrobial strategies in veterinary medicine.
