Project description:BCG therapy is the most effective immunotherapy for bladder cancer, but it sometimes causes serious adverse events because of infection with live BCG bacteria. To develop more effective and less toxic immunotherapeutic agents, we have developed formulations using BCG cell wall components. Trehalose dimycolate (TDM) is a predominant glycolipid that constitutes the cell wall of BCG, and reported to show activation of innate immunity and have adjuvant effect. We already established hydrophilic cationic liposomes incorporating TDM, and reported that local administration of the cationic liposome TDM (Lip-TDM) exerted antitumor effect to subcutaneously inoculated tumors by inducing activation of CD8+ T cells via dendritic cell maturation. In present study, we demonstrated that intraperitoneal administration of Lip-TDM exerted antitumor effects in orthotopic bladder carcinogenesis mouse model induced by N-butyl-N-(4-hydroxybutyl) nitrosamine (BBN). In this model, intraperitoneal administration of Lip-TDM enhanced the systemic activation of dendric cells and macrophages, and increased infiltration of adaptive immune cells such as CD8+ T cells. Furthermore, Lip-TDM treatment induced Natural Killer (NK) cells to be cytotoxic NK cells in tumors and regulatory NK cells in lymphoid tissues. Lip-TDM treatment induced activation of DCs and cytotoxic CD8+ T cells and exerted antitumor effect in WT mice, but not in NK cell-depleted mice. These results indicate NK cells are essential for Lip-TDM to induce sufficient acquired immune response. Our findings elucidate the mechanism of the antitumor effect by Lip-TDM and suggest that Lip-TDM is a non-infectious agent which can be administered intraperitoneally and has potential to be an alternative to BCG.

Project description:Bladder cancer (BLCA) is a highly prevalent malignancy on a global scale, characterized by significant tumor heterogeneity. The investigation of molecular mechanisms that can be leveraged for the management of aggressive BLCA represents a particularly pertinent objective.DLGAP5 is mitosis-related protein widely existed in eukaryotic cells. The molecular mechanism of DLGAP5 in bladder cancer is still not clear, despite its role in tumorigenesis. Bladder cancer tissues exhibit a noteworthy increase in DLGAP5 expression, compared to normal bladder tissues. Furthermore, the expression of DLGAP5 shows a positive correlation with the clinical stage of the tumor and a negative impact on the prognosis. Cell proliferation and migration are significantly impacted by alterations in DLGAP5 expression, as demonstrated by in vitro and in vivo experiments. Further investigation reveals that DLGAP5 regulated E2F1 protein stability via USP11 deubiquitinase activity, and DLGAP5 is a direct target of E2F1. DLGAP5-E2F1 forms a positive feedback loop and influences the efficacy of BLCA progression. Collectively, DLGAP5 could potentially serve as a new therapeutic target for the treatment of bladder cancer.

Project description:Interstitial cystitis (IC) and bladder pain syndrome are terms used to describe a heterogenous chronic pelvic and bladder pain disorder of unknown etiology. The goal of this pilot study was to determine if gene expression profiling of bladder biopsy tissue from patients experiencing symptoms could be used to separate the patients based on some clinical parameter. Gene expression profiles in bladder biopsy tissue from patients with: (1) low bladder capacity (defined here as <400 ml upon hydordistension), (2) normal capacity (M-bM-^IM-%400 ml), and (3) controls were compared. Gene expression profiles from low bladder capacity tissues differed significantly from normal capacity and control tissue, suggesting gene expression profiling may be a useful tool for better understanding IC disease pathophysiology.

Genes encoding components of the specialized urothelial membrane, the uroplakins, were downregulated in Get1-/- mice. At least one of these genes, Uroplakin II, is a direct target of Get1. The urothelial-specific activation of the Uroplakin II gene is due to selective binding of Get1 to the Uroplakin II promoter in urothelial cells, most likely regulated by histone modifications. These results demonstrate a key role for Get1 in urothelial differentiation and barrier formation.

Project description:ATM plays a central role in the cellular response to DNA damage and ATM alterations are common in several tumor types including bladder cancer. However, the specific impact of ATM alterations on therapy response in bladder cancer is uncertain. Here, we combine preclinical modeling and clinical analyses to comprehensively define the impact of ATM alterations in bladder cancer. We show that ATM loss is sufficient to increase sensitivity to DNA damaging agents including cisplatin and radiation. Furthermore, ATM loss drives sensitivity to DNA repair targeted agents including PARP and ATR inhibitors. ATM loss alters the immune microenvironment and improves anti-PD1 response in preclinical bladder models but is not associated with improved anti-PD1/PD-L1 response in clinical cohorts. Finally, we show that ATM expression by IHC is strongly correlated with response to chemoradiotherapy. Together these data define a potential role for ATM as a predictive biomarker in bladder cancer.

Project description:To explore circular RNA (circRNA) expression profiling and their biological functions in bladder cancer, we surveyed the circRNA expression signature of 4 pairs of matched bladder cancer and para-cancer tissues from clinical patients using microarray. Hundreds of significantly changed circRNAs were identified. Our findings provide a novel perspective on transcriptional group and lay the foundation for future research of potential roles of circRNAs in bladder carcinoma.

Project description:RNASeq profiling of bladder carcinomas from different histologies (urothelial carcinomas or squamous cell carcinomas) which were collected at different moments of the Neodurvarib-related clinical procedure (pretreatment ("SCR") or post-treatment ("Q")). The main goals of the screening were 1) to define potential predictive/prognostic biomarkers (genes or molecular pathways) through the comparison of diagnostic samples ("SCR") according to the clinical response observed in the patients under study; ii) to evaluate the molecular changes observed in the tumors under study by comparing the profiles of diagnostic samples ("SCR") versus cystectomies ("Q"), which would be a direct consequence of the neoadjuvant therapy used in this clinical trial.
