Detection of STAT3 ubiquitination modification sites

ABSTRACT: Identify all the lysine sites of STAT3 that may be ubiquitinated,The search criteria were as follows: no enzyme was choosed as unspecific; two missed cleavage was allowed; carbamidomethylation (C) were set as the fixed modifications; the oxidation (M) and ubiquitination were set as the variable modification; precursor ion mass tolerances were set at 20 ppm for all MS acquired in an orbitrap mass analyzer; and the fragment ion mass tolerance was set at 0.02Da for all MS2 spectra. Confidence levels were set to 1% FDR (high confidence)

Ubiquitination, a critical post-translational modification, regulates diverse cellular processes including protein degradation, signal transduction, and cell cycle progression. Dysregulation of the ubiquitin system is closely associated with tumor development and progression. HERC4, a member of the HECT-type E3 ubiquitin ligase family, remains poorly characterized in colorectal cancer (CRC). In this study, we report that HERC4 is significantly downregulated in CRC tissues and is associated with  ...[more]
