Project description:<p _msttexthash='40138319' _msthash='351'>High ammonia nitrogen stress significantly compromises the survival of Litopenaeus vannamei under low-salinity conditions, posing a common challenge in low-salinity aquaculture. However, existing studies predominantly focus on ammonia nitrogen responses under single stressors or normal seawater salinity. The molecular regulatory mechanisms, metabolic remodeling patterns, and key pathway interactions in shrimp subjected to high ammonia-nitrogen stress under low-salinity environment remain unclear. In this study, we adopted the transcriptomics and metabolomics analysis to unveil the underlying molecular responses and metabolic biomarkers in the gills of L. vannamei to ammonia stress under low salinity condition. First, L. vannamei underwent low-salinity acclimation from 30ppt to 5ppt salinity and was then temporarily reared for one week to acclimatize to the experimental environment. Subsequently, shrimp were treated with 42.32 mg/L ammonia nitrogen for a consecutive 96h period. Integrated transcriptomic and metabolomic analyses elucidated the stress response patterns in the gills of L. vannamei under low-salinity ammonia nitrogen exposure. Specifically, 352, 802, and 140 differentially expressed genes (DEGs) were identified at 12h, 48h, and 96h post-exposure, respectively. GO and KEGG enrichment analyses revealed that the significant DEGs were primarily enriched in six pathways: autophagy, immune-related pathway, ABC transporter, fatty acid degradation and metabolism, metabolic pathway, and PPAR signaling pathway. Metabolomic profiling identified numerous differentially accumulated metabolites (DAMs) in both positive and negative ion modes, with significantly altered DAMs mainly consisting of organic acids and their derivatives, phospholipids, and other related metabolites. Key DAMs included taurine, guanosine, 1-palmitoyl-sn-glycero-3-phosphocholine, pseudouridine, and betaine. Integrative multi-omics analysis revealed that L. vannamei mediates stress responses by modulating five core pathways under low-salinity/high-ammonia-nitrogen dual stress: fatty acid degradation and metabolism (e.g., acyl-CoA dehydrogenase short chain (Acads), acetyl-CoA acetyltransferase 2 (ACAT2)), autophagy (e.g., autophagy-related protein 101-like (atg101)), immune regulation pathway (e.g., V-type proton ATPase subunit H-like, (VhaSFD), actin-5C-like (Act5C)), metabolic pathway (e.g., molybdopterin synthase catalytic subunit-like (Mocs2B), cytochrome P450 2U1-like (Cyp2b1)), and ABC transporter (e.g., ATP-binding cassette sub-family D member 3-like (ABCD3), ATP-binding cassette sub-family B member 10 (ABCB10)). Through characterization of these core pathways, this study reveals the fundamental mechanisms by which L. vannamei responds to high ammonia nitrogen stress following low-salinity acclimation, providing a theoretical foundation for estuarine shrimp farming.</p>

The assembled sequence data comprising 349,012 unique transcripts were obtained, and the established transcriptional database provided a comprehensive sequence source for this non-model organism.

Project description:Saline is a key environmental parameter in aquaculture. Research on acute salinity stress can help farmers determine the optimal salinity level to reduce stress responses in fish larvae, improve survival rates, and increase production efficiencies. It can also aid in the selection or breeding of species suited to environments with specific salinities. However, the molecular mechanisms underlying the negative effects of high salinity on Pseudobagras ussuriensis and the regulatory mechanisms underlying its tolerance remain unclear. In this study, Pseudobagras ussuriensis was exposed to 10 g/L NaCl for 96 h, and the physiological and biochemical changes in the gill and kidney tissues were continuously monitored. Transcriptomic and metabolomic analyses of the gill and kidney tissues were conducted at 24 h to elucidate the molecular adaptation mechanisms to salt stress. A total of 2,554 differentially expressed genes (DEGs) were identified in gill and 1,066 DEGs in the kidney tissue, respectively. Compared with the control group, 826 genes were upregulated and 1,728 were downregulated in the gill tissue under acute salinity stress at 24 h, whereas 508 genes were upregulated and 558 were downregulated in the kidney tissue. These DEGs were involved in oxidative stress, energy metabolism, ion transport, and immune responses. Metabolomic analysis showed that compared to the control group, 85 differential metabolites (DMs) were identified in the gill tissue, with 49 upregulated and 36 downregulated, whereas 433 DMs were identified in the kidney tissue, with 252 upregulated and 181 downregulated. Notably, the levels of phosphatidylcholine (PC) and phosphatidylethanolamine (PE) in the kidney and phosphatidic acid (PA) and PC in the gills were significantly increased, while sphingomyelin (SM) decreased. Integrated analysis of metabolomics and transcriptomics through the KGML network map revealed the downregulation of ALDH, ALDH7A1, AOX, and HADHA in the gill, and ENPP1_3, CD203, L-glutamine (downregulated), and Succinic Acid (upregulated) in the kidney. This study combined metabolomics, transcriptomics, and physiological-biochemical analyses to provide new insights into the molecular mechanisms of oxidative stress, energy metabolism, and immune regulation in gill and kidney tissues under acute salinity stress.

Project description:In low rainfall regions soils are naturally conditioned with frequent co-occurrence of salinity and alkalinity. Plant salinity responses both at physiological and molecular level have been extensively researched. However, effects of the combined treatment of alkaline salinity that could greatly reduce plant growth and the mechanisms responsible for tolerance remain indeterminate. In Brassica juncea, large reductions in biomass and increased leaf Na+ concentration under alkaline salinity indicates that the combined treatment had greater negative effect than salinity on both growth and the physiological responses of the plant. To determine molecular mechanisms potentially controlling adaptive tolerance responses to salinity and alkaline salinity, the moderately tolerant genotype NDR 8501 was further investigated using microarray analysis. The transcripts of treated leaf tissues verses those of the untreated control sample were analysed after prolonged stress of four weeks. In total, 528 salinity responsive and 1245 alkaline salinity responsive genes were indentified and only 101 genes were expressed jointly in either of the two treatments. Transcription of 37% more genes involved in response to alkaline salinity than salinity alone, which suggests the increased impact and severity of the combined stress on the plant, indicating the transcription of a far greater number of genes likely involved in mitigation and damage control. Transcription of KUP2 and KUP7 genes involved in potassium homeostasis under salinity alone and NHX1 and ENH1 genes for ion (K+ and Na+) homeostasis under alkaline salinity, clearly demonstrated that different genes and genetic pathways are involved in response to each stress. They further provide supporting evidence for the physiological responses that occur in the plant, with massive reprogramming of the transcriptome leading to partial ion exclusion, shuttling and compartmentation.
