Project description:The arms race between plants and herbivorous insects involves sophisticated molecular strategies. Tuta absoluta ranks among the most devastating pests of Solanaceous crops. While herbivorous insects commonly secrete effector proteins to modulate plant immunity, how T. absoluta accomplishes this remains largely unknown. This study identifies TaCRVP, a novel salivary effector protein from the T. absoluta. We demonstrate that TaCRVP is delivered into plant cells during feeding and functions as a key factor by targeting the host catalase SlCAT2. We found that TaCRVP promotes SlCAT2 tetramerization, thereby enhancing its enzymatic activity to scavenge H₂O₂. Furthermore, TaCRVP enhances SlCAT2 stability by competitively inhibiting its interaction with the plant E3 ubiquitin ligase SlAdBiL, thereby blocking the SlAdBiL-mediated targeting of SlCAT2 for 26S proteasomal degradation. The consequent increase in both SlCAT2 activity and abundance effectively suppresses H₂O₂ accumulation, a critical defense signal, leading to the downregulation of jasmonic acid-mediated defenses and the biosynthesis of key compounds. Our findings reveal a convergent evolutionary strategy whereby herbivorous insect effector, like pathogen effectors, manipulates host redox homeostasis by targeting catalase. This study advances our understanding of plant-insect interactions and lays the groundwork for developing novel pest management strategies by identifying potential molecular targets.

Project description:Plants respond to attack by chewing insects through the recognition of herbivore-associated molecular patterns (HAMPs) that are present in oral secretions (OS) and released at the wound site, leading to appropriate deployment of plant immune responses. Because insect feeding is accompanied by severe wounding of the leaf tissue, the specific contribution of HAMPs to defense is not well characterized. In addition, OS contain effectors that interfere with activation of defenses, including the suppression of wound-induced genes. Here, we analyze the transcriptome of Arabidopsis thaliana leaves in response to wounding and application of OS from Spodoptera littoralis or Pieris brassicae. We report that OS amplify wound-induced responses and specifically promote the activation of hormonal pathways, as well as pathogen- and cell wall-related responses. Furthermore, application of OS on wound sites extensively suppresses the expression of genes involved in the regulation and biosynthesis of aliphatic glucosinolates. Also, OS suppress the expression of genes involved in wound-healing processes and callus formation, including ERF114, DOFs and CYCD3s, in addition to genes encoding cell-wall modifying enzymes, whose activation completes wound sealing. Altogether these results highlight the contribution of OS components to defense and unveil the potential role of conserved OS-derived effector(s) in inhibiting the production of defense compounds and interfering with wound healing processes.

Project description:The whitefly Bemisia tabaci is a notorious pest of worldwide agriculture. It is believed to secrete saliva to counter plant defenses, but the underlying mechanism remains to be elucidated. Here, we characterize the gene/protein repertoires of B. tabaci salivary glands and secreted saliva by transcriptomic and LC–MS/MS analysis. A total of 698 salivary gland-higher expressed unigenes, as well as 172 saliva proteins are identified. Comparative analysis of the saliva composition in different arthropod species illustrates that proteins associated with binding, hydrolysis and oxidation-reduction are widely distributed in herbivorous saliva. There are 74 saliva proteins exclusively identified in B. tabaci, with 34 of them being B. tabaci-specific. In addition, eleven B. tabaci-specific saliva proteins plastically regulated in response to different diets, which might be associated with wide host range of this pest. Our results gain insight into whitefly–plant interactions, and provide a good resource for functional characterization of effectors

Project description:Lasiodiplodia theobromae is one of the causal agents of Grapevine trunk diseases, which becomes a tremendous threat to the grapevine production worldwide. Plant pathogens secrete diverse effectors to suppress host immune responses or regulate the host metabolism to promote diseases. Our results suggest that L. theobromae LtCre1 targets host VvRHIP1 to manipulate the sugar signaling pathway, through disrupting the association of VvRHIP1 and VvRGS1 complex.

Project description:The resistance mechanisms evolved by insects to overcome host-plant allelochemicals is a key consideration in pest management. Camphor oil (EO) and its main component (i.e D-camphor) form a specific terpenoid-defensive system in camphor trees, Cinnamomum camphora. However, an emerging insect pest, Pagiophloeus tsushimanus has recently caused serious damage to this intractable plant species, which is largely elusive. Here, we used feeding bioassays and RNA-seq to investigate the mechanism underlying the resistance of the beetle to host-specific terpenoid defenses. Firstly, a hormetic response in both larval weight and developmental time was observed in terpenoid-feeding individuals, which is a highly generalized dose-response phenomenon in toxicology whereas occurs infrequently in the context of insect-plant interactions. Then, comparative transcriptome analysis between terpenoid-feeding and control groups indicated that both CYP450s-mediated metabolic resistance and CPs-mediated cuticular resistance were jointly employed to cope with terpenoid-induced stress. In addition, a small portion of genes involved in glucose transport pathway were up-regulated at the low D-camphor dose, suggesting an extra intake of glucose used for larval growth may contribute to a hormetic response. These findings probably implied that the dual terpenoid-resistance mechanisms existing in this specialist is an essential precondition for hormetic response in larval growth, ultimately contributing to successfully colonize host camphor trees with impunity. Overall, our study will open new avenues for understanding the insect-plant coevolutionary adaptation and developing durable pest control strategies.

Project description:Sugarcane aphids (SCA; Melanaphis sacchari Zehntner) is a key piercing-sucking type pest of sorghum (Sorghum bicolor) which cause significant yield losses. While feeding on host plants, complex signaling networks are invoked from recognition of insect attack to induction of plant defenses. Consequently, these signaling networks lead to the production of insecticidal compounds or limited access to nutrients to insects. Previously, several studies are published on the transcriptomics analysis of sorghum in response to SCA infestation, but no information is available on the physiological changes of sorghum at proteome level. We used SCA resistant sorghum genotype SC265 for the global proteomics analysis after 1 and 7 days of SCA infestation using TMT-plex technique.

Project description:Pathogens rely on a diverse complement of effector proteins to manipulate host cellular processes. The devastating plant pathogen Phytophthora produces numerous effectors that consist of tandem repeats of the “(L)WY” motif. Here, we discovered a specific (L)WY-LWY combination that enables efficient recruitment of the Serine/Threonine protein phosphatase 2A (PP2A) core enzyme in plant hosts. Structural characterization of an effector-PP2A complex and biochemical analyses demonstrate multiple effectors adopt this module at the amino terminus to form functional PP2A holoenzymes but have divergent C-terminal LWY units. Our results discovered a virulence strategy widely employed by Phytophthora in which an essential phosphatase complex of the host is appropriated to promote disease and highlight how diversification in an effector repertoire is promoted by protein modularity.

Project description:Mutualistic symbionts are increasingly recognized as hidden players in plant-insect interactions. Understanding how symbionts affect plant responses to herbivory provides important insight into the fascinating complexity of terrestrial ecological networks and how we could specifically target symbionts to achieve sustainable pest management. Although the role of herbivore-associated symbionts is gaining increased attention in ecology of plant-insect interactions, how symbionts of members of higher trophic levels affect these interactions is poorly understood. The natural enemies of insect herbivores such as parasitic wasps that are used in biological pest control are associated with mutualistic symbionts (polydnaviruses) that they inject into the herbivore along with eggs in an event of parasitisation. We showed previously that polydnavirus-genes are expressed in the salivary glands of parasitized caterpillars suggesting that third-trophic level symbionts may alter the herbivore’s salivary composition and consequently, affect the way the herbivore interacts with the plant. By using a novel multidisciplinary approach that integrates molecular, chemical, behavioral and field ecology, the proposed project aims to manipulate the phenotype of caterpillars to study the effect of symbiotic viruses of third-trophic-level insects at the plant-herbivore interface. We will address this issue at different levels of biological organization, from plant gene expression to plant-mediated ecological consequences. The project will thereby provide novel fundamental insights into the hidden role played by parasitic wasp symbionts in plant responses to herbivory and these insights will generate important knowledge for pest management programs that focus on polydnaviruses as biological control agents.

Project description:The soldier fly is an endemic pest of sugarcane in Australia. Small numbers of larvae can cause significant damage to roots and reduce the crop yields. Little is known about the composition and function of the soldier fly salivary gland, its secretions, and their roles in insect-plant interactions. In this study, we performed transcriptome analysis of the salivary glands of starved and sugarcane root-fed soldier fly larvae. A total of 31,119 highly expressed assembled contigs were identified in the salivary glands and almost 50% of them showed high levels of similarity to known proteins in Nr databases. Of all the obtained contigs, only 9,727 sequences contain an open reading frame of over 100 amino acids. Around 31% of contigs were predicted to encode secretory proteins, including some digestive and detoxifying enzymes and potential effectors. Some known salivary secreted peptides such as serine protease, cysteine proteinase inhibitors, antimicrobial peptides and venom proteins were among the top 100 highly expressed genes. Differential gene expression analysis revealed significant modulation of 850 transcripts in salivary glands upon exposure to plant roots or starvation stress. Here, we identified some venom proteins which were significantly upregulated in the salivary glands of soldier fly larvae exposed to sugarcane roots. In other insects and nematodes some of these proteins have been used to manipulate host plant defense systems and facilitate the invasion of the host plant. These findings provide a further insight into the identification of potential effector proteins involved in soldier fly- sugarcane interactions.

Many effectors in the devastating plant pathogen Phytophthora contain tandem repeats of the “(L)WY” motif, which are structurally conserved but variable in sequences. Crystal structure of an effector-PP2A complex shows that the (L)WY-LWY module enables hijacking of the host PP2A core enzyme to form functional holoenzymes. While sharing the PP2A-interacting module at the amino terminus, these effectors possess divergent C-terminal LWY units and regulate distinct sets of phosphoproteins in the host. Our results highlight the appropriation of an essential host phosphatase through molecular mimicry by pathogens and diversification promoted by protein modularity in an effector repertoire.
