Proteomic analysis of cadmium immobilization by bacterial consortia of Achromobacter insuavis SL8 and Enterobacter cancerogenus SL12

ABSTRACT: Strains Achromobacter insuavis and Enterobacter cancerogenus, as well as their co-culture, were grown in LB medium with/without 100 mg/L Cd²⁺ for 48 h. For label‑free proteomics, cells were lysed in urea/thiourea/CHAPS buffer, homogenized, sonicated, and centrifuged. The protein precipitate was analyzed by LC-MS/MS (Agilent 1290‑TripleTOF 5600).

Project description:Cadmium (Cd) is a metal present in working and living environments known to be toxic and carcinogenic, but its mechanism of action remains still unclear. We investigated the gene expression modulation in human hepatoma cell line HepG2 after exposure to 2 M-BM-5m and 10 M-BM-5m cadmium using Agilent Whole human genome expression microarray. HepG2 cells were treated for 24 hours with 2 M-NM-<M and 10 M-NM-<M Cd and normal medium as control. Three independent replicates were used for the each type of stimuli.

Accompanying anion affects cadmium toxicity during Arabidopsis thaliana development

Project description:Cadmium (Cd) is a significant environmental pollutant with widespread detrimental effects on living organisms, making it a frequent subject of laboratory studies. However, different types of Cd salts are used to spike media, often without considering the possibility that accompanying anions may influence the effects of metal cations. Using two commonly used Cd salts, CdSO4 and CdCl2, we observed distinct toxicity effects on Arabidopsis thaliana development. On a physiological level, 7-day-old seedlings exposed to 50 µM CdSO4 had shorter roots than those treated with CdCl2. Proteomic analysis revealed strong downregulation of proteins involved in microtubule organization and primary cell wall synthesis in the root of plants exposed to CdSO4. Additionally, these plants exhibited higher Cd uptake from the medium and greater Cd accumulation in the shoot, indicating that the SO42-, as an accompanying anion, exacerbates Cd toxicity. These findings highlight the critical but often overlooked role of accompanying anions in modulating the toxic effects of heavy metals on plants.

Genome-wide profiling of responses to cadmium in the diatom Phaeodactylum tricornutum

Project description:The only group of organisms in which a biological function for cadmium has been shown is the diatoms, which are unicellular phytoplankton. Yet diatoms exhibit similar sensitivity to Cd as other groups of phytoplankton. We have investigated responses of Cd on molecular, metabolic and physiological levels in the diatom Phaeodactylum tricornutum. P. tricornutum apparently has a high tolerance to Cd; only minor responses were observed on growth, pigment and transcriptional changes at cadmium concentrations of 123 µg/l. No significant changes in chlorophyll and xanthophyll levels were observed, and the very few transcripts affected strongly indicate that the cells were able to respond to the increased Cd2+ levels without changing proteins levels. At 10 times this concentration, 1230 µg/l, a much clearer response was observed, including transcripts encoding proteins involved in metal transport, cell signalling and detoxification processes. Our results point towards putative pathways for the removal or detoxification of Cd and its metabolites, as well as a possible Cd uptake mechanism. We predict that ATPase5-1B is involved in removal of Cd by pumping Cd2+ ions out of the cell, whereas VIT1/CCC1 sequesters Cd2+ in the vacuole. Cultures of Phaeodactylum tricornutum were treated with two concentrations of Cd2+, 123 µg/l and 1230 µg/l. The reference samples were grown in artificial seawater base AK modified with added f/2-medium nutrients. Three biological replicates were harvested for all samples.
