Proteomic Profiling of Inflammatory versus Noninflammatory Cartilage Regions in Knee Osteoarthritis Patients

ABSTRACT: This study performed data-independent acquisition (DIA)-based quantitative proteomic analysis on paired cartilage samples from inflammatory and noninflammatory regions of knee osteoarthritis (OA) patients. The objective was to characterize the proteomic alterations associated with cartilage inflammation and degeneration in OA, and to elucidate the functional connections between oral-gut microbiota dysbiosis and joint pathological changes through multiomics integration.

Single-cell sequencing of murine articular cartilage

Project description:Osteoarthritis (OA) is a degenerative joint disease characterized by progressive cartilage loss, bone remodeling, synovial inflammation, and significant joint pain, often resulting in disability. Injury to the synovial joint such as the anterior cruciate ligament (ACL) tear is the major cause of OA in young adults. Currently, there are no approved therapies available to prevent joint degeneration or rebuild articular cartilage destroyed by OA, primarily because our understanding of the cellular and molecular changes that contribute to joint damage is very limited. The synovial joint is a complex structure composed of several tissues including articular cartilage, subchondral bone, synovium, synovial fluid, and tensile tissues including tendons and ligaments. In the present study, using single-cell RNA sequencing (scRNA-seq), we examined the cellular heterogeneity in articular cartilage from mouse knee joints and determined the knee joint injury-induced early molecular changes in the chondrocytes that could contribute to OA.

Characterization of the cartilage DNA methylome in knee and hip osteoarthritis.

Project description:The aim of this study was to characterise the genome-wide DNA methylation profile of osteoathritis (OA) chondrocytes from both knee and hip cartilage, providing the first comparison of DNA methylation between OA and non-OA hip cartilage, and between OA hip and OA knee cartilage. The study was performed using the Illumina Infinium HumanMethylation450 BeadChip array. LOXL2 deletion in post-traumatic injury dysregulates knee joint homeostasis and promotes pain sensitivity, inflammation, and osteoarthritis

Project description:Traumatic knee injuries lead to cartilage degeneration and osteoarthritis (OA). Cartilage has limited potential for self-regeneration, and any damage can lead to structural, molecular, and functional aberrations in the knee joint. Early changes in extracellular matrix (ECM) affecting cartilage are primarily asymptomatic and progress towards knee joint dysfunction, pre-OA, and finally OA. This study aimed to elucidate the mechanism of lysyl oxidase-like 2 (LOXL2) in maintaining healthy knee joint articular cartilage, its regeneration, and potential therapeutic applications. LOXL2 loss-of-function was evaluated using Acan promoter-specific inducible Loxl2 knockout, followed by immunohistochemistry, RNA-seq, transcriptional analysis, and knee joint functional and pain analysis. Our results showed that LOXL2 deletion increases the severity of destabilized medial meniscus (DMM) -induced cartilage damage. LOXL2-overexpressing mice were protected against degenerative cartilage changes in the knee joint compared with their wild-type littermates. Interestingly, Intra-articular injection of adenovirus-delivered LOXL2 protected knee joint function, alleviated cartilage degeneration, restored treadmill running capability, and reduced allodynia. Overall, LOXL2 loss initiates cartilage damage, inflammation, and pain, leading to OA. The gain of LOXL2 protects against progressive cartilage damage and relieves pain and inflammation. Thus, we identified a novel function for LOXL2 in OA-related local pain.

Project description:Subchondral insufficiency fracture of the knee (SIFK) is a common cause of knee joint pain that mainly afflicts the elderly. Yet its transcriptional profiles of cartilage in SIFK and OA patients are largely unknown. We performed single-cell RNA sequencing (scRNA-seq) on the cartilage of a SIFK patient, then compared with the processed scRNA-seq files of normal and OA cartilage tissues which were acquired from GSE169454.
