4D-Label-free quantitative proteomic analysis of differential proteins in the lung tissues of AT2 cell-specific VDR knockout mice

ABSTRACT: Six-week-old control mice and VDR CKO mice were administered tamoxifen (100 mg/kg body weight) via intraperitoneal injection once daily for five consecutive days. After a two-week interval, mice of each genotype were randomly allocated into two groups: the saline group and the BLM group. On day 0, mice in the BLM subgroup received an intratracheal injection of BLM (1 mg/kg), while those in the saline subgroup were given an equivalent volume of saline via intratracheal injection. All mice were euthanized on day 21, and their lungs were harvested for 4D label-free quantitative proteomics analysis.

Xijiao Dihuang decoction protects against sepsis-induced cardiac dysfunction

Project description:The goals of this study is to identify the differential expressed genes in heart of C57BL/6 mice with or without LPS treatment, and compare the differential expressed genes in the heart of LPS injection C57BL/6 mice after Xijiao Dihuang decoction(XJDHT) treatment. Briefly, the mice were randomly divided into 3 groups: control, LPS and LPS + XJDHT groups (n=4 for each group). Mice in Control and LPS groups were intraperitoneal injection with saline and 10mg/kg of LPS respectively, and orally administrated with saline; the mice in LPS + XJDHT groups were intraperitoneal injection with LPS (10 mg/kg) and orally administrated with 50 mg/kg /day of XJDHT for total 3 days. Then the heart were used to identify differentially expressed genes among different groups.

Transcription profiling of mouse neocortex and hippocampus kainate- and nicotine-injected juvenile mice

Project description:To examine irreversible changes in the developing brain following seizures, juvenile inbred mice were intraperitoneally injected with kainate and nicotine. Experiment Overall Design: Kainate and nicotine were dissolved in sterile saline, and intraperitoneally injected into postnatal day 23 males at a dose of 15 mg/kg and 10 mg/kg, respectively. Control mice received a comparable injection of saline. The injection was repeated three times at 2-hour intervals in the daytime, and after one week, the P30 survivors were sacrificed for sample preparation. Neocortices and hippocampi were isolated from mice that had received the repeated injections of saline, kainate, and nicotine (n=6, 5, and 6 mice, respectively).

Project description:Transcriptomic profiling of mouse liver tissue to characterize the acute and long-term gene expression response to the carcinogen N-nitrosodimethylamine (NDMA) in juvenile mice. RaDR+/+;GPT+/+ (WT) and RaDR+/+;GPT+/+;Mgmt-/- (Mgmt knockout) mice received intraperitoneal injections of NDMA (3.5 mg/kg at postnatal day 8 and 7 mg/kg at postnatal day 15) or saline vehicle. Liver tissue was collected at Day 1, Day 2, Day 5, Week 10, and Month 10 post-treatment and subjected to bulk RNA-seq to identify differential gene expression patterns across genotypes, sexes, treatments, and timepoints.

Gene expression changes induced by Dendrocalamus latiflorus leaf extract (DLE) and white tea extract (WTE) and pioglitazone (PGZ) in db/db mice liver

Project description:Eight-week-old C57BL/6J and T2DM db/db mice randomly divided into 4 groups of 8 based on equal bodyweight and fasting blood glucose (FBG) levels (mice were fasted for 6 h). The C57BL/6J mice (normal group) were treated with saline. The T2DM db/db mice were treated with saline, DLE (200 mg/kg), WTE (180 mg/kg), or pioglitazone (PGZ, 20 mg/kg) for 12 weeks, and were called the model group, DLE group, WTE group and PGZ group, respectively. All mice received oral administration of the indicated treatments once a day. After 12 weeks drug administration, liver aliquots from four randomly selected mice from the normal, model, PGZ and DLE groups were ground into powder in liquid nitrogen, followed by RNAseq for differentially expressed gene determination and pathway enrichment analysis.

Project description:The goal of the study was to determine how 24-hour withdrawal from cocaine affects the expression profile of transcripts in the nucleus accumbens using genome-wide RNA-sequencing. 11-week old male C57/BL6 mice were injected mice intraperitoneally (IP) with saline (0.9%) or cocaine (20 mg/kg dissolved in 0.9% saline) for 7-days consecutive days, and 24-hours after the last injection, NAc tissue was dissected. Mice were housed in group of 5 mice per cage, and all mice in a cage were injected with either saline or cocaine and thus there was no counter-balancing between groups.
