Proteome analyse of tomato seed exudates

ABSTRACT: Proteomic dataset of exudates released during imbibition by tomato seeds in dormant and non-dormant states. The study includes treatments with cordycepin and cycloheximide to assess the contribution of de novo transcription and translation to the abundance of defense-related proteins, including PR proteins.

Project description:During maturation seeds acquire several physiological traits to enable them to survive drying and disseminate the species. Few studies have addressed the regulatory networks controlling acquisition of these traits at the tissue level particularly in endospermic seeds such as tomato, which matures in a fully hydrated environment and does not undergo maturation drying. Using temporal RNA-seq analyses of the different seed tissues during maturation, gene network and trait-based correlations were used to explore the transcriptome signatures and identify hubs associated with desiccation tolerance, longevity, germination under water stress and dormancy.

Project description:To investigate and compare the influence of root exudates of tomato and maize on Pseudomonas donghuensis P482, we have grown the strain up to a stationary phase in M9 0.4% glucose medium supplemented with maize exudates (Maize), tomato exudates (Tomato) or without supplementation (Control). We then performed differential gene expression analysis, identifying changes in transcriptome profiles between each treatment (Tomato, Maize) and the Control as reference conditions, and between the two treatments.

Nitrate control of Arabidopsis seed dormancy

Project description:Freshly harvested Arabidopsis seeds (ecotype Col0 obtained under 10 mM nitrate nutrition, referred to as C10) are dormant and do not germinate when sown on an agarose-based medium. RNA-Seq of seed dormancy in red rice

Project description:Red rice fully dormant seeds do not germinate even under favourable germination conditions. In several species, including rice, seed dormancy can be removed by dry-afterripening (warm storage); thus, dormant and nondormant seeds can be compared for the same genotype. A weedy (red) rice genotype with strong dormancy was used for mRNA expression profiling, by RNA-Seq, of dormant and nondormant dehulled caryopses (here addressed as seeds) at two temperatures (30 °C and 10 °C) and two durations of incubation in water (8 hours and 8 days). Aim of the study was to highlight the differences in the transcriptome of dormant and nondormant imbibed seeds.

Analysis of light induced secondary seed dormancy in Aethionema arabicum, Cyprus accession

Project description:Light inhibits the seed germination in the Cypriot accession (CYP) of Aethionema arabicum. Extended light illumination also induces a secondary seed dormancy that inhibits the germination even if the seeds were transferred back to darkness. This analysis aims to compare the seed transcriptome before or after the dormancy was established by light illumination (100 µmol/m2s; white light). One day light treatment (1dL) inhibits the germination but the seeds are not yet dormant. Seven day (7dL) or 14-day (14dL) long light treatment induces dormancy, and the seeds stay dormant after 7 dal-light plus 7 day-dark (7dL7dD) treatment. To understand the dormancy establishment, transcriptome was compared between non-dormant (1dL) and dormant (7dL, 14dL, 7dL7dD) seed samples.

Effect on tomato plants of asymptomatic infection with southern tomato virus

Project description:Southern tomato virus (STV) often infects healthy tomato plants (Solanum lycopersicum). In this study, we compared STV-free and STV-infected cultivar M82 plants to determine the effect of STV infection on the host plant. STV-free plants exhibited a short and bushy phenotype, whereas STV-infected plants exhibited increased height. STV-infected plants produced more fruit than STV-free plants, and the germination rate of seeds from STV-infected plants was higher than that of seeds from STV-free plants. This phenotypic difference was also observed in progeny plants (siblings) derived from a single STV-infected plant in which the transmission rate of STV to progeny plants via the seeds was approximately 86%. These results suggest that the interaction between STV and host plants is mutualistic. Transcriptome analyses revealed that STV infection affected gene expressions in host plants, notably evidenced by down-regulation of genes involved in ethylene biosynthesis and signaling. STV-infected tomato plants thus might be artificially selected due to their superior traits as a crop.
