Proteomics of the co-culture system of INS-1 cells with conditioned medium from LPS-induced in vitro model of Hashimoto's thyroiditis

ABSTRACT: We treated the rat thyroid follicular epithelial cell line FRTL-5 with LPS to establish an in vitro cell model mimicking Hashimoto’s thyroiditis. Subsequently, the conditioned medium derived from LPS-stimulated FRTL-5 cells was collected and used to co-culture rat pancreatic β-cell line INS-1. Finally, the INS-1 cells after co-cultivation were harvested for proteomic sequencing analysis.

Project description:Bone metastases is a common severe complication for breast cancer. We previously showed that conditioned medium (CM) from osteocytes stimulated with oscillatory fluid flow, mimicking bone mechanical loading during routine physical activities, reduced breast cancer cell extravasation across endothelial monolayers. Endothelial cells are situated at an ideal location to mediate signals between osteocytes in the bone matrix and metastasizing cancer cells in the blood vessels. Therefore, we used RNA sequencing to show that CM from endothelial cells conditioned in CM from flow-stimulated osteocytes significantly altered gene expression in bone-metastatic breast cancer cells. This This provides insights into the capability of bone-loading activity in preventing bone metastases.

Project description:Here we aim to dissect the complex nature of the acute systemic response of HSCs to lipopolysaccharide (LPS), mimicking a bacterial infection. Acute LPS challenge transiently induced proliferation of quiescent HSCs in vivo, without yet impairing HSC function. We hypothesized that the effect of LPS on HSCs is indirectly mediated via proinflammatory cytokines. Our results showed that Acute LPS exposure depends in part on IFN signaling to activate HSCs.
