Multi-omics Study of Breast Cancer Initiation and Progression Across the Disease Continuum

ABSTRACT: This project is based on multi-stage disease samples obtained from the same patient with breast invasive ductal carcinoma and innovatively proposes a “space-for-time substitution” research strategy. Under a shared genetic background, we reconstruct the continuous evolutionary trajectory from normal mammary ducts through ductal hyperplasia and ductal carcinoma in situ (DCIS) to invasive ductal carcinoma (IDC), thereby overcoming the limitations of conventional approaches that infer disease progression by integrating samples from different individuals at different stages. In addition, we will establish a “super-omics” framework that integrates single-cell spatial proteomics, single-cell spatial transcriptomics, and nineteen large-scale omics modalities, enabling a comprehensive and multidimensional characterization of breast cancer evolution.
