Influence of diabetic retinopathy on peripheral blood gene expression

Project description:To understand the association between systemic factors and the diabetic retinopathy, blood samples were collected in 50 healthy controls, 74 diabetic patients without diabetic retinopathy (DM group), and 69 diabetic retinopathy (DR Group). T1DM and T2DM in this study was based on the diagnostic criteria for diabetes proposed by the WHO in 1999. The diagnosis of DR was based on the international clinical classification of DR proposed at the 2002 International Ophthalmology Conference. We then performed gene expression profiling analysis using data obtained from RNA-seq of three patient groups.

A single cell atlas of circulating immune cells involved in diabetic retinopathy

Project description:Circulating immune cells play a role in the pathophysiology of diabetic retinopathy (DR). Our study focused on examining the exact role of circulating immune cells in the development of DR. Single-cell RNA sequencing (scRNA-seq) techniques revealed unique differentially-expressed genes and pathways in circulating immune cells among non-diabetic retinopathy (NDR) patients and DR patients. These findings highlight the notable alterations in the immunophenotypes of circulating immune cells in patients with type 1 DR.

Identification of plasma exosomal protein markers associated with diabetic retinopathy progression by proteomic analysis

Project description:Protein biomarkers derived from plasma exosome have been extensively identified in various diseases. This study aimed to explore the biomarkers at different stages of diabetes retinopathy (DR) progression. Two cohorts, discovery and validation cohorts, were included in this study. The discovery cohort was used for proteomic analysis to screen differentially expressed proteins (DEPs). Hub proteins were identified by Wilcox.test, and their correlation with clinical characteristics were further analyzed. The validation cohort was used for enzyme-linked immunosorbent assay (ELISA) to verify the expression of hub proteins. Receiver operator characteristic (ROC) curve was conducted to evaluate the diagnostic values of hub proteins for DR progression. The correlation with clinical indicators was further analyzed.

Identification and Validation of microparticle proteins as biomarkers for diabetic retinopathy

Project description:Diabetic retinopathy (DR) is a microvascular complication that develops in diabetic individuals. There are currently no methods to detect the onset or progression of DR. Biomarkers can help identify the subset of diabetics who are at high risk of developing DR or those DR patients with a high probability to progress quickly to advanced stage. In this study, we examined the serum microparticles as a source of biomarkers for DR. We profiled the microparticle proteome and compared across six different sample groups representing the onset and progression of DR. We found that the microparticle proteins participate in DR pathogenesis. We also validated eight of these proteins and found two to be biomarkers for DR.
