Endothelial FGF13 impairs angiogenesis by accelerating oxygen-independent HIF-1α degradation

ABSTRACT: Anti-Flag antibody was added to the lysate of HUVECs transfected with Flag-FGF13 for co-immunoprecipitation, and IgG was used as a negative control. LC-MS/MS analysis was carried out by PTM Bio Co., Ltd (Zhejiang, China). The substrate proteins that may bind to FGF13 were screened out according to the score and Flag/IgG ratio of the detected proteins in the mass spectrometry data.

Project description:Angiogenesis is a tightly regulated process essential for both development and disease, yet the role of fibroblast growth factor 13 (FGF13) in vascular remodeling has remained unclear. Here, we identify FGF13 as a key negative regulator of physiological and pathological angiogenesis. Using endothelial cell–specific knockout and overexpression mouse models, combined with retinal vascularization assays, hindlimb ischemia and myocardial infarction models, and in vitro endothelial cell functional assays, we demonstrate that loss of FGF13 enhances, whereas its overexpression impairs, neovascularization. Mechanistically, FGF13 binds to the PAS domain of HIF-1α and facilitates its interaction with RACK1, leading to oxygen-independent degradation of HIF-1α and suppression of downstream angiogenic signaling. Importantly, adenoviral overexpression of HIF-1α reversed the inhibitory effects of FGF13, confirming its essential role in mediating FGF13-driven angiogenic regulation. These findings reveal FGF13 as a critical endothelial-intrinsic brake on angiogenesis and suggest its potential as a therapeutic target in ischemic vascular disease.

Indoxyl sulfate stimulates angiogenesis by regulating reactive oxygen species production via CYP1B1

Project description:Introduction: Indoxyl sulfate (IS) is an accumulated protein-bound uremic toxin found in patients with kidney disease. Although it is known that IS impairs the vascular endothelium, the precise underlying mechanism remains unclear. Methods: We performed RNA sequencing in human umbilical vein endothelial cells (HUVECs) that were treated with either IS or control medium. Gene enrichment analyses were performed based on differentially expressed genes. We further examined the main enriched biological processes at the functional level. Results: We identified 1,293 genes that were affected in IS-treated HUVECs when compared to control and gene enrichment analysis indicated altered vascular formation and cell metabolism. We observed decreased viability and increased cell senescence due to IS treatment at the functional level. Furthermore, we demonstrated that IS exposure of HUVECs promoted angiogenesis as shown by the increase in total tubule length in a 3D HUVECs/pericytes co-culture assay. We also observed an increase in ROS production in IS-treated endothelial cells. Notably, the pro-angiogenic response of IS and the increased ROS production were abolished when CYP1B1, one of the main target genes that was upregulated by IS, was silenced. Conclusion: IS promotes angiogenesis and CYP1B1 is an important factor in IS-activated angiogenic response.

YBX1 Drives Post-Myocardial Infarction Angiogenesis via an m⁶A-IGF2BP1/3 Dependent Stabilization of HIF-1α mRNA

Project description:Angiogenesis plays a vital role in myocardial repair following myocardial infarction (MI). Y-box binding protein 1 (YBX1), an RNA-binding protein known for its role in cancer, is found to be significantly upregulated in endothelial cells within the peri-infarct zone and in hypoxic human umbilical vein endothelial cells (HUVECs). Using in vitro and in vivo models, we demonstrate that YBX1 enhances endothelial cell viability, proliferation, migration, and tube formation. Endothelial-specific overexpression of YBX1 in a murine MI model improved cardiac function and angiogenesis, while YBX1 knockdown aggravated cardiac injury. Transcriptome (RNA-seq) analysis revealed that YBX1 deficiency disrupts HIF-1α signaling. Mechanistically, YBX1 forms a complex with IGF2BP1/3 to stabilize HIF-1α mRNA through N⁶-methyladenosine (m⁶A) modification, thereby promoting angiogenesis and cardiac repair.
