Project description:Human citrate Plasma was collected from the left atrial appendage directly after catheter ablation for atrial fibrillation with either teh Boston Scientific Cryo-Balloon System or the Boston Scientific Pulsed Field Ablation System. A total of 154 microRNAs (miRs) were consistently detected across all samples. While some variability in miR expression levels was observed, no statistically significant differences were found after adjusting for multiple comparisons.

Microsecond pulsed electric fields induce myocardial ablation by damaging mitochondria

Project description:Catheter ablation is an effective treatment to prevent recurrence of Atrial fibrillation (AF) and can be used to maintain sinus rhythm and improve symptoms of AF, but to some extent it can cause a range of adverse effects associated with catheter ablation. Pulsed electric field is a newer treatment modality to replace catheter ablation for atrial fibrillation due to its fewer side effects. Different from radiofrequency ablation, which destroys diseased myocardial tissue by thermal energy, pulsed electric field ablation achieves the purpose of atrial fibrillation ablation by inducing damage to diseased myocardial cells through irreversible electroporation. However, some experimental parameters and mechanism of pulsed electric fields remain unclear.

Multi-Omics Analysis Reveals Disturbance of Nanosecond Pulsed Electric Field in the Serum Metabolic Spectrum and Gut Microbiota

Project description:<p>Nanosecond pulsed electric field (nsPEF) is a novel ablation technique that based on high-intensity electric voltage to achieve tumor-killing effect in the target region, and increasingly considered for treating tumours of the liver, kidneys, and other organs with rich blood supply. When we were looking at the safety and feasibility of applying nsPEF on porcine livers, we found that the indicators of liver function after ablation fluctuated. To further study the influence of this technique in the treatment of liver, we collected serum and fecal specimens of the pigs pre-treatment and post-treatment. The gut microbiota of pigs was sequenced by Illumina Miseq platform for analyzing the diversity and alterations of gut microbiota. Liquid chromatography-mass spectrometry (LC-MS) based metabonomic analysis and Pearson coefficient method were also used to construct the interaction system of different metabolites, metabolic pathways and flora. </p>

Establishment of a dopaminergic neuron purification system in mice

Project description:Transcriptomic analysis of enriched of neuron population collected from in-vivo mouse brains has been a challenge in the neuroscience field due to its fragility in withstanding harsh condition during isolation and collection process. We established a fluorescent reporter mouse Ddc-hKO1 to facilitate the identification and collection of Ddc-expressing neurons (dopaminergic, serotonergic, cholinergic and adrenergic neurons) by the detection of red fluorescence signals using a FACs. We utilized an improved isolation protocol that ensure high yield and quality of viable neurons during collection that is suitable for RNA-sequencing. This is the first report of transcriptomic profiling of Ddc expressing (hKO1(+)) neurons. Successful collection of Ddc-expressing neurons were verified by gene markers of dopaminergic, serotonergic, cholinergic neurons in hKO1(+) populations while other neuron types in hKO1(-) population. Furthermore, GSEA analysis were performed on both hKO1(+) and hKO1(-) to further support the neuron types collected in respective population.
