Project description:Plastic pollution, especially from mismanaged waste degrading into nanoplastics (NPs), poses a growing threat to environmental and human health. Polystyrene NPs (PS-NPs) are particularly prevalent, infiltrating ecosystems and accumulating in organs, including the brain. While NPs are linked to behavioral, metabolic, and developmental effects, their impact on reproduction and fertility remains poorly understood. Declining fertility rates suggest environmental toxicants like NPs may contribute to adverse reproductive outcomes. Reproductive function relies on the hypothalamus-pituitary-gonad (HPG) axis, centrally regulated by gonadotropin-releasing hormone (GnRH) neurons. These neurons originate in the nasal placode and migrate to the hypothalamus during embryogenesis. Altered GnRH neuron development or release can lead to absent puberty and infertility, whose genetic causes explain only 50% of known cases. In this context, environmental factors including NPs might contribute to the etiology of these disorders. In this work, we take advantage of an established model of migrating GnRH neurons (GN11) to assess PS-NPs effects and to provide the first evidence that these environmental factors interfere with cellular and molecular processes of GnRH neurons, thus suggesting a novel mechanism contributing to idiopathic infertility.

Project description:Nanoplastics (NPs), plastic particles smaller than 1 μm, have gained increasing attention due to their potential health effects. Experimental studies in animals have shown that NPs can pass biological barriers, reach the female reproductive tract, and enter the ovary. Their presence in the ovary might have profound consequences for reproductive endpoints, such as oocyte development. This study aimed to investigate the uptake and associated effects of polystyrene (PS) NPs on oocyte and embryo development using a bovine in vitro fertilization (IVF) model. Bovine cumulus-oocyte complexes (COCs) were exposed to pristine PS-NPs (50 nm or 200 nm) at different doses during in vitro maturation (23 hours), followed by IVF and embryo culture. RNA sequencing was performed to investigate mechanisms involved in possible effects on oocyte maturation. Confocal microscopy confirmed the internalization of 50 nm and 200 nm PS-NPs into cumulus cells, however, only 50 nm particles were able to transfer into oocytes. Additionally, exposure to 50 nm PS-NPs at 3 μg/mL significantly delayed oocyte nuclear maturation and early embryo development. At this dose, RNA sequencing did not identify differentially expressed genes in the COCs, however, gene set enrichment analyses did indicate effects on mitochondrial processes. Nevertheless, subsequent analysis on mitochondrial activity remained unaffected.

Project description:Nanoplastics pollution is a rising environmental concern whose impacts on biodiversity and human health are far from being understood. This is particularly salient in aquatic ecosystems, where the majority of species depend on external fertilization for reproduction. Here we evaluated the effects of a short-term exposure to engineered polystyrene nanoplastics (NPs) in the zebrafish germ line to further explore their impact on reproduction. To this end, zebrafish (Danio rerio) were exposed to 5 mg/L of 45 nm polystyrene (PS)-NPs via water for 96h. We show that, in males, nanoplastics induced testicular histological alterations with abnormal sperm clustering and chromatin compaction, resulting in viable spermatozoa but with reduced motility. Moreover, in females we observed an alteration in oocyte stages frequencies during oogenesis, possibly reflecting alterations in oocyte growth. RNA-sequencing analysis in male testis links nanoplastic induced alterations in the expression of genes involved in chromatin structure, meiosis and DNA double-strand break formation and repair progression, and gametes recognition. Flow cytometry analysis revealed that the observed effects in males were directly due to nanoplastics penetrating the testicular barrier and being internalized within germline cells. Overall, our results demonstrate that acute exposure to NPs can compromise reproductive fitness, underscoring the environmental and health impacts of NPs pollution.

Project description:Plastic particles in water environment can adsorb heavy metals, leading to combined toxicity to aquatic organisms. However, current conclusions are mostly obtained based on cell population-average responses. Heterogeneity effects among cell populations in aquatic organisms remain unclear. This study analyzed the heterogeneity effects of 200 μg/L 100 nm polystyrene nanoplastics (PS-NPs), 50 μg/L lead (Pb), and their combined exposures on zebrafish intestine cells by single-cell RNA sequencing.A total of 38640 cells in the zebrafish intestine was obtained and identified as seven cell populations, including enterocytes, macrophages, neutrophils, B cells, T cells, enteroendocrine cells, and goblet cells.Co-exposure of PS-NPs and Pb caused similar transcriptome profiles with PS-NPs exposure in macrophages, which changed immunological recognition processes. The Pb exposure influenced the macrophages by direct cytotoxicity. However, the Pb alone and combined exposures induced similar modes of action in the enterocytes, including the generation of oxidative stress and abnormal lipid metabolism.

The mice were treated with water in control group. We found that exposure to PS NPs caused cognitive decline. PS NPs exposure influenced the prefrontal cortex cells with more pathological alteration with increasing dosage. High-throughput RNA sequencing was conducted to explore miRNA expression in prefrontal cortex. Twenty-nine differentially expressed miRNAs were detected, including 12 upregulated and 17 downregulated miRNAs. This finding provided a reference for further studies on the development mechanisms of ncRNA during cognitive dysfunction.

Effects were investigated on transcriptomic responses, in digestive gland gonads and oocytes of exposed oysters. Transcriptomic profiles in the tissues of the exposed oyster showed endocrine disrupting signals, notably highlighting alteration in glucocorticoid response, insulin pathway and fatty-acid metabolism in response to micro-PS exposition. In oocytes from exposed females, several transcripts coding for proteins involved in Ca2+ binding were differentially expressed suggesting a disruption of the Ca2+ signaling pathway with crucial consequences on oocyte maturation.

Project description:In vitro fertilization (IVF) is an assisted reproductive technology that has enabled millions of births. Although considered safe, emerging evidence suggests that IVF may have long-term health adverse outcomes in offspring, though its impact on female reproduction, especially on reproductive aging is largely unknown. Here, we use a mouse model to investigate how IVF influences female reproductive and transgenerational health outcomes. We assessed IVF-conceived fetal (E18.5) and adult (12-week and 39-week) female offspring, identifying alterations in ovarian-to-body weight ratios, ovarian morphology, serum sex hormone levels, and transcriptomic and DNA methylation profiles in ovaries, oocytes, and cumulus cells, consistent with biomarkers of accelerated reproductive aging. The offspring of IVF females and wild-type males exhibited altered fetal-to-placental weight ratios, placental morphology, and dysregulated placental gene expression. At 12-weeks-of-age, offspring showed increased body weight, disrupted lipid and glucose metabolism, and transcriptomic and epigenetic changes in liver and reproductive organs. Our results highlight the need for deeper mechanistic understanding of assisted reproduction’s long-term and multigenerational impacts on female reproductive health.

Adults were sampled 2 and 8 weeks after the beginning of exposure (corresponding to T1 and T3, respectively, 8-9 replicates per time of sampling and condition for a total of 56). Tissues were immediately dissected from each oyster, frozen in liquid nitrogen, then crushed to a fine powder at -196°C with an oscillating mill mixer and stored in liquid nitrogen until RNA extraction. Oocytes were collected from 5 females per condition, filtered in a 40 µm sieve, counted and transferred into 1.5 mL of Extract-all reagent (Eurobio, Courtaboeuf, France) (20,000 oocytes). Total RNA was isolated using 1.5 mL of Extract-all Reagent per 50 mg of gonad powder. For microarray hybridizations, 200 ng of total RNA were indirectly labeled with Cy3 using the Low Input Quick Amp Labeling kit One-Color. Hybridization was performed using the Agilent Gene expression hybridization kit (5188-5242), with 1.65 μg of labeled RNA, for 16 h at 65 °C. The employed arrays were Agilent 60-mer 4x44K custom microarrays, containing 31,918 C. gigas ESTs, designed by Dheilly et al. (2011). Slides were scanned on an Agilent Technologies G2565AA Microarray Scanner system at 5 μm resolution, using default parameters. Features were extracted using the Agilent Feature Extraction software 6.1.
