Dysregulation of mechanotransduction is implicated in the progression of various diseases such as cancer and cardiovascular diseases. The actin cytoskeleton plays a crucial role in transmitting mechanical stimuli. Actin filaments, essential for cell motility and shape changes, respond to mechanical cues by remodeling, influencing gene expression via the LINC complex and mechanosensitive transcription factors. This study employs the DSP-MNase DNA-seq method to explore the relationship between cellular mechanosensing and chromatin architecture.

Project description:The actin cytoskeleton is a highly conserved structural network that supports diverse cellular processes, but its contribution to organismal aging is not well understood. This project investigates how genetic and pharmacological perturbations of actin and actin-binding proteins influence global gene expression and aging phenotypes in Caenorhabditis elegans. Whole-animal RNA interference was used to knock down key actin regulatory genes, including arx-2 (Arp2/3 complex), unc-60 (cofilin), and lev-11 (tropomyosin). Here Bulk RNA sequencing was performed to assess transcriptional responses to actin cytoskeletal disruption using genetic knockdown (RNAi) of actin-binding proteins and actin de/stabilizing molecules. These datasets capture transcriptomic signatures associated with actin dysfunction, aging trajectories, and stress responses, and provide a resource for exploring the molecular links between cytoskeletal integrity and longevity regulation.

Project description:Cell-cell fusion is a frequent and essential event during development, and its dysregulation causes diseases ranging from infertility to muscle weakness. Fusing cells repeatedly need to remodel their plasma membrane by orchestrated formation and disassembly of cortical actin filaments, but how the dynamic reorganization of the actin cytoskeleton control is still poorly understood. Here, we identified a ubiquitin- dependent toggle switch that establishes reversible actin bundling during mammalian cell fusion. We found that EPS8-IRSp53 complexes stabilize cortical actin bundles at sites of cell contact, which in part pushes cells towards each other. Conversely, EPS8 monoubiquitylation by CUL3KCTD10 displaces EPS8-IRSp53 from membranes and counteracts actin bundling, a dual activity that allows apposed cells to progress with fusion. We conclude that cytoskeletal rearrangements during development are precisely controlled by ubiquitylation, raising the possibility to modulate the efficiency of cell-cell fusion for therapeutic benefit.

Project description:Members of the NETWORKED (NET) family are involved in actin-membrane interactions. Here we show that two members of the NET family, NET4A and NET4B, are essential for normal guard cell actin reorganization, which is a process critical for stomatal closure in plant immunity. NET4 proteins interact with F-actin and with members of the Rab7 GTPase RABG3 family through two distinct domains, allowing for simultaneous localization to actin filaments and the tonoplast. NET4 proteins interact with GTP-bound, active RABG3 members, suggesting their function being downstream effectors. We also show that RABG3b is critical for stomatal closure induced by microbial patterns. Taken together, we conclude that the actin cytoskeletal remodelling during stomatal closure involves a molecular link between actin filaments and the tonoplast, which is mediated by the NET4-RABG3b interaction. We propose that stomatal closure to microbial patterns involves the coordinated action of immune-triggered osmotic changes and actin cytoskeletal remodelling likely driving compact vacuolar morphologies.

Project description:The cytoskeletal actin-binding ERM (ezrin, radixin and moesin) proteins play essential role in modulating numerous signal transduction events in the cytoplasm by crosslinking actin filaments with plasma membrane proteins. The nuclear localization of ERMs has been also reported and the involvement of the single Drosophila ERM protein in nuclear mRNA export was recently described. However, because the cytoskeletal activity of ERMs is essential in the cell, and the exact mechanism of their nuclear transport is still unknown, the biological significance of the presence of ERM proteins in the nucleus has not been yet determined. Here we present the first attempt to reveal the in vivo relevance of the nuclear localization of the cytoskeletal ERM protein, Drosophila Moesin. With the help of a nuclear export signal (NES) we decreased the amount of Moesin in the nuclei of the animal. As a result, we observed various developmental defects, demonstrating for the first time the importance of ERM function in the cell nucleus. Transcriptome analysis of the mutant flies revealed, that the lack of nuclear Moe function leads to expression change of about 700 genes, among them a group of heat shock genes. This result together with additional findings demonstrates that in Drosophila, the expression of protein chaperons requires the nuclear functions of Moesin.

Moreover, auxin transcriptional regulation is enhanced on the mutant background, supporting the idea that shaping actin filaments is sufficient to modulate BR-mediated auxin responsiveness. The actin cytoskeleton thus functions as a scaffold for integration of auxin and BR signaling pathways. Three biological replicates were performed for each sample (wild-type and actin2-5) and hybridized to the the Affymetrix ATH1 GeneChips.
