Project description:The entomopathogenici fungus B. bassiana is of great imporatnce in biological control of insect pests. In integrated pest management (IPM) program, unsaturated fatty acids and their derivatives are used as fungicides. The compatibility of mycoinsecticides with fungicides has attracted more and more attention. The genome-wide exprssion analysis involved in fungal resonse to unsaturated fatty acids was analyzed by using high throughput sequencing (RNA-Seq) at different concentrations of linoleic acid. Our transcriptional profiles revealed that numerous differentially expressed genes (DEGs), of which involved in metabolism, cell transport and cell rescue, were significantly involved in the tolerance of B. bassiana to linoleic acid stress.

Project description:The arms race between plants and herbivorous insects involves sophisticated molecular strategies. Tuta absoluta ranks among the most devastating pests of Solanaceous crops. While herbivorous insects commonly secrete effector proteins to modulate plant immunity, how T. absoluta accomplishes this remains largely unknown. This study identifies TaCRVP, a novel salivary effector protein from the T. absoluta. We demonstrate that TaCRVP is delivered into plant cells during feeding and functions as a key factor by targeting the host catalase SlCAT2. We found that TaCRVP promotes SlCAT2 tetramerization, thereby enhancing its enzymatic activity to scavenge H₂O₂. Furthermore, TaCRVP enhances SlCAT2 stability by competitively inhibiting its interaction with the plant E3 ubiquitin ligase SlAdBiL, thereby blocking the SlAdBiL-mediated targeting of SlCAT2 for 26S proteasomal degradation. The consequent increase in both SlCAT2 activity and abundance effectively suppresses H₂O₂ accumulation, a critical defense signal, leading to the downregulation of jasmonic acid-mediated defenses and the biosynthesis of key compounds. Our findings reveal a convergent evolutionary strategy whereby herbivorous insect effector, like pathogen effectors, manipulates host redox homeostasis by targeting catalase. This study advances our understanding of plant-insect interactions and lays the groundwork for developing novel pest management strategies by identifying potential molecular targets.

Project description:Background: The horn fly, Haematobia irritans (L.), is an obligate blood-feeding parasite of cattle and control of this pest is a continuing problem in the United States and other parts of the world. Worldwide annual economic losses attributable to this pest surpass $1 billion. The fly is becoming resistant to pesticides and new control technologies are needed by cattle producers. Results: Dominant conditional lethal gene systems are being investigated as population control technologies against agricultural insect pests. One of the critical components of these systems is a highly expressed female-specific gene promoter which can be used to drive expression of a lethality-inducing gene. To identify candidate genes to supply this gene promoter, microarrays were designed from a recently developed horn fly EST database and probed to identify female-specific and larval-specific differential gene expression. Analysis of dyeswap experiments found 432 and 417 transcripts which were over- and under-expressed in adult female flies, respectively, compared to adult male flies. Additionally, 419 and 871 transcripts were over- and under-expressed in first instar larvae compared to adult flies. Three transcripts were identified which were over-expressed in adult females flies compared to adult males and which also were over-expressed in the first instar larval lifestage compared to adult flies. Conclusion: We have identified 3 strong candidates for further evaluation as a gene promoter source for the development of a female-specific conditional lethality system in the horn fly. One of these candidates, the putative nanos orthologue, has a high female-to-male expression ratio, has a moderate expression level in first instar larvae, and has been well characterized in D. melanogaster. Further investigations leading from this microarray analysis will include transformation of the horn fly and evaluation of the female conditional lethal system components for applicability to the specific biological parameters of natural populations of the horn fly. Project description:In cotton, after the Bt introduction, lepidopteron pest attack been reduced to a greater extent, however, the sucking pests became major problem. Among the sucking pests, the leafhopper Amrasca biguttula biguttula (Ishida), considered to be a severe pest. Here, we reported the label-free quantitative proteomics to identify putative differentially expressed proteins contributing to leafhopper resistance. The results obtained from this work not only aimed to provide a new clue that will facilitate better understanding of molecular interaction between cotton plant and leafhopper, but also explored the key genes for the development of functional markers in cotton towards leafhopper resistance.

Project description:Protease inhibitors (PIs) are among the most ubiquitous plant defences against herbivorous insects. These inhibitors arrest proteolytic digestion by binding to the proteases in the gut. However, many pest insects have coevolved mechanisms to counteract these effects. While the nature of these counter-defensive strategies has been well described for several insect pests, our understanding of how these responses are triggered and regulated remains limited. In this study we investigated the initiation of this adaptive response in the economically damaging migratory locust (Locusta migratoria), via microarray analysis of gut tissues following the dietary uptake of PIs. For this purpose, the total number of ESTs available for locusts was narrowed down to transcripts that were found expressed in gut and/or brain tissue. The remaining subset was used for the construction of an optimized microarray platform.

Project description:Being a sessile organism, plants are constantly confronted by various biotic (pest and pathogen) and abiotic (drought, salinity, flood, extreme temperatures, etc.) stresses. In response to these environmental stresses, plants have developed numerous defense mechanisms. One of the basal defense responses in plants are mediated by trypsin inhibitors (TIs). Putranjiva roxburghii trypsin inhibitor (PRTI), a potent trypsin inhibitor from P. roxburghii showing sequence similarity with a group of genes known to have defense and storage function such as wound inducible (WIN) proteins, vegetative storage proteins (VSPs), and Bark storage protein (BSPs) was overexpressed in Citrus aurantifolia. PRTI overexpressing lines were tolerant to various abiotic stresses (salinity, drought, and alkalinity) and two pests namely, Scirtothrips citri and Papilio demoleus. The molecular insights underlying the heterologous overexpression of PRTI at the transcriptomic level reveals the upregulation of stress responsive genes and involvement of hormonal signal transduction and transporters. Further, genes related to DNA repair, amino acid synthesis, and development were also found to be upregulated. Our study also reveals the nuclear-cytoplasmic localization and alteration phytohormone profile by PRTI overexpression in transgenic lines as compared to wild-type which clearly indicates the role of abscisic acid ABA in stress tolerance.
