Project description:Disuse osteoporosis is a condition characterized by rapid bone loss induced by reduced mechanical loading on bones due to physical inactivity. An increase in osteoclast number is associated with bone loss in disuse osteoporosis. Project description:Fatty acid-binding protein 4 (FABP4), a key lipid protein in metabolism and inflammation, has been suggested to be linked to osteoporosis (OP), though direct evidence is scarce. Here, we present the first clear evidence of FABP4's significant role in OP, supported by clinical data and comprehensive in vivo and in vitro experiments. Elevated serum FABP4 in OP patients inversely correlates with bone mineral density (BMD), with similar trends observed in OVX mice. While FABP4 does not influence osteoblast differentiation, it promotes osteoclast formation and bone resorption. The FABP4 inhibitor BMS309403, with an IC50 of 0.89 μM, inhibits osteoclast differentiation by modulating calcium ions and suppressing the Ca2+-Calcineurin-NFATc pathway. Oral BMS309403 increased BMD in OVX mice, albeit less effectively than alendronate, whereas bone-targeted PLGA nanoparticles showed comparable efficacy to alendronate. This research identifies FABP4 as a promising therapeutic target for OP, with significant clinical implications.

But patients with subclinical hyperthyroidism, decline in the level of thyroid stimulating hormone (TSH) alone, are at equal risk of osteoporosis. Research has shown that there is thyroid-stimulating hormone receptor (TSHR) on the osteoclast, but whether TSH regulates osteoclast directly and its mechanisms are still not be stated clearly. In this study, in combination of in vivo and in vitro, we took the osteoclast precursor cell conditional and systemic knockout TSHR mouse models to study the effects of TSHR knockout on bone metabolism in mice and the changes of osteoclast differentiation and function in vitro. Transcriptomics was used to screen the differential genes and signal pathways, and the in vitro experiments were verified. We found that Tshr flox/ flox Lyz2-Cre (TSHR MKO) mice display decreased femoral biomechanics and bone density, damaged bone microstructure. Serum bone resorption marker CTX-I was significantly increased, accompanied by an increase in the number of osteoclasts in bone sections in TSHR MKO mice. The percentage of CD11b+ in TSHR knockout mice was decreased compared with control littermates. Osteoclasts from Tshr -/- mice had stronger osteoclast differentiation and acid secretion ability. In vitro cell experiments confirmed TSH effecting osteoclast differentiation and function by activation of AMPK signaling pathway. In conclusion, TSH inhibits osteoclast differentiation and bone resorption by activating AMPK signaling pathway, increases bone mass, and has osteoprotective effect. This will provide a reference for the diagnosis and treatment of osteoporosis, especially thyroid function related bone metabolism bnormalities. TSH structural analogues or AMPK activators are expected to provide new ideas for the development of drugs for the prevention and treatment of osteoporosis.

Intermittent administration of a fragment of Parathyroid hormone (PTH) activates osteoblast-mediated bone formation and is used in patients with severe osteoporosis. However, the mechanisms by which PTH elicits its anabolic effect are not fully elucidated. Here we show that the absence of the homeodomain protein TG-interacting factor 1 (Tgif1) impairs osteoblast differentiation and activity, leading to a reduced bone formation. Deletion of Tgif1 in osteoblasts and osteocytes decreases bone resorption due to an increased secretion of Semaphorin 3E (Sema3E), an osteoclast-inhibiting factor. Tgif1 is a PTH target gene and PTH treatment failed to increase bone formation and bone mass in Tgif1-deficient mice. Thus, our study identifies Tgif1 as a novel regulator of bone remodeling and an essential component of the PTH anabolic action. These insights contribute to a better understanding of bone metabolism and the anabolic function of PTH.

Moreover, these treatments have drawbacks, including jaw osteonecrosis and skin eczema. Hence, there is an urgent need for new drugs to treat osteoporosis. Drug library screening was performed by alkaline phosphatase (ALP) staining in osteoblasts to identify potential candidates for osteoporosis. qPCR, Western blot, ALP staining, alizarin red staining, and TRAP staining were conducted to assess the impact of ZM-306416 (ZM) on osteoblast and osteoclast differentiation in vitro. Additionally, RNA sequencing and pathway analysis were carried out to explore molecular mechanisms. Micro-CT scan and immunostaining were used to determine bone phenotypes in vivo. Drug library screening demonstrated that ZM enhances ALP activity in osteoblasts, indicating its potential as a pro-osteogenic agent. ZM exerts dual effects by promoting osteoblast differentiation through the Wnt/β-catenin signaling pathway and simultaneously inhibiting osteoclast differentiation by the NF-κB and MAPK signaling pathways. In an OVX mouse model, ZM effectively prevents bone loss by stimulating osteoblast formation and inhibiting osteoclast development. Our study revealed that ZM has a dual anti-osteoporosis effect by promoting osteoblastogenesis and inhibiting osteoclastogenesis, mediated via activation of Wnt/β-catenin signaling and suppression of NF-κB/MAPK cascades. These findings suggest that ZM could be a promising therapeutic strategy for alleviating osteoporosis.

Project description:Objectives：Dysregulated mitophagy coupled with osteoclast activation orchestrates the development and progression of osteoporosis.Although albiflorin (ALB) exhibits bone-protective effects through anti-inflammatory and antioxidant activities, its precise mechanism—particularly regarding mitochondrial regulation—remains unknown. This study therefore investigates ALB as a novel osteoclast inhibitor by examining its molecular mechanism in regulating mitophagy via the Rap1a/ERK signaling pathway. Materials and Methods: ALB was evaluated using murine models of postmenopausal osteoporosis. Key methodologies included RNA sequencing (RNA-seq) for gene expression pathway analysis, transmission electron microscopy (TEM) for visualization of mitochondrial and autophagic structures, MitoTracker/LysoTracker co-staining for assessment of mitophagy, and Western blotting for protein signaling validation. The effects of ALB on osteoclast differentiation and bone loss mitigation were assessed both in vitro and in vivo. Results:ALB significantly inhibited osteoclastogenesis and osteoclast differentiation, thereby effectively reducing osteoporosis in murine models. RNA-seq analysis revealed that ALB modulated mitophagy by regulating the expression of Rap1a and components of the ERK signaling pathway. Validation through TEM demonstrated suppressed mitochondrial autophagy, while MitoTracker/LysoTracker co-staining confirmed a reduction in mitophagy. Furthermore, Western blot analysis showed that ALB inhibited osteoclast activation via the Rap1a/ERK signaling axis. Conclusion:ALB mitigates postmenopausal osteoporosis by suppressing osteoclast activation through Rap1a/ERK-dependent inhibition of mitophagy. These findings identify ALB as a promising therapeutic strategy for osteoporosis, addressing the need for safer long-term treatment options.

Project description:Osteoporosis poses an increasing health and socioeconomic burden on aging societies. Current therapeutic options often present potentially severe side effects or lack long-term efficacy, underscoring the need for more effective treatments. Establishing novel drug targets requires an in-depth understanding of their physiological roles. Here, we describe the impact of the adhesion G protein-coupled receptor GPR133/ADGRD1 on osteoblast differentiation and function. Constitutive and osteoblast-specific knockouts of GPR133/ADGRD1 in mice result in reduced cortical bone mass and increased trabecularization in femurs and vertebrae—features characteristic of osteoporosis. Genome-wide association studies in humans link GPR133/ADGRD1 gene variants to differences in bone mineral density and body height. At the molecular level, GPR133/ADGRD1 influences osteoblast function and differentiation through a combined activation mechanism involving interaction with its endogenous ligand PTK7 and mechanical forces. In vitro analysis shows that osteoblast differentiation is driven by cAMP-dependent activation of the b-catenin signaling pathway. Activation of GPR133/ADGRD1 with the receptor-specific ligand AP503 promotes osteoblast function and differentiation both in vitro and in vivo, significantly alleviating osteoporosis in a mouse ovariectomy model. This renders GPR133/ADGRD1 a promising therapeutic target for osteoporosis and other diseases characterized by reduced bone mass.
