Identification of RXRA-Interacting Proteins in M2 Macrophages by Co-Immunoprecipitation Coupled with LC-MS/MS
Ontology highlight
ABSTRACT: Co-immunoprecipitation (co-IP) coupled with LC-MS/MS was performed to identify RXRA-interacting proteins in M2-type macrophages (Mus musculus). RXRA was immunoprecipitated from M2 macrophage lysates using an anti-RXRA antibody, with IgG serving as the negative control. Immunoprecipitated proteins were reduced, alkylated, and digested with trypsin, then separated on a nanoElute nano-flow UHPLC system (Bruker) and analyzed on a timsTOF Pro mass spectrometer (Bruker) in PASEF mode with positive ion detection (scan range 100-1700 m/z). Raw data were searched with MaxQuant (v1.6.14) against the mouse protein database to identify the proteins in the RXRA immunoprecipitates. This dataset provides a resource for studying RXRA-mediated protein interaction networks and transcriptional regulation in M2 macrophage polarization and function.
