Spermatophylax of Poecilimon ampliatus

ABSTRACT: Nuptial gifts of P. ampliatus were divided into ampullae and spermatophylax. Both were separated by high-resolution 2-DE with a gel size of 23 cm x 30 cm and stained with FireSilver. 16 spots of the spermatophylax were excised, trypsin digested and analyzed by LC-FTICR.

Project description:The spermatophore was removed and separated into ampulla and spermatophylax. Each of these compartments was solubilized in 6M urea, 3 M thiourea, 70 mM DTT and centrifuged. The supernatant was separated by 2-DE and spots 9,10, and 11 were analyzed by LC-FTICR MS. Spots 9,10, and 11 were occuring on 2-DE patterns of both spermatophylax and ampulla.

Immunoreactive proteins in the oesophageal gland cells of Anisakis simplex sensu stricto detected by MALDI-TOF/TOF analysis

Project description:Two hundred ventricles were manually dissected from L3 stage larvae of Aniskakis simplex s.s. to allow direct protein analysis. Denaturing gel electrophoresis followed by monochromatic silver staining revealed the presence of differential (enriched) proteins when compared to total nematode extracts. Such comparison was performed by means of 1D and 2D electrophoresis. Pooled antisera from Anisakis spp allergic patients were used in western blots revealing the presence of 13 immunoreactive bands in the ventricular extracts in 1D, while 82 spots in 2D. The corresponding protein bands and spots were excised from the silver stained gel and protein assignation was made by MALDI-TOF/TOF. A total of eleven proteins (including isoforms) were unambiguosly identified. The majority of these proteins are known to be secreted by nematodes into the external environment, of which three are described as being major allergens in other organisms with different phylogenetic origin.

Yersinia enterocolitica SOR17 - The RNA chaperone Hfq impacts growth, metabolism and production of virulence factors in Yersinia enterocolitica

Project description:We work with the bacterium Yersinia enterocolitica, a gastrointestinal pathogen. In this study we characterized a bacterial mutant strain called SOR17. We compared the protein extracts of SOR17 with that of the parental strain JB580v. Bacteria were grown in triplicate for 5 h at 37°C, and following bacterial disruption by French press, proteins were fractionated into soluble or membrane proteins. We also compared total protein extracts of bacteria grown for 16 h at 27°C. Two-dimensional electrophoresis were performed and proteins were stained with Coomassie brilliant blue. Spots whose abundance changed more than 2 fold were excised and sent to mass spectrometry for identification by either MALDI-TOF PMF or LC-MS/MS.

Rat liver mitochondrial membrane proteome

Project description:The rat liver mitochondrial lysate was analyzed by the blue native PAGE. Then gel bands were excised and plated into tubes. De-stained excised slices were reduced and alkylated. Subsequently, the proteins of gel were digested by trypsin. The collected peptides were subjected to an NSI source followed by tandem mass spectrometry (MS/MS) in Orbitrap Exploris 480 MS coupled online to the UPLC.

Na2CO3-responsive proteins in Puccinellia tenuiflora leaves

Project description:Puccinellia tenuiflora is a monocotyledonous halophyte species belonging to the Gramineae. It has strong ability for surviving in the extreme saline-alkali soil (pH range of 9-10). In the present study, proteins from leaves under various Na2CO3 treatments were separated and visualized on Coomassie Brilliant Blue-stained two-dimensional gel electrophoresis (2-DE) gels. After gel image analysis on the base of the calculated average vol% values of each protein spot, 174 protein spots were detected as abundance changed protein spots (1.5-fold changes, p < 0.05). Among them, 104 protein spots were identified using MALDI-TOF MS/MS and Mascot database searching. Thus the 104 protein identities represented 80 unique proteins were taken as Na2CO3-responsive proteins in leaves. After integrative analysis by BLAST alignment, Kyoto Encyclopedia of Genes and Genomes, and referring information from related literature, they were classified into 10 functional categories, including photosynthesis, carbohydrate and energy metabolism, other metabolisms, stress and defense, membrane and transport, signaling, protein synthesis, folding, and turnover, cell wall mechanism, cell cycle, and miscellaneous or unknown.

RNA-Seq time-series of Adventitious root development in Populus ussuriensis
