Injection of IP-001 into thermally ablated solid tumors

Project description:Background and study aims Treatment in patients with advanced colorectal cancer, (CRC) non-small cell lung cancer (NSCLC) and soft tissue sarcoma (STS) represent a significant unmet need. Local thermal ablation of tumor tissue using techniques such as radiofrequency ablation (RFA) or stereotactic radiotherapy is an established treatment option for different tumors, in particular, CRC and NSCLC. There is strong nonclinical and early clinical evidence that combining thermal ablation with the investigational product, 1.0% IP-001 for Injection might be able to turn ‘cold’ tumors into ‘hot’ tumors, inducing a long-term systemic immune response mediated by the patient’s immunological defense system against any remaining tumor cells both at the site of the ablation treatment and those that are outside or distant from the treated area (known as the abscopal effect). The 2 components of treatment play unique roles in inducing an antitumor immune response: 1. During thermal ablation, treated tumor undergoes immunogenic cell death via a well-defined heat gradient 2. Adding 1.0% IP-001 for Injection elicits a tumor-specific adaptive immune response As a result, it is hypothesized that ablation + IP-001 can be deployed against a wide variety of tumor types to induce customized immune activation against a patient’s own, unique cancer. Who can participate? Adult patients with confirmed CRC, NSCLC, or STS What does the study involve? In this study, a solid tumor will first be thermally ablated by a standard ablation modality, and then 1.0% IP-001 for Injection will be injected into the area of the ablated tumor. Patients can be treated every 6 weeks for up to 4 treatments with RFA + 1.0% IP-001 for Injection.

Identification of miRNA targets in breast cancer cells (Ago2-IP)

Project description:miRNAs regulate mRNA stability and translation through the action of the RNAi-induced silencing complex. In this study, we systematically identified endogenous miRNA target genes by using AGO2 immunoprecipitation (AGO2-IP) and microarray analyses in two breast cancer cell lines, MCF7 and MDA-MB-231, representing luminal and basal-like breast cancer, respectively. The expression levels of ~70% of the AGO2-IP mRNAs were increased by DROSHA or DICER1 knockdown. In addition, integrated analysis of miRNA expression profiles, mRNA-AGO2 interaction, and the 3'-UTR of mRNAs revealed that >60% of the AGO2-IP mRNAs were putative targets of the fifty most abundantly expressed miRNAs. To identify mRNAs associated with AGO2, cell lysate was precleaned with control IgG and immunoprecipitated with anti-human Ago2 (Clone 2E12-1C9, Abnova). Total RNA from cell lysate or coimmunoprecipitated with AGO2 was extracted with Trizol and subjected to microarray analysis, with three biological repeats for each experimental condition.

Identification of the interaction proteins of PRSS37 by GFP-IP/MS using testis proteins from PRSS37-EGFP knock-in mice and pAcr-EGFP transgenic mice

Project description:To identify the potential interaction partners of PRSS37, three testis lysates of Prss37E/+ mice (PRSS37-EGFP knock-in heterozygous mice) and three testis lysates of pAcr-EGFP transgenic mice were immunoprecipitated using anti-GFP mAb magnetic beads. The GFP-IP products of six samples from two groups were subsequently analyzed by mass spectrometry (MS) to identify differentially immmunoprecipitated proteins (DIPs) in the PRSS37-EGFP group. Proteins detected in pAcr-EGFP group were used as negative controls to exclude the endogenous proteins and polypeptides that interact nonspecifically with the anti-GFP mAb magnetic beads.

Signaling protein antibody microarray analyses for islets of control and IFT88 knockout mice [SET100]

Project description:β-cell specific IFT88 knock-out mice recapitulate human diabetes with impaired insulin secretion and altered islet hormone paracrine regulation. To examine the signaling pathways regulating islet cell function, we subjected protein lysates of whole islets from control and IFT88 knockout mice to a commercial signaling-protein array analysis (Full Moon Bio, Inc). Samples were probed against 1358 antibodies with 2 replicates per antibody on 76 x 25 x 1mm glass slides.

CCDC188 IP-MS data in mouse testis protein lysates

Project description:We immunoprecipitated endogenous CCDC188 from testis protein lysates of adult wild-type mice using our homemade CCDC188 antiserum. No CCDC188 protein was identified in protein lysates in which mouse IgG was added. Thus, testis protein lysates added to mouse IgG were utilized as the negative control. A high-throughput technology, LC/MS–MS (MS), was applied to identify the candidate interacting proteins of CCDC188.
