Project description:The gastrointestinal tract is colonized by trillions of microorganisms collectively known as the gut microbiota. These microbes provide essential signals to support healthy gut function. The microbiota is separated from internal tissue by a single layer of intestinal epithelial cells that not only provides a physical barrier but also relays luminal signals to underlying gut immune cells. Altered microbiota composition including loss of anti-inflammatory microbes or outgrowth of mucosa-associated bacteria such as adherent-invasive E. coli (AIEC) are hallmarks of inflammatory disease including inflammatory bowel disease (IBD). In contrast to their hypothesized role in pathology, we recently identified select AIEC isolates that improve outcomes in mouse colitis models. These AIEC induce macrophage production of the anti-inflammatory cytokine IL-10 which limits gut inflammation and supports barrier repair. These benefits were lost if the AIEC was unable to attach to epithelial cells. However, the epithelial signaling underlying this protection remained unclear. To understand if intestinal epithelial cells signaled to immune cells after microbial attachment, we utilized human colonic organoid monolayers and found co-culture with a subset of AIEC isolates upregulated immune regulatory genes including CCL2, a macrophage recruiting chemokine. This effect was only observed in undifferenced epithelial cells, indicating epithelial stem cell recognition of microbes leads to macrophage recruitment. In vivo, antibody blockade of CCR2 abrogated the protective effect of AIEC colonization. Using bacterial transcriptome analysis, we identified high flagellin expression in AIEC isolates that activated epithelial signaling, with lost signaling in organoids deficient for TLR5, the receptor for flagellin. Together our findings suggest intestinal epithelial cells recognize microbial signals to coordinate macrophage recruitment that support intestinal repair, protecting from colitis.

Project description:The intestinal ecosystem is balanced by dynamic interactions between resident and incoming microbes, the gastrointestinal barrier, and the mucosal immune system. However, in the context of inflammatory bowel diseases (IBD) where the integrity of the gastrointestinal barrier is compromised, resident microbes contribute to the development and perpetuation of inflammation and disease. In this context, probiotic bacteria exert beneficial effects enhancing epithelial barrier integrity. However, the mechanisms underlying these beneficial effects are only poorly understood. Here, we comparatively investigated the effects of four probiotic lactobacilli, namely L. acidophilus, L. fermentum, L. gasseri, and L. rhamnosus in a T84 cell epithelial barrier model. Results of DNA-microarray experiments indicating that lactobacilli modulate the regulation of genes encoding in particular adherence junction proteins such as E-cadherin and b-catenin were confirmed by qRT-PCR. Furthermore, we show that epithelial barrier function is modulated by Gram-positive probiotic lactobacilli via their effect on adherence junction protein expression and complex formation. In addition, incubation with lactobacilli differentially influences the phosphorylation of adherence junction proteins and of PKC isoforms such as PKCd which thereby positively modulates epithelial barrier function. Further insight into the underlying molecular mechanisms triggered by these probiotics might also foster the development of novel strategies for the treatment of gastrointestinal diseases (e.g. IBD).

Project description:Repairing large bone defects remains a significant clinical challenge. This study explores the development of collagen-enhanced piezoelectric microfiber barrier membranes (PLLA/ZnO@COL) for bone regeneration. The membranes' physicochemical properties, especially their piezoelectric characteristics, significantly influenced cellular behavior. Scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDS) confirmed the uniform dispersion of ZnO nanoparticles within the PLLA matrix, essential for optimal piezoelectric performance. The inclusion of collagen layers enhanced the electrical properties, cytocompatibility and osteogenic activity of the membranes. In vitro analyses, including bioinformatics, revealed that PLLA/ZnO@COL membranes upregulated key osteogenic pathways such as PI3K-AKT, MAPK, and Ras, thereby enhancing osteoblast proliferation, migration, and differentiation. Low-intensity pulsed ultrasound (LIPUS) stimulation further amplified these effects, indicating a synergistic role in enhancing osteoblast functions. In vivo studies using a rat mandibular bone defect model demonstrated that PLLA/ZnO@COL membranes, with the collagen layer facing the defect, significantly fostered new bone formation compared to control groups. Micro-CT analysis at 6 and 12weeks post-implantation showed higher bone mineral density, volume, and trabecular number in the PLLA/ZnO@COL group, indicating greater osteogenic activity. In conclusion, PLLA/ZnO@COL membranes, in conjunction with LIPUS, effectively enhance osteoblast functions and induce bone regeneration, showing potential as innovative barrier membranes in bone tissue engineering. The combination of piezoelectric properties, collagen enhancement, and LIPUS stimulation presents a novel approach to improving bone healing outcomes.

Project description:Background: The selective absorption of nutrients and other food constituents in the small intestine is mediated by a group of transport proteins and metabolic enzymes, often collectively called ‘intestinal barrier proteins’. An important receptor that mediates the effects of dietary lipids on gene expression is the peroxisome proliferator-activated receptor alpha (PPARα), which is abundantly expressed in enterocytes. In this study we examined the effects of acute nutritional activation of PPARα on expression of genes encoding intestinal barrier proteins. To this end we used triacylglycerols composed of identical fatty acids in combination with gene expression profiling in wild-type and PPARα-null mice. Treatment with the synthetic PPARα agonist WY14643 served as reference. Results: We identified 74 barrier genes that were PPARα-dependently regulated 6 hours after activation with WY14643. For eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA) and oleic acid (OA) these numbers were 46, 41, and 19, respectively. The overlap between EPA-, DHA-, and WY14643-regulated genes was considerable, whereas OA treatment showed limited overlap. Functional implications inferred form our data suggested that nutrient-activated PPARα regulated transporters and phase I/II metabolic enzymes were involved in a) fatty acid oxidation, b) cholesterol, glucose, and amino acid transport and metabolism, c) intestinal motility, and d) oxidative stress defense. Conclusion: We identified intestinal barrier genes that were PPARα-dependently regulated after acute activation by fatty acids.This knowledge provides a better understanding of the impact dietary fat has on the barrier function of the gut, identifies PPARα as an important factor controlling this key function, and underscores the importance of PPARα for nutrient-mediated gene regulation in intestine. Project description:Accumulating evidence suggests that a compromised intestinal epithelial barrier (IEB) contributes to the progression of metabolic dysfunction-associated steatotic liver disease (MASLD); however, the exact mechanisms remain unclear. Here we reveal that intestinal mucin 1 (MUC1) levels and glycosylation are decreased in both humans and mice with MASLD. Enterocyte-specific Muc1 knockout aggravates high-fat diet (HFD)-induced IEB impairment and MASLD progression in mice. Mechanistically, HFD feeding reduces the glycosylation of intestinal epithelial MUC1, triggering its clathrin-mediated endocytosis and NEDD4-mediated lysosomal degradation, which subsequently induces β-Catenin degradation and ultimately impaires the IEB. Notably, enterocyte-specific overexpression of cytoplasmic-tail-deleted MUC1 protects against IEB impairment and mitigates MASLD progression. These findings indicate that reduced intestinal epithelial MUC1 levels facilitate the progression of MASLD. Preserving the glycosylation and levels of intestinal MUC1 to maintain IEB integrity is a potential therapeutic strategy to explore for MASLD.

Project description:Abstract: Our intake of ultra-processed foods has dramatically increased over the past few decades in line with the prevalence of obesity and diabetes, key risk factors for microvascular diseases such as chronic kidney disease (CKD). The extent to which long-term intake of highly processed food influences CKD outcome is unclear. Here, we show in rodent models that a highly processed diet drives intestinal barrier permeability and an increased risk of CKD. Inhibition of the advanced glycation pathway, which generates Maillard reaction products within foods upon thermal processing, reversed kidney injury. Consequently, a highly processed diet leads to innate immune complement activation and local kidney inflammation via the potent proinflammatory effector molecule complement 5a (C5a). C5a receptor inhibition ameliorated albuminuria. In a mouse model of diabetes, a high resistant starch fiber diet led to a redistribution of the gut commensal consortium, prevented impaired gut barrier function and decreased the severity of kidney injury via suppression of complement. These results provide mechanistic insight into the role of highly processed foods on inflammation and chronic disease risk. One Sentence Summary: Ultra-processed diets promote chronic kidney disease

Project description:Introduction: Intestinal epithelial cells (IECs) constitute a critical first line of defense against microbes by providing a physical barrier capable of producing antimicrobial peptides (AMPs) and cytokines. While IECs are known to respond to various microbial signals, the precise upstream cues regulating diverse IEC responses are not clear. Method: Mouse intestinal crypts were isolated from colon tissue. Crypts were differentiated into 3D spheroids in matrigel by stimulating with LWRN media and growth factors. After differentiation colonoids were stimulated with IL-1b, IL-22, or IL-1b and IL-22 for 12 hours then RNA was isolated for RNA sequencing. Conversely, endothelial maladaptation can lead to pathological angiogenesis and the perpetuation of inflammation in chronic inflammatory diseases. However, whether enteric ECs actively engage in the regulation of intestinal homeostasis and pathology through integration of environmental cues is currently unknown. Here, we show that the aryl hydrocarbon receptor (AHR), a ligand-activated transcription factor, acts as critical node for EC-sensing of dietary and microbial metabolites. We first established a comprehensive single-cell endothelial atlas of the mouse small intestine uncovering the cellular complexity and functional heterogeneity of blood and lymphatic ECs, identifying transcriptional networks and putative biological roles across endothelial subtypes. Analyses of AHR mediated responses at single-cell resolution identified tissue-protective transcriptional signatures and regulatory networks promoting quiescence and vasculoprotection. Endothelial AHR-deficiency in mice resulted in cellular activation, proliferation, and initiation of angiogenic pathways disrupting tissue homeostasis. In human ECs, AHR signalling promoted quiescence through cell cycle arrest and tempered endothelial inflammatory responses. Together, our data provide a comprehensive dissection of the impact of environmental sensing across the spectrum of enteric endothelia, demonstrating that endothelial AHR signalling serves to promote homeostasis and prevent aberrant inflammatory responses at the intestinal barrier.
