Prinary breast cancers and brain metastasis

Project description:Sixteen paired matched samples from primary breast cancers and brain metastases diagnosed between April 1, 2001 and December 31, 2012 were collected from 8 institutions. Brain metastases were identified based on magnetic resonance imaging and/or computed tomography findings. The clinical characteristics of all the patients were obtained from their medical records. This study was approved by the institutional review board of each participating institute (Tokai University School of Medicine; National Hospital Organization Osaka National Hospital; Kinki University School of Medicine; Niigata Cancer Center Hospital; Shizuoka General Hospital; Hokkaido Cancer Center; National Hospital Organization, Tokyo Medical Center; and Gunma Prefectural Cancer Center). Matching primary breast cancers and brain metastases Formalin-Fixed Paraffin-Embedded (FFPE) specimens for gene expression analysis were collected into RNA. RNA from specimens was isolated, and quantity and quality of the each RNA was using an Agilent 2100 Bioanalyzer (Agilent Technologies). Genome-wide expression levels of transcripts were analyzed using the Affymetrix U133A gene chips (Affymetrix) according to the manufacture’s instructions.

N-glycan profiling of Pseudomyxoma peritonei

Project description:Pseudomyxoma peritonei (PMP) is a subtype of mucinous adenocarcinoma of the appendix, We compared N-linked glycan profiles of PMP tissues to those of normal appendix. N-glycosidase F liberated N-glycan profiles of eight normal appendix samples and eight low-grade and eight high-grade PMP specimens were analyzed by MALDI-TOF mass spectrometry. The most prominent alteration was increased and complex fucosylation in high-grade carcinoma. Small RNA Solexa sequencing of human liver samples

Project description:We applied small RNA Solexa sequencing technology to identify microRNA expression in human liver samples from surgically removed liver tissues including three normal liver tissues (distal normal liver tissue of liver hemangioma), an hepatitis B virus (HBV)-infected liver, a severe chronic hepatitis B liver, two HBV-related hepatocellular carcinoma (HCC), an hepatitis C virus (HCV)-related HCC, and an HCC without HBV or HCV infection. All samples were collected with the informed consent of the patients and the experiments were approved by the ethics committee of Second Military Medical University, Shanghai, China. Project description:In this study, neutrophils isolated from human peripheral blood were cultured with PBS, tPA(10μg/ml) or tPA+HRG (0.5μm) for 2.5 hours at 37 ° C . Then, neutrophils were collected for proteomic analysis (n=6). Identification of Multiomics and Immune Infiltration-Associated Biomarkers for Gastric Cancer

Project description:Fresh peripheral blood was collected from gastric cancer patients in the second hospital of Lanzhou University. There were 30 patients with non metastatic gastric cancer (GCMN), 29 patients with healthy control group (NC), 31 patients with gastric mucosal lesions (GML), 8 patients with non metastatic gastric cancer for 1 month (gcmno), 11 patients with non metastatic gastric cancer for 4 months (gcmnsf), 15 patients with metastatic gastric cancer for 1 month (gcmso), 12 patients with metastatic gastric cancer for 4 months (gcmsf), and 15 patients with microsatellite unstable gastric cancer (gcmsi). Blood collection vessels containing coagulant were used to obtain serum. All patients had not received chemotherapy or radiotherapy before sampling, and individuals with autoimmune diseases, infectious diseases or multiple primary cancers were excluded. The clinical stage of the tumor was determined according to the TNM classification system of the International Anti Cancer Alliance (8th Edition). This study has been approved by the medical ethics committee of the second hospital of Lanzhou University (approval No. 2023A-459) and signed a written informed consent with each patient.

CircRNAs differentially expressed in brain metastases and primary lung cancer tissues of three pairs of non-small cell lung cancer patients with brain metastases

Project description:We searched for three NSCLC patients with brain metastases in the First Affiliated Hospital of Nanjing Medical University. After obtaining informed consent, the lung cancer brain metastasis surgical specimens and lung carcinoma in situ puncture specimens were collected from the three patients. The histopathological characteristics were examined using hematoxylin and eosin (H&E) staining. We searched for three pairs of differentially expressed RNAs in brain metastasis tissues and lung carcinoma in situ tissues by ceRNA microarray.
