Bone marrow Mac1 and Gr1 positive cells of CALR-deficient and wild-type mice

ABSTRACT: We analyzed the effect of Calr deficiency on Mac1 and Gr1 positive bone marrow cells using hematopoietic cell specific Calr knockout (Mx-cre;Calrf/-) mice and control (Mx-cre;Calr+/+) mice.

IL-4 stimulation of Dicer deficient bone-marrow derived macrophages in vitro

Project description:Macrophages were derived from the bone-marrow of 3 x fl/+ Dicer LysCre +/- (wild-type) and 3 x fl/fl Dicer LysCre +/- mice and stimulated with IL-4 (50ng/mL) for 72h. Total RNA was isolated and analyzed by gene array. In this experiment, we derived Dicer deficient bone-marrow macrophages using Dicer fl/+ LysM-Cre by Dicer fl/+ crossed mice to obtain Dicer fl/fl LysM-cre progeny (and Dicer deficient macrophages). Next, we studied the effects of IL-4 stimulation in macrophage with a deficiency in Dicer/microRNAs.

Project description:Seeking to challenge the current dogma that the nuclear core-complex proteins function in an entirely epistatic manner, Dr. Clapp's group developed a new double-knockout mouse nullizygous for Fancc and Fancg. Because the hematopoietic phenotype was more severe than single knockout mice, we reasoned that transcriptomal differences would exist and lead to the identification of molecular defects unique to each FA gene. RNA was purified from unfractionated and uncultured bone marrow cells from three types of Fanconi anemia gene knockout mice: nullizygous for Fancc, Fancg, or both Fancc and Fancg.. Three wild type C57Bl/6 mice served as controls. Each of three marrow samples provided one RNA sample (the RNA samples were not pooled).

In this study, we showed the BCL11A is required in vivo for developmental silencing of gamma-globin genes in adult animals. We used microarray to determine the changes in gene expression profile after loss of BCL11A in adult erythroid cells CD71+Ter119+ erythroid progenitor cells were FACS-sorted from bone marrows of 6-week old control (Bcl11a +/+) and BCL11A knockout (Bcl11a fl/fl EpoR-Cre+) mice.

Bone marrow-derived macrophage responses to classical activation: wild-type vs. MLL1-deficient (Lys2Cre MLL1 fx/fx)

Project description:mRNA from wild-type (Cre-) and MLL1-deficient (Cre+) BMDMs were analyzed via gene chip (Mouse Gene ST 2.1, Affymetrix) for relative expression changes. Isolated mRNA from Cre- and Cre+ BMDMs stimulated with classical activation signals (IFNg, LPS or IFNg+LPS) was analyzed using a gene chip panel of >40,000 RefSeq transcripts, and resulting fold expression was determined by analyzing quality-controlled expression values for validated probesets. Bone marrow derived macrophages from wild-type (Cre-) or MLL1-deficient (Cre+) mice were stimulated in vitro with IFNgamma (10 ng/ml), LPS (100 ng/ml) or the combination of IFNgamma+LPS for six hours. Cells were then processed in Trizol reagent for RNA extraction.

Single cell sequencing of murine hepatic CD45+ cells from mice transplanted with Trem2-deficient or wild-type bone marrow with NASH

Project description:We report the single cell transcriptome of CD45+ cells from the livers of bone marrow chimeric wildtype mice with NASH that had previously been transplanted with bone marrow from TREM2-deficient or wildtype littermate mice. We show that hematopoietic TREM2 deficiency is associated with alterations in the hepatic immune cell composition in the context of NASH.
