Mitochondrial proteome of human leuekmia.

ABSTRACT: Proteome characterization of isolated mitochondria samples prepared from three commonly used acute leukemia cell lines (HL-60, KG-1, MV-4-11). Data were compared to isolated mitochondria prepared from peripheral blood mononuclear cells (PBMC), isolated from healthy human subjects.

Currently there is great interest in targeting mitochondrial oxidative phosphorylation (OXPHOS) in cancer. However, notwithstanding the targeting of mutant dehydrogenases, nearly all hopeful 'mito-therapeutics' cannot discriminate cancerous from non-cancerous OXPHOS and thus suffer from a limited therapeutic index. The Illumina 450K methylation beadchip array was used to obtain DNA methylation profiles across approximately 450,000 CpG dinucleotide methylation loci in DNA isolated from PBMCs. Gene expression of CD8+ T cells isolated from human subjects during acute and convalescent phase of EBV infection

Project description:Affymetrix HuGene ST 1.0 microarrays were used to study and compare gene expression in peripheral blood CD8+ T cells of human patients with Acute Infectious Mononucleosis (AIM; acute EBV infection) and during convalescence (CONV; 6-12 months after AIM visit). Blood samples were drawn from ten human patients with AIM and again during their covalescence (CONV). Peripheral blood mononuclear cells were isolated and cryopreserved. Paired AIM and CONV samples were thawed and CD8+ T cells purified with magnetic beads. RNA was isolated and processed for hybridization according to the Affymetrix protocol CD8+ T cells acute and convalescent phase of EBV infection (10 subjects)

Gene expression profiling of acute myeloid leukemia samples from Polish adult AML-M1 and M2 patients

Project description:Acute myeloid leukemia (AML) is the most common and severe acute leukemia diagnosed in adults. Although it is one of the most studied cancers, this is the first study of Polish population of AML patients. The data were collected with the use of home-made boutique array. The additional advantage is pre-selection of patients - our sample set includes only two AML FAB subtypes with the highest content of immature myeloid cells: M1 (11 samples) and M2 (22 samples). From the majority of patients two sources of material were used for mononuclear cell separation and RNA extraction: bone marrow and peripheral blood. 15 samples from healthy volunteers were used as a control.

Project description:Early T-cell precursor acute lymphoblastic leukemia (ETP-ALL) is a high-risk T-ALL subtype with distinct immunophenotypes and genetic alterations, complicating diagnosis and treatment. Although cytogenetic and gene expression studies have advanced the diagnostic and therapeutic strategies, the proteomics signatures of ETP-ALL remain underexplored. Herein, we performed 4D label-free proteomic analyses on mononuclear cells isolated from ETP-ALL, T-ALL, acute myeloid leukemia (AML), and healthy donor samples to identify molecular signatures specific to ETP-ALL.

Project description:Gene expression of patient samples with acute myeloid leukemia (AML) were compared to normal controls to study dysregulated signalling pathways. Peripheral blood mononuclear cells (PBMCs) from primary AML patient samples were isolated using the Ficoll-Paque gradient separation method. RNA from CD34+ bone marrow cells of healthy donors were purchased from AllCells LLC (Alameda, CA; catalog number RNA-BM003C). Total RNA were harvested from patient samples using Trizol and subjected to microarray-based gene expression analysis.
