Project description:Development of the vertebrate hair bundle is a precisely orchestrated event that culminates in production of a tightly ordered arrangement of actin-rich stereocilia and a single axonemal kinocilium. To understand how the protein composition of the bundle changes during development, we isolated bundles from young (postnatal days P4-P6) and mature (P21-P25) mouse utricles using the twist-off method, then characterized their constituent proteins using liquid-chromatography tandem mass spectrometry with data-dependent acquisition. Using label-free quantitation, we measured relative abundances of proteins in both bundles and in the whole utricular epithelium; comparison of protein abundance between the two fractions allows calculation of enrichment in bundles. These data will be useful for examining the proteins present in mammalian vestibular bundles and how they change over development.

Project description:Transcriptional profiling of the mantle tissue across the four stages of female gonads development (winter peak) in a natural population of the marine mussel Mytilus galloprovincialis sampled in the Bizerta Lagoon, Tunisia, across November 2007 -March 2008. Background: Seasonal environmental changes may affect the physiology of Mytilus galloprovincialis (Lam.), an intertidal filter-feeder bivalve occurring commonly in Mediterranean and Atlantic coastal areas. We investigated seasonal variations in relative transcript abundance of the digestive gland and the mantle (gonads) of males and females. To identify gene expression trends, we used a medium-density cDNA microarray (1.7 K probes) in dual-color competitive hybridization analyses. Results: Hierarchical clustering of digestive gland microarray data showed two main branches, distinguishing profiles associated with the “hot” months (May–August) from the other months. Genes involved in chitin metabolism, associated with mussel nutrition and digestion, showed higher expression during summer. Moreover, we found different gene expression patterns in the digestive glands of males and females during the four stages of mussel gonadal development. Microarray data from gonadal transcripts also displayed clear patterns during the different developmental phases with peak relative mRNA abundance at the ripe phase (stage III) for both sexes. Conclusion: These data showed a clear temporal pattern in gene expression profiles of mussels sampled over an annual cycle. Physiological response to thermal variation, food availability, and reproductive status across months may contribute to variation in gene expression. Mantle tissue from individual animals in different gonad maturation stages were analyzed in a complete loop design. Dual color competitive hybridizations (stage 4 vs stage 1, 2 vs 1, 3 vs 2, 4 vs 3) including label swap. Single individuals. Four biological replicates. One replicate per array.

Project description:Gene expression changes during biofilm formation processes were investigated. The gene expression was compared at attachment, colony formation and maturation during biofilm formation. At the same time, the gene expressions were also compared with exponential phase and stationary phase in planktonic cells. The gene expression pattern at attachment and colony formation processes showed similar pattern with those in planktonic exponential phase, and the gene expression pattern at maturation process showed similar pattern with those in planktonic stationary phase. During the maturation process, metabolic activities of the cells in the biofilms decreased, and the genes involved in the anaerobic respiration and efflux pumps were induced. The analysis revealed that gene expression pattern was changed and the physiological states were changed dramatically during maturation process in the biofilms. Keywords: time course

Project description:Studies have been done to search for changes in miRNA expression during the muscle development in different pigs. However, research in this area has largely focused on dissecting miRNAs that are important for fetal and young muscle development. Little is known about the role of miRNAs in adult muscle development. In this study, we performed a comprehensive study on miRNA expression pattern across the muscle developmental process from embryo to adult (E90-7Years). We identified a number of differentially expressed muscle development-associated miRNAs including new members during developmental stages.
