Mitochondrial Proteomics in a murine model of CKD

ABSTRACT: This project involves TMT-labeled proteomics analysis of mitochondria isolated from skeletal muscle, heart, and kidney of mice with and without chronic kidney disease (CKD). CKD was induced by adenine-supplementation of the diet for 6 months. Mouse strain was C57BL6J mice from Jackson Lab, aged 6 months prior to start of the diet. Mitochondria were isolated from skeletal muscle, heart, and kidney and immediately snap frozen and stored at -80C until processing.

Transcription profiling of mouse heart, brain, and skeletal muscle to determine the rffect of age, calorie restriction and resveratrol on gene expression

Project description:Resveratrol in high doses has been shown to extend lifespan in some studies in invertebrates and to prevent early mortality in mice fed a high-fat diet. We fed mice from middle age (14-months) to old age (30-months) either a control diet, a low dose of resveratrol (4.9 mg kg-1 day-1), or a calorie restricted (CR) diet and examined genome-wide transcriptional profiles. We report a striking transcriptional overlap of CR and resveratrol in heart, skeletal muscle and brain. Both dietary interventions inhibit gene expression profiles associated with cardiac and skeletal muscle aging. Gene expression profiling suggests that both CR and resveratrol may retard some aspects of aging through alterations in chromatin structure and transcription. Resveratrol, at doses that can be readily achieved in humans, fulfills the definition of a dietary compound that mimics some aspects of CR. Project description:Study designed to identify genes that are induced in both BAT and skeletal muscle during acute adaptive thermogenesis in mouse using gene expression microarray. Animal studies were performed under approved UCLA animal research protocols and according to guidelines established in the Guide for Care and Use of Laboratory Animals. C57BL/6J mice were maintained in 12-h light/dark conditions and fed a regular chow diet (Purina 50010, Lab Diet, USA). Cold exposure was performed as described. After euthanasia, tissues were collected and frozen until use. Total RNA was isolated from mouse tissues by extraction with TRIzol from either brown adipose tissue (BAT) or skeletal muscle (quadriceps) after exposure to cold (4 degrees C for 4 hours). The mice were of the C57BL/6J strain and were maintained in 12-h light/dark conditions and fed a laboratory chow diet that consisted of 4.5% fat, 50% carbohydrate by weight (Lab Diet, Purina 50010).

Effect of in utero undernutrition on skeletal muscle gene expression

Project description:In utero undernutrition is associated with obesity and insulin resistance, although its effect on skeletal muscle remains poorly defined. We report that, in mice, adult offspring from undernourished dams have decreased energy expenditure, decreased skeletal muscle mitochondrial content, and altered energetics in isolated mitochondria and permeabilized muscle fibers. Strikingly, when these mice are put on a 40% calorie restricted diet they lose half as much weight as calorie restricted controls. Our results reveal for the first time that in utero undernutrition alters metabolic physiology having a profound effect on skeletal muscle energetics and response to calorie restriction in adulthood. We have used a mouse model of low birth weight generated through 50% food restriction of mouse dams during the third week of gestation. We have studied in utero food restricted offspring and control offspring that were not food restricted in utero in both the ad libitum and calorie restricted states. Gene expression profiling was performed on tibialis anterior muscle from 8 mice per group, pooled in pairs.

Ketone flux through BDH1 supports metabolic remodeling of skeletal and cardiac muscles in response to intermittent time-restricted feeding

Project description:Time-restricted feeding (TRF) has gained attention as a dietary regimen that promotes metabolic health. This study sought to determine if ketone flux in skeletal and cardiac muscles plays an essential role in conferring the health benefits of an intermittent TRF (iTRF) schedule. Notably, we found that the ketolytic enzyme, beta-hydroxybutyrate dehydrogenase 1 (BDH1), is uniquely enriched in isolated mitochondria derived from heart and red/oxidative skeletal muscles, which also have high capacity for fatty acid oxidation (FAO). Using mice with muscle/heart-specific BDH1 deficiency, we discover that this enzyme is required for optimal FAO efficiency and exercise tolerance during acute fasting. Additionally, iTRF leads to robust molecular remodeling of muscle tissues and BDH1 flux is indeed required for the full adaptive benefits of this regimen, including increased lean mass, mitochondrial hormesis, and metabolic rerouting of pyruvate. In sum, ketone utilization enhances mitochondrial bioenergetics and supports iTRF-induced remodeling of skeletal muscle and heart.

Profiling carbonylated proteins in heart and skeletal muscle mitochondria from trained and untrained mice

Project description:Understanding the relationship between physical exercise, reactive oxygen species and skeletal muscle modification is important in order to better identify the benefits or the damages that appropriate or inappropriate exercise can induce. Heart and skeletal muscles have a high density of mitochondria with robust energetic demands and mitochondria plasticity has an important role in both cardiovascular system and skeletal muscle responses. The aim of this study was to investigate the influence of regular physical activity on oxidation profiles of mitochondrial proteins from heart and tibialis anterior muscles. To this end, we used mouse as animal model. Mice were divided in two groups: untrained and regularly trained. The carbonylated protein pattern was studied by two-dimensional gel electrophoresis followed by Western Blot with anti-dinitrophenyl hydrazone antibodies. Mass spectrometry analysis allowed the identifications of several different protein oxidation sites including methionine, cysteine, proline and leucine residues. A large number of oxidized protein were found in both untrained and trained animals. Moreover, mitochondria from skeletal muscles and heart showed almost the same carbonylation pattern. Interestingly, exercise training seems to increase carbonylation level mostly of mitochondrial protein from skeletal muscle.

Project description:To investigate microRNAs related to mitochondria biogenesis in skeletal muscle, microRNA expressions during skeletal muscle differentiation and exercise were analyzed in vivo and in vitro. Murine skeletal muscle cell (C2C12) were assigned to undifferentiated, differentiated, and passively stretched (exercise mimicked). C57BL/6S mice were assigned to resting, acute exercise (1day), and chronic exercise (7days). Low molecular weight RNA (< 200 nucleotides) was isolated from C2C12 cell or tibialis anterior muscle of mice and hybridized to Ncode microRNA microarrays. The experiment was performed using a loop design for the data analysis.
