A comparison of mono-culture and co-culture model containing fibroblasts

Project description:Organotypic three dimensional cultures of epithelial cells are grown at the air–liquid interface (ALI) and resemble the in vivo counterparts. Although the complexity of in vivo cellular responses could be better manifested in co-culture models in which additional cell-types such as fibroblasts were incorporated, the presence of another cell-type could mask the response of the other. This study reports the impact of whole combustible cigarette smoke (CS) on organotypic mono- and co-culture models to evaluate the relevancy of organotypic models for toxicological assessment of aerosols. Two organotypic bronchial models were directly exposed to low and high concentrations of CS of the reference research cigarette 3R4F: mono-culture of bronchial epithelial cells without fibroblasts (BR) and co-culture with fibroblasts (BRF) models. Adenylate kinase-based cytotoxicity, cytochrome P450 (CYP) 1A1/1B1 activity, tissue histology, and concentrations of secreted mediators in the basolateral media, as well as transcriptomes were evaluated following the CS exposure. The results demonstrated similar impact of CS on the AK-based cytotoxicity, CYP1A1/1B1 activity, and tissue histology in both models. However, a greater number of secreted mediators were found in the basolateral media of the mono-culture than in the co-culture models. Furthermore, annotation analysis and network-based systems biology analysis of the transcriptomic profiles indicated a more prominent cellular stress and tissue damage following CS in the mono-culture epithelium model without fibroblasts. Finally, our results indicated that the in vivo smoking-induced xenobiotic metabolism response in the bronchial epithelial cells was better reflected on the in vitro co-culture model upon CS exposure.

A normalization method to decipher quantitative proteome changes in microbial co-culture systems

Project description:The value of synthetic microbial communities in biotechnology is gaining traction due to their ability to undertake more complex metabolic tasks than monocultures. However, a thorough understanding of strain interactions, productivity and stability is often required to optimize growth and scale up cultivation. Quantitative proteomics can provide valuable insights into how microbial strains adapt to changing conditions in biomanufacturing or bioremediation scenarios. However, current workflows and methodologies are not suitable for simple artificial co-culture systems where strain ratios are dynamic. Here, we established a standard workflow for co-culture proteomics using an exemplar system containing two key members, Azotobacter vinelandii and Synechococcus elongatus. Factors affecting the quantitative accuracy of co-culture proteomics were investigated, including peptide physicochemical characteristics such as molecular weight, isoelectric point, hydrophobicity, and dynamic range, as well as factors relating to protein identification such as varying proteome size and shared peptides between species. Different quantification methods based on spectral counts and intensity were evaluated, demonstrating good correlations between protein amount and the six quantification methods at the protein level. We propose a new normalization method, named “LFQRatio”, to reflect the relative contributions of the two distinct cell types emerging from the cell ratio changes during co-cultivation. LFQRatio can be applied to real co-culture proteomics experiments, providing accurate insights into quantitative proteome changes in each strain.

Project description:Rho-GDP dissociation inhibitors (RDI) are repressors of Rho-type monomeric GTPases that allow for precise control of their target processes, e.g. cytoskeletal arrangement, vesicle trafficking, and polarized growth. In the human pathogenic yeast Cryptococcus neoformans, maintenance of normal cell morphology is vital for pathogenicity. We identified and deleted the gene encoding an RDI homolog in the human fungal pathogen Cryptococcus neoformans and investigated its impact on pathogenicity in animal models of cryptococcosis. Rdi1 deletion resulted in altered vacuole size in tissue culture medium, with corresponding alterations in expression of vesicle trafficking related genes. The rdi1∆ mutant strain showed reduced intracellular survival in macrophages, and severe attenuation of virulence in murine models of cryptococcosis. This reduction in virulence of the rdi1 mutant occurs in the absence of major defects in growth, morphology, or classical virulence-associated phenotypes.
