The nutrient solution contains 0.25 mM Ca(NO3)2.4H2O, 1mM KH2PO4, 0.5 mM KNO3, 1mM MgSO4.7H2O, 50 µM H3BO3, 19 µM MnCl2.4H2O, 10 µM ZnCl2, 1 µM CuSO4.5H2O, 0.02 µM Na2MoO4.2H2O and 50 µM FeNa-EDTA. Plants were subjected to an 8 h light/16 h dark cycle, at 19°C, with 70% relative humidity. Leaves of six week old hydroponically grown A. thaliana Col0 plants were infiltrated with 1mM deferoxamine or sterile distilled water. Leaves were harvested 7 and 24 h.p.i. Project description:This project details a quantitative phosphoproteomic analysis of PC3 cells treated with the compound KBU2046. The study evaluated cellular responses to vehicle control, 1 µM KBU2046, and 10 µM KBU2046 at three time points (15 minutes, 1 hour, and 4 hours) using three biological replicates per condition. To enable robust comparative quantification, samples were multiplexed using Tandem Mass Tag 10 plex reagents. Phosphorylated peptides were enriched utilizing titanium dioxide affinity chromatography prior to analysis on an Orbitrap Eclipse Tribrid mass spectrometer operating in data-dependent acquisition mode with synchronous precursor selection MS3 quantitation.

Project description:To compare the genome-wide transcriptional effect of ABA and iSB09 in tomato plants, we performed RNA-seq analysis of mock-, 10 uM ABA- or 20 uM iSB09-treated plants. Differential gene expression analysis between mock- and ABA-treated or iSB09-treated seedlings was done with DESeq2 and genes with an absolute value of log2 fold change (log2FC) > 1 or (log2FC) < -1 and p-adjusted value (padj) < 0.05 were selected. iSB09 upregulated and downregulated genes represent a subset of the ABA-responsive genes, which reflects the activation of PYL1-like and PYL4-like ABA receptors in tomato seedlings.
