ABSTRACT: we performed a label-free quantitative mass spectrometry-based proteomic analysis comparing the dry seed proteome of four genotypes in three independent biological replicates: Col-0, dwa1 dwa2 double mutant, and YFP-AFP2 over expressing lines in both Col-0 and dwa1 dwa2 backgrounds. This assay aimed to identified protein accumulation pattern correlating with seed longevity and or ABA resistance during germination.

Overexpression of ABA-INSENSITIVE5 binding proteins (AFPs) results in extreme ABA resistance of seeds and failure to acquire desiccation tolerance, at least in part through effects on chromatin modification. We tested the hypothesis that AFPs promote germination in Arabidopsis (Arabidopsis thaliana) by also functioning as adapters for E3 ligases that ubiquitinate ABI5, leading to its degradation.
