Conditioned medium from bone marrow derived macrophages

ABSTRACT: Conditioned medium (CM) from bone marrow derived macrophages untreated or treated with LPS was collected and filtered through a 0.22-μm filter. The filtered CM was sequentially fractionated with 50-kDa and 100-kDa Amicon filters. The 50–100 kDa fraction of CM was analyzed by mass spectrometry.

Project description:We have employed whole genome microarray expression profiling to identify differently expressed genes following stimulation of wild-type bone-marrow derived macrophages with Escherichia coli lipopolysaccharide (LPS). Macrophages were stimulated with LPS (100 ng/ml) for 6 hours and a signature was identified that distinguished between infected and control samples. Expression of several genes from this signature was quantified in the same RNA samples by real-time PCR, confirming the predicted macrophage response pattern.

RNAseq analysis of GM-CSF and M-CSF polarized bone marrow derived macrophages cultured on TC or noTC dishes

Project description:Bone marrow cells were isolated, primed with M-CSF (M BMDM) or GM-CSF (GM BMDM) and cultured on tissue culture dishes (TC) or low adherent dishes (noTC) for 7 days. BMDMs were treated with LPS (100 ng/ml) for 3 hours. The difference between TC- and noTC-cultured BMDMs were analyzed to describe the phenotye and function of each population. Moreover, the influence of LPS treatment on GM-BMDMs and M-BMDMs were analyzed.

Mouse macrophages from bone marrow treatment

Project description:The noncanonical inflammasome induced by intracellular lipopolysaccharide (LPS) leads to caspase-11-dependent pyroptosis, which is critical for induction of endotoxic shock in mice. However, the signaling pathway downstream of caspase-11 is unknown. We found that cytosolic LPS stimulation induced caspase-11-dependent cleavage of the pannexin-1 channel followed up by ATP release, which in turn activated the purinergic P2X7 receptor to mediate cytotoxicity. In the absence of P2X7 or pannexin-1, pyroptosis induced by cytosolic LPS was abrogated. Cleavage of pannexin-1 required the catalytic activity of caspase-11 and was essential for ATP release and P2X7-mediated pyroptosis. Priming the caspase-11 pathway in vivo with LPS or Toll-like receptor-3 (TLR3) agonist resulted in high mortality in wild-type mice after secondary LPS challenge, but not in Casp11(-/-), Panx1(-/-), or P2x7(-/-) mice. These results reveal a critical role for pannexin-1 and P2X7 downstream of caspase-11 for pyroptosis and susceptibility to sepsis induced by the noncanonical inflammasome. Project description:In this experiment we investigated the effect of HDAC3 inhibition on the transcriptome of IFNg-primed macrophages under different tolerization conditions. Peripheral blood mononuclear cells (PBMCs) were isolated from 3 healthy donors. PBMCs were isolated from whole blood of healthy donors using Ficoll gradient (Invitrogen). Monocytes (CD14+ cells) were positively selected from PBMCs using CD14 Microbeads according to the manufacturer’s instructions (Miltenyi Biotec). Monocytes were subsequently treated with or without 500 nM HDAC3i (ITF3100) for 30 minutes prior to overnight IFNg priming (50 ng/mL). Cells were then kept without LPS (non-LPS; N), treated with 10 ng/mL LPS once (non-tolerized; NT), or treated with LPS twice (tolerized; T; second LPS concentration: 100 ng/mL).
