Project description:The development and propagation of an adaptive immune response to an invading pathogen is a highly orchestrated process that involves the precise regulation of cytokine expression. Naïve CD4+ T lymphocytes give rise to T helper (Th) cell subsets with functions that are tailored to their respective roles in host defense. MicroRNAs are important regulators of most cellular processes, including many responses in the immune system. To identify novel microRNAs that might be important in human T helper cell differentiation to different subsets we purified T cell subsets from peripheral blood and performed microRNA arratys at Exiqon. Naïve, Th1, Th2, Th17 and Tregs were FACS-sorted ex-vivo from peripheral blood of 6-11 donors. Due to the very low amount of starting material total RNA from at least 6 different donors was pooled and anlaysed on the arrays. All samples were analyzed against a common reference, which was made by pooling together a small amount of total CD4+ cells from all donors.

Multiple functionally distinct helper CD4+ T cell subsets can arise in response to a single invading pathogen, complicating the identification of rare memory CD4+ T cells. Here we found that expression of Id3, an inhibitor of E protein transcription factors, identified a population of cells within both the CD4+ Tfh and Th1 helper lineages that exhibited memory potential in response to secondary infection. Notably, a subset of Th1 memory cells expressing Id3 exhibited enhanced expansion upon response to pathogen, generating both Th1 and Tfh secondary effector cell populations, and displayed enrichment of key molecules associated with memory potential when compared to Id3lo Th1 cells. Thus, Id3 expression serves as an important marker of multipotent memory CD4+ T cells.

Project description:Plant guard cells perceive pathogens and close stomata to prevent invasion. Biomolecular condensates are central to life growth and stress adaptation, while their roles in regulating stomatal immunity remain unclear. Here, we identify a guard cell-preferential RRM-type RNA-binding protein (RBP) named SAIR1 (stomatal immune RBP 1). Upon pathogen infection and the pathogen-associated molecular pattern flg22 signaling, SAIR1 undergoes phase separation and forms condensates in guard cells to mediate stomatal immunity. Upon flg22 signals, the activated kinases MPK3/6 phosphorylate SAIR1 to initiate its condensation in guard cells. SAIR1 condensates associate with critical defense mRNAs, including those involved in salicylic acid (SA) signaling. SAIR1 interacts with pivotal immune translation regulators such as PABPs and eIFiso4G, and enhances translation of defense mRNAs, ultimately promoting stomatal closure. Our findings reveal that SAIR1 phosphorylation, condensation, and interactions with mRNAs and translation regulators link flg22-MPK3/6 signaling with SA pathways to mediate stomatal immunity.

Project description:The capacity for T cells to become activated and clonally expand during pathogen invasion is pivotal for protective immunity. Our understanding of how T cell receptor (TCR) signaling prepares cells for this rapid expansion remains limited. We have identified an E3 ubiquitin ligase, Cul4b, that regulates this process. Cul4b levels and activity increase following TCR stimulation. Disruption of Cul4b resulted in impaired proliferation and survival of activated T cells. Additionally, Cul4b-deficient CD4 T cells accumulated DNA damage. In T cells, Cul4b preferentially associated with the substrate receptor DCAF1, and Cul4b and DCAF1 were found to interact with proteins that promote the sensing or repair of damaged DNA. While Cul4b-deficient CD4 T cells showed evidence of DNA damage sensing, downstream phosphorylation of SMC1A did not occur. These findings reveal an essential role for Cul4b in promoting the repair of damaged DNA to allow survival and expansion of activated T cells.

Project description:Defense against attaching and effacing (A/E) bacteria requires the sequential generation of IL-23 and IL-22 to induce protective mucosal responses. While the critical source of IL-22 has been identified as CD4+ and Nkp46+ innate lymphoid cells (ILCs), the precise source of IL-23 is unclear. Here, we use genetic techniques to deplete specific classical dendritic cell (cDC) subsets and analyze immunity to the A/E pathogen Citrobacter rodentium. We find that Zbtb46+ cDCs, and specifically Notch2-dependent intestinal CD11b+ cDCs, but not Batf3-dependent CD103+ cDCs, are required for IL-23 production and immunity against C. rodentium. Notch2 controls cDC differentiation at a terminal step mediated by lymphotoxin signaling. Importantly, these results provide the first demonstration of a non-redundant function of CD11b+ cDCs in vivo. Analysis of differentially expressed genes in ESAM+ and ESAM- CD11b+ and DEC205+ splenic classical DC subsets. Splenocytes were harvested from WT C57Bl/6 or WT Cx3cr1-gfp mice and cDC subsets sorted to >95% purity on the FACSAriaII.

Project description:Tissue mononuclear phagocytes (MNP) are specialised in pathogen detection and antigen presentation. They are the first cells of the immune system to encounter HIV and play a key role in transmission as they deliver the virus to CD4 T cells, which are the primary HIV target cell in which the virus undergoes replication. Most studies have investigated the role that epithelial MNPs play in HIV transmission but, as mucosal trauma and inflammation are strongly associated with HIV transmission, it is also important to examine the role that sub-epithelial MNPs play. Sub-epithelial MNPs are present in a diverse array of subsets which differ in their function and the pathogens they detect. Understanding how specific subsets interact with HIV and deliver the virus to CD4 T cells is therefore of key importance to vaccine and microbicide development. In this study we have shown that, after topical application, HIV can penetrate to interact with sub-epithelial resident myeloid cells in anogenital explants and defined the full array of MNP subsets that are present in all the human anogenital and colorectal sub-epithelial tissues that HIV may encounter during sexual transmission. In doing so we have identified two subsets that preferentially take up HIV, become infected and transmit the virus to CD4 T cells; CD14+CD1c+CD11c+ monocyte-derived dendritic cells and langerin-expressing conventional dendritic cells 2 (cDC2).
