Project description:The transplacental transfer of maternal IgG to the developing fetus is critical for infant protection against infectious pathogens in the first year of life. However, factors that modulate the transplacental transfer efficiency of maternal IgG that could be harnessed for maternal vaccine design remain largely undefined. HIV-infected women have impaired placental IgG transfer, yet the mechanism underlying this impaired transfer is unknown, presenting an opportunity to explore factors that contribute to the efficiency of placental IgG transfer. We measured the transplacental transfer efficiency of maternal HIV and other pathogen-specific IgG in historical U.S. (n=120) and Malawian (n=47) cohorts of HIV-infected mothers and their HIV58 exposed uninfected and HIV-infected infants. We then examined the role of maternal HIV disease progression, infant factors, placental Fc receptor expression, and IgG Fc region subclass and glycan signatures and their association with transplacental transfer efficiency of maternal antigen-specific IgG. We established 3 distinct phenotypes of placental IgG transfer efficiency in HIV-infected women, including: 1) efficient transfer of the majority of antigen-specific IgG populations; 2) generally poor IgG transfer phenotype that was strongly associated with maternal CD4+ T cell counts, hypergammaglobulinemia, and frequently yielded non-protective levels of vaccine-specific IgG; and 3) variable transfer of IgG across distinct antigen specificities. Interestingly, maternal IgG characteristics, such as binding to placentally expressed Fc receptors FcgRIIa and FcgRIIIa, IgG subclass frequency, and Fc region glycan profiles were associated with placental IgG transfer efficiency. These maternal IgG transplacental transfer determinants were distinct among different antigen-specific IgG populations. Our findings suggest that in HIV-infected women, both maternal disease progression and Fc region characteristics modulate the selective placental transfer of distinct IgG subpopulations, with implications for both the health of HIV-exposed uninfected infants and maternal vaccine design.

Project description:Immunoglobulin Gs (IgGs) have become highly successful drug scaffolds by combining very specific target binding with the ability to induce cellular cytotoxicity. Of further advantage, IgGs possess unusually long half-lives in the blood (2-3 week). IgGs achieve such extraordinary half-lives through a pH-dependent interaction with the Neonatal Fc receptor (FcRn) whereby IgGs are recycled. No high-resolution structure of FcRn in complex with a full-length IgG is available, and the interaction was long thought to be mediated solely via the IgG Fc. However, some IgGs with identical Fc parts, but different Fab domains, exhibit different half-lives, suggesting involvement of the Fab domains in FcRn binding. Here, we have employed a combination of HDX-MS and cross-linking MS (XL-MS) to explore the interaction of a full-length IgG with FcRn. HDX-MS and XL-MS analysis confirms an interaction between FcRn and the Fc-domain of IgGs, as a reduction of HDX was observed in both the Fc-domain and FcRn upon complex formation, and three cross-links were identified between FcRn and the Fc-domain. However, FcRn-induced changes in HDX were also observed in the Fab domains, supported by multiple cross-links between the Fab domains and the C-terminal α3 domain of FcRn. Our results thus provide direct evidence for a Fab-FcRn interaction. We envision that these result could advance the engineering of therapeutic IgGs with tailored pharmacokinetics and enhanced efficacy.

Project description:In this study, we introduce a targeted microflow LC-MS/MS-PRM method for the quantification of multiple glycopeptides in unfractionated serum samples. The entire preparation of 16 samples in a batch is completed within 3 h, and the LC-MS quantification of all the glycoforms in a sample is completed in 15 min in triplicate, including online capture and desalting. We demonstrate applicability of the workflow on a multiplexed quantification of eight N-glycoforms of immunoglobulin G (IgG) together with two O-glycoforms of hemopexin (HPX). We applied the assay to a serologic study of fibrotic liver disease in patients of HCV etiology. The results document that specific IgG- and HPX-glycoforms detect efficiently fibrotic disease of different degree, and suggest that the LC-MS/MS-PRM assays may provide rapid and reproducible biomarker assay targeting simultaneously the N- and O-glycoforms of the peptides.

Project description:Vaccination triggers the production of antigen-specific antibodies, including IgG. IgG molecules are glycosylated at the Fc region, and these glycan modifications markedly influence Fc receptor binding and downstream immune functions. Notably, infections with enveloped viruses such as SARS-CoV-2 can trigger the production of afucosylated IgG, which enhances FcγRIIIa binding and promotes antibody-dependent cellular cytotoxicity. Despite its importance, the glycosylation profiles of antigen-specific IgG following vaccination remain understudied, particularly in animal models. In this study, we investigated the Fc glycosylation patterns of antigen-specific IgG in hamsters and rhesus macaques following immunization with the Ad26.COV2.S COVID-19 vaccine. Overall, our findings demonstrate that IgG Fc glycosylation dynamics in these animal models largely parallel those in humans. For example, we observed a transient afucosylated IgG response in both species, resembling the response previously reported in humans. These results indicate that IgG Fc glycosylation responses to vaccination in macaques and hamsters recapitulate key features of the human response, supporting their use as translational models for adenovirus vector-based vaccination studies.

Project description:Fc γ-receptor IIIa (FcγRIIIa, or CD16a) is a receptor for the Fc region of IgG antibodies and plays an essential role in the regulation of antibody effector functions. While the activation of CD16a plays a role in a healthy immune response, dysregulated CD16a activity may contribute to tissue damage and chronic inflammation by over-activating immune cells or, conversely, by compromising the removal of immune complexes. Functionally distinct glycosylated forms of CD16a exist, with CD16a glycoforms modified with high mannose N-linked glycans at site N162 demonstrating a higher binding affinity for IgG compared to other forms of CD16a. Here, we developed and tested a nano-liquid chromatography tandem mass spectrometry (nLC-MS/MS) parallel reaction monitoring (PRM) method for the site-specific study of CD16a glycosylation. The PRM method was tested by spiking recombinant CD16a (F158 or V158) into a complex matrix consisting of THP-1 cell lysate digest, then the CD16a glycopeptide PRM method was applied to measure statistically significant changes in site N162 N-glycan compositions corresponding to high affinity glycoforms of CD16a in a model of monocyte differentiation. Analyses were extended to the analysis of CD16a derived from human primary monocytes and peripheral blood mononuclear cells. CD16a site N162 N-glycosylation demonstrated remarkable similarity across the surveyed cell types, including in THP-1 cells – a monocyte cell line isolated from the peripheral blood of an acute monocytic leukemia patient, as well as primary human monocytes, and PBMCs. Critically, CD16a glycopeptide PRM facilitated measurement of changes in CD16a glycosylation at site N162, with high mannose N-glycan compositions, corresponding to high affinity glycoforms of CD16a, increasing in a model of monocyte differentiation into macrophages. Here, we have demonstrated the feasibility of utilizing a glycopeptide nLC-MS/MS-PRM targeted approach to study CD16a glycosylation in material from primary cells in a glycosylation site-specific manner. This strategy is well-suited for adaptation to clinical studies focused on understanding the role of CD16a glycosylation in physiology and disease, with a focus on immune disorders.

Project description:We report the comparison of mass-spectral-based abundances of tryptic glycopeptides with fluorescence abundances of released labeled glycans, both derived from therapeutic monoclonal antibodies. These proteins were selected for their very high purities, which assures the origin of all observed released glycans. After excluding in-source fragmentation of glycopeptide ions from the results, a linear correlation was observed between fluorescently labeled N-glycan and glycopeptide abundances over a dynamic range of 500. The primary glycoforms derived from Rituximab, NISTmAb, Evolocumab, and Infliximab were high mannose and biantennary complex galactosylated and fucosylated and, in some cases, sialylated N-glycans. Except for Evolucumab, in-source ions derived from the loss of HexNAc or HexNAc and hexose sugars to HexNAc(4)Hex(4)Fuc(1) glycoform are prominent for other therapeutic IgGs. After excluding in-source fragmentation of glycopeptide ions from the results, a linear correlation was observed between fluorescently labeled N-glycan and glycopeptide abundances across all mAb samples over a dynamic range of 500. Different charge states of human IgG-derived glycopeptides containing a wider variety of abundant attached glycans were also investigated closely to examine the effect of charge state on ion abundances. These results confirmed and refined results for the therapeutic proteins, clearly revealing a linear dependence of relative glycopeptide abundance on the mass of the glycan, with higher charge states favoring higher mass glycans. Overall, our findings indicate that the mass-spectrometry-based bottom-up approach can provide results as accurate as those of glycan release studies while revealing the origin of each attached glycan. These site-specific relative abundances are conveniently displayed and compared using previously described Glycopeptide Abundance Distribution Spectra GADS representations.

Project description:Autoantibodies to nuclear antigens are hallmarks for diagnosis of the autoimmune disease systemic lupus erythematosus (SLE) and they contribute to SLE pathogenesis. However, there remains a gap in our knowledge regarding how different isotypes of autoantibodies contribute to key disease processes, including the production of the critical type I interferon (IFN) cytokines by plasmacytoid dendritic cells (pDCs) in response to immune complexes (ICs). Here, we show that individuals with SLE have IgA autoantibodies against most nuclear antigens assessed, largely correlating with the amounts of IgG against the same antigen. We investigated whether IgA autoantibodies against a major SLE autoantigen, Smith ribonucleoproteins (Sm/RNPs), play a role in IC activation of pDCs. We found that pDCs express the IgA-specific Fc receptor, FcRI, and there was a striking ability of IgA1 isotype autoantibodies to synergize with IgG in RNA-containing ICs to generate robust pDC IFN responses. pDC responses to these ICs required both FcRI and FcRIIa, suggesting the most potent ICs for pDCs contained autoantibodies of both isotypes. Sm/RNP IC binding to and internalization by pDCs were greater when ICs contained both IgA1 and IgG. In addition, binding of Sm/RNP ICs generated with IgA1-sufficient serum correlated to pDC FcRI expression, but not FcRIIa expression. pDCs from individuals with SLE had higher binding of IgA1-containing ICs and higher expression of FcRI than pDCs from healthy control individuals, correlating with expression of IFN response genes. Taken together, our data indicate that IgA1 ANAs contribute to SLE pathology and warrant further investigation.
