A multiplexed microflow LC-MS/MS-PRM assay for serologic quantification of IgG N- and HPX O- glycoforms in liver fibrosis

Project description:In this study, we introduce a targeted microflow LC-MS/MS-PRM method for the quantification of multiple glycopeptides in unfractionated serum samples. The entire preparation of 16 samples in a batch is completed within 3 h, and the LC-MS quantification of all the glycoforms in a sample is completed in 15 min in triplicate, including online capture and desalting. We demonstrate applicability of the workflow on a multiplexed quantification of eight N-glycoforms of immunoglobulin G (IgG) together with two O-glycoforms of hemopexin (HPX). We applied the assay to a serologic study of fibrotic liver disease in patients of HCV etiology. The results document that specific IgG- and HPX-glycoforms detect efficiently fibrotic disease of different degree, and suggest that the LC-MS/MS-PRM assays may provide rapid and reproducible biomarker assay targeting simultaneously the N- and O-glycoforms of the peptides.

Identification and relative quantification of hFSH glycoforms in women sera by PRM MS based approach.

Project description:This study presents an alternative preliminary strategy to identify and quantify serum hFSH glycoforms based on immunopurification assay and mass spectrometry (MS) using parallel reaction monitoring (PRM) analysis. In this study, we provide MS-PRM data acquisition method for hFSH glycopeptides identification with high specificity and their quantification by extracting the chromatographic traces of selected fragments of glycopeptides. Once set up in all its features, the proposed method could be transferred to the clinic to improve the fertility treatments and follow-ups in men and women.

Human serum glycopeptide LC-MS/MS

Project description:The heterogeneity caused by post-translational modifications and wide dynamic range of relative protein abundances of the human serum proteome, challenge the capabilities of existing mass spectrometry-based methodologies in accurate identifying N-linked glycopeptides from human serum. To overcome these challenges, we utilized pParse 2.0 to find monoisotopic peak of each precursor ion as well as co-eluted ones, then applied spectral library search to intact glycopeptide identification through pMatchGlyco. Spectra of deglycosylated peptide including semi-tryptic peptides and common modifications were used for spectral library construction. Through scoring of ion matches on MS/MS spectra and target-decoy false positive rate control, and manual check of co-eluted precursor ions at MS1 level, the accuracy of identification was improved. In total, this method identified 1,194 N-linked glycosites and 448 unique glycoproteins in human serum.
