Project description:To examine the changes of miRNA in the small extracellular vesicles of breast cancer cells receiving DOX chemotherapy. Human breast cancer cells (MDA-MB-231 cells) were cultured with exorcism-free serum, and treated with DOX. After 24h, cell supernatant was collected and small extracellular vesicles were extracted by ultra-fast centrifugation method, and miRNA content in small extracellular vesicles was obtained for high-throughput miRNA sequencing.

Proteomic analysis of vitreous in WT and CD11b KO mice.

Project description:Our data support that following optic nerve crush injury, ocular infiltration of immune cells causes damage to the retinal vasculature and compromises the integrity of the blood-retina barrier (BRB). Functional ablation of integrin alphaM (CD11b) attenuates immune cell trafficking, protects the BRB, and strongly enhances β-glucan-elicited retinal ganglion cells axon regeneration. Here, we performed an unbiased analysis of the vitreal proteome; we harvested vitreous humor from WT and Itgam-/- mice, β-glucan treated after optic nerve injury, and used mass spectrometry-based proteomics for differential analysis of the WT and Itgam-/- vitreous proteome (label free and tandem mass tagging).

Proteomic signatures of extracellular vesicles secreted by non-mineralizing and mineralizing human osteoblasts and stimulation of tumor cell growth

Project description:Beyond forming bone, osteoblasts play pivotal roles in various biological processes, including hematopoiesis and bone metastasis. Extracellular vesicles (EVs) have recently been implicated in intercellular communication via transfer of proteins and nucleic acids between cells. Here, we focused on the proteomic characterization of non-mineralizing (NMOBs) and mineralizing (MOBs) human osteoblast (SV-HFOs) EVs and investigated their effect on human prostate cancer (PC3) cells by microscopic, proteomic and gene expression analyses. Proteomic analysis showed that 97% of the proteins were shared among NMOB and MOB EVs, and 30% were novel osteoblast-specific EV proteins. Label-free quantification demonstrated mineralization stage-dependent five-fold enrichment of 59 and 451 EV proteins in NMOBs and MOBs, respectively. Interestingly, bioinformatic analyses of the osteoblast EV proteomes and EV-regulated prostate cancer gene expression profiles showed that they converged on pathways involved in cell survival and growth. This was verified by in vitro proliferation assays where osteoblast EV uptake led to two-fold increase in PC3 cell growth compared to cell-free culture medium-derived vesicle controls. Our findings elucidate the mineralization stage-specific protein content of osteoblast-secreted EVs, show a novel way by which osteoblasts communicate with prostate cancer, and open up innovative avenues for therapeutic intervention. Label-free quantitative proteomics analysis of bone marrow cells of MK-LCM WT versus KO mice

Project description:High ploidy large cytoplasmic megakaryocytes (LCM) are critical negative regulators of hematopoietic stem cells (HSC) and are responsible for platelet formation. Using a mouse knockout model with normal megakaryocyte numbers but essentially devoid of LCM (MK-LCM KO), we demonstrated a pronounced increase in bone marrow HSC concurrent with endogenous mobilization and extramedullary hematopoiesis. When HSC isolated from a MK-LCM KO microenvironment were transplanted in lethally irradiated mice, the absence of LCM increased HSC in BM, blood and spleen. Severe thrombocytopenia was observed in animals with diminished LCM, although there was no change in megakaryocyte ploidy distribution. In contrast, WT HSC-generated LCM regulated a normal HSC pool and prevented thrombocytopenia. The present label-free quantitative LC-MSMS data was used to determine proteins that are differentially expressed in bone marrow cells of MK-LCM WT versus MK-LCM KO mice.
