GPX4 methylation in HEK-293T cells

Project description:After transfecting HEK-293T cells with Flag-GPX4, the cells were immunoprecipitated using an anti-FLAG antibody. the GPX4 proteins were isolated using sodium dodecyl-sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) on a 4-12% gradient gel (Invitrogen, Carlsbad, CA, USA) and stained with Coomassie blue. Thylakoid protein phosphorylation in Physcomitrella patens

Project description:Physcomitrella patens is a model plant representing the group of mosses, which diverged from vascular plant ancestors early after land colonization. To investigate the unique characteristic of the composition and post-translational regulation of Physcomitrella patens photosynthetic apparatus, we have separated the thylakoids protein complexes in their subunits composition by means of two-dimensional large-pore Blue-Native/sodium dodecyl sulfate-polyacrylamide gel electrophoresis (2D lpBN/SDS-PAGE), followed by protein in-gel tryptic digestion of single spots. To identify the kinase responsible for phosphorylation of the major thylakoid protein complex photosystem II, a mutant line lacking the ortholog of kinase STN8 in the model angiosperm Arabidopsis thaliana has been generated and the identified thylakoid protein phosphorylation pattern have been compared with the WT moss.

Project description:methane yield was promoted during propionate anaerobic digestion process.The total proteins of three samples were extracted, and the concentration and quality of proteins were determined by BCA Protein Assay Kit and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), respectively. Next, protein was digested, and the peptides after protein digestion were vacuum dried, and then re-suspended with 2% acetonitrile and 0.1% trifluoroacetic acid (TFA). The samples were desalted with a Waters Oasis HLB and vacuum dried. The assay of samples was performed on a Q-Exactive HF-X coupled with Easy-nLC 1200.

Novel peptidomics-based evaluation of pork freshness via a potential peptide biomarker

Project description:This study applied peptidomics to investigate potential biomarkers for evaluating pork-meat freshness. Meat samples stored at -2, 4, 10, and 25 °C were collected at specific time points to evaluate meat freshness indicators (color, total viable count, pH, and total volatile basic nitrogen). The sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) profile was analyzed, and substantial protein degradation (myosin heavy chain, paramyosin, troponin) was detected at the end of storage, regardless of the temperature. Peptidomics analysis was performed using a UHPLC-LTQ-Orbitrap mass spectrometer, and the potential peptide marker MVHMASKE was filtered via multivariate analysis and quantified by parallel reaction monitoring combined with external standard quantitation. In addition, the relationship between peptide content and change in meat freshness was verified using real-life samples and the content of MVHMASKE showed an obvious decline during storage, presenting a period of pork meat from fresh to spoilage. This study provides favorable evidences to evaluate pork meat freshness by mass spectrometry-based pep-tidomics.

Gene expression profiling predicts processing properties in short-term imbibition of wheat

Project description:Objective: Germination of wheat maximizes phytochemical content and antioxidant activity while altering chemical composition, gluten content, and pasting properties. We previously reported marked changes in gene expression in soybean prior to germination before the radicle had grown from a seed. The present study investigated whether imibibition induces similar changes in wheat. Methods: Changes in gene expression profiles of wheat during short-term imbibition (0, 16, and 24 h) were evaluated by DNA microarray analysis. Gene Ontology (GO) analysis was carried out to categorize the function of genes with altered expression. The expression of genes encoding enzymes associated with starch breakdown was evaluated by quantitative real-time PCR, and changes in enzymatic activity were assessed with functional assays. Pasting properties of flour made from wheat seeds imbibed for different times were examined with a Rapid Visco Analyzer. The protein profile was determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis, and gluten content was quantified. Results: The GO analysis revealed that genes related to cellulose and cell wall synthesis were upregulated by imbibition for 16 h whereas those associated with polysaccharide catabolism and nucleosome assembly were upregulated in the subsequent 8 h. α-Amylase expression was highest after 24-h imbibition, with a corresponding increase in enzymatic activity. The pasting properties of wheat flour decreased when seeds were imbibed for over 16 h. Gluten was not degraded until 48 h imbibition. Conclusion: Short-term imbibition of wheat can produce a new type of starch with improved physical and functional properties that may be more appealing to consumers.
