Characterization of the SFPQ-forming meshwork condensate for multi-dimensional gene regulation and its implications in autism spectrum disorder

ABSTRACT: To comprehensively identify the entire gene expression regulatory complex associated with SFPQ in vivo, we conducted purification of biotinylated proteins within the LLPS structure and subsequently characterized them using mass spectrometry. HEK 293T cells expressing TurboID-SFPQ and cells expressing SFPQ lacking TurboID as a negative control were both labeled with 500 μM biotin for 10 minutes, and the cell lysates were subjected to pull-down using streptavidin beads for purification.
