Single-Cell Secretome Profiling by NanoLC-MS/MS Analysis


ABSTRACT: Here, we reported a novel single cell secretome profiling method that integrates cell metabolic labeling, on-capillary enrichment based on click chemistry, in-situ sample preparation, and nanoLC-MS/MS analysis. By this method, an average of circa 389 protein groups were confidently identified as the secretome from single HeLa cells, an order of magnitude higher than previously reported targeted secretome analysis by antibody recognition.
