Serum peptide barcode and peptidome profile of horses with Equine Gastric Ulcer Syndrome

ABSTRACT: Early and accurate diagnosis Equine Gastric Ulcer Syndrome (EGUS) is critical for effective treatment and management. This study aimed to analyse the serum peptide barcode and peptidome profile of horses with EGUS using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry and liquid chromatography-tandem mass spectrometry.

Gene expression profiling in equine polysaccharide storage myopathy

Project description:Standardized muscular biopsies of the dorsal compartment of the gluteus medius muscle were performed in 7 horses suffering from equine polysaccharide storage myopathy (PSSM) and 6 sound Norman Cob horses . Gene expression analysis was performed using an equine oligonucleotide microarray which included 384 equine gene probes of the nuclear genome and all the mitochondrial genes. All the samples of PSSM muscles were hybridized against the reference control muscles. This reference was made by pooling together all the mRNA extracted after in vitro transcription from the 6 control muscles of the sound horses. Briefly, the hybridization protocol was adapted from Le Brigand et al. (2006). An open-access long oligonucleotide microarray resource for analysis of the human and mouse transcriptomes. Distinct molecular profiles underpin mild to moderate equine asthma cytological profiles

Project description:This project used a state-of-the-art, multi-omics approach to improve our understanding of the aetiopathogenesis of a highly prevalent, performance-limiting disorder of racehorses; mild to moderate equine asthma (MMEA). This advanced knowledge is a prerequisite to improving prophylactic, management and therapeutic options for this condition. Although a number of risk factors for MMEA have been identified, incomplete understanding of the precise aetiopathogenesis of this syndrome means that options for intervention are currently limited. This study aimed to apply a multi-omic approach, reveal key inflammatory pathways involved in inflammatory cell recruitment to the lower airways and highlight pathways specific to different MMEA inflammatory cell profiles (MMEA endotypes). Thus, we compared BALF cell gene and protein expression data from horses with normal bronchoalveolar lavage fluid (BALF) cytology (n = 8) with those isolated from horses with neutrophilic (n = 4), mastocytic (n = 8), mixed neutrophilic/mastocytic (n=4) and mixed eosinophilic/mastocytic (n=3) inflammation. Analyses on transcriptomic and proteomic data derived from BALF cells from horses with neutrophilic cytology showed enrichment in classical inflammatory pathways, while horses with mastocytic cytology showed enrichment in pathways involved in hypersensitivity reactions related to non-classical inflammation that could mimic a Th2 immune response. The mixed eosinophilic/mastocytic group also presented with a non-classical inflammatory profile while, the mixed neutrophilic/mastocytic group revealed profiles consistent with both neutrophilic inflammation and hypersensitivity. Our adopted multi-omics approach (both gene and protein expression) has enabled a more holistic assessment of the immunological status of the lower airways associated with the different cytological profiles of equine asthma.

Project description:Equine Papillomavirus Type 2 (EcPV2) appears to be a causal factor for the development of genital especially penile squamous cell carcinomas (SCC) and as such have an important clinical impact on horses. However, the pathomechanisms associated with this cancer transformation are not known, yet. To analyze the host’s and viral transcriptome in EcPV2 affected horses, tissue samples were collected from horses with EcPV2-positive genital lesions as well as from healthy EcPV2-negative horses. Expression levels of host and viral genes were evaluated by RNA-Seq.

Project description:Through this comparative proteomic study of equine CD4+ cells 376 membrane proteins could be identified. Several proteins showed changed abundance in horses affected by a spontaneous autoimmune disease (equine recurrent uveitis, ERU). Findings provide novel knowledge about changes in the CD4+ immune cell membrane proteome in a spontaneously and naturally occurring autoimmune disease in horses. The data is highly relevant for veterinary medicine and has proven translational quality for autoimmune uveitis in man.

RNA-seq of equine and canine mammary stem cells treated with DMBA

Project description:Whereas canines are susceptible to mammary cancer at a frequency similar to humans, equines are largely resistant to the disease. We treated mammary stem cells from both species with the chemical carcinogen, DMBA, and found that equine cells are significantly more susceptible to treatment. These RNA-seq results represent samples from three individuals of dogs and horses in order to reveal transcriptional differences in response to treatment that may explain the phenotypic differences we observed.
