Proteomic Analysis of Mouse Liver in Response to a Choline-Deficient, L-Amino Acid-Defined, High-Fat Diet (CDAHFD)

ABSTRACT: We investigated proteomic changes in the livers of mice subjected to various dietary and genetic interventions. Three comparisons were made: (1) Mice fed CDAHFD (n = 6) vs. mice fed regular chow (RC) (n = 6); (2) Mice treated with Coasy ASO under CDAHFD conditions (n = 6) vs. mice treated with control ASO under CDAHFD conditions (n = 6); (3) Mice treated with Coasy ASO and 2% cholesterol under CDAHFD conditions (n = 6) vs. mice treated with Coasy ASO under CDAHFD conditions (n =6).

RNA Sequencing of livers from ApoE-/- mice treated with HIF1a inhibitor PX-478 or Saline

Project description:Purpose: HIF1a inhibitor PX-478 prevents hypercholesterolemia in ApoE-/- mice fed Western diet for 3 months. The goal of this study was to determine the mechanisms in the liver through which PX-478 regulates cholesterol. Methods: 4-week old ApoE-/- mice were fed a Western diet and injected intraperitoneally, bi-weekly with either PX-478 (40 mg/kg) or Saline for 3 months. The livers of 3 PX-478-treated mice and 4 Saline-treated mice were used for this RNA-seq study. Results: We identified 800+ differentially expressed genes in the PX-478-treated ApoE-/- mice livers relative to the saline-treated control livers. A subset of these genes were found to play important roles in cholesterol regulation and were further validated by qPCR analysis. Conclusions: Our study details potential hepatic cholesterol regulating mechanisms for HIF1a inhibitor PX-478. These results, coupled with our findings on the effect of PX-478 on two different mouse models of atherosclerosis, demonstrate that PX-478 is a potential anti-atherogenic and anti-hypercholesterolemic drug.

RNA-sequencing of liver tissue from wild type and Gramd1c knockout mice fed with high and low cholesterol diet

Project description:The Aster-C protein (encoded by the Gramd1c gene) is an endoplasmic reticulum (ER) resident protein that has been reported to transport cholesterol from the plasma membrane to the ER . Although there is a clear role for the closely-related Aster-B protein in cholesterol transport and downstream esterification in the adrenal gland, the specific role for Aster-C in tissue cholesterol homeostasis is not well understood. Here, we have examined whole body cholesterol balance in mice globally lacking Aster-C under low or high dietary cholesterol conditions.transport and metabolism under divergent dietary cholesterol conditions. These results strongly suggest that Aster-C alone is not sufficient to control whole body cholesterol balance, but can modestly impact circulating cortisol and bile acid levels when dietary cholesterol is limited.

Expression profiling of aging liver in low-fat diet mice

Project description:Dietary interventions are effective ways to extend or shorten lifespan. By examining midlife hepatic gene expressions in mice under different dietary conditions, which resulted in different lifespans and aging-related phenotypes, we were able to identify genes and pathways that modulate the aging process. To determine how our dietary intervention-based transcriptomic approach for predicting aging-regulatory genes compares to more traditional approach of using age-dependent transcriptional changes, we examined the hepatic gene expression changes in LF-fed mice during aging at 4, 8, 13 and 21 months. Male C57BL/6J mice at 4 weeks of age were purchased from Shanghai Animal Co, Ltd. Mice were maintained under a 12-hour dark/light cycle (lights on at 6:30 am) at a temperature of 22 ± 3 °C in accredited animal facilities. Prior to the start of experiment, mice were maintained on a low-fat diet (Research Diets Inc., New Brunswick, NJ) for one week. Liver RNAs extracted from six male C57BL/6J mice of the same age were pooled at each age point to obtain an average microarray profile at 4, 8, 13 and 21 months.
