ABSTRACT: The activated PBMCs were then divided into four subgroups: PBMC alone, PBMC co-cultured with PMN after 4 h (PBMC-PMN), PBMC co-cultured Mtb (PBMC-TB), and PBMC co-cultured Mtb then added PMN after 4 h (PBMC-TB-PMN). After a total of 6 h incubation, the protein of culture supernatant was collected and measured in concentration. The protein samples were separated by SDS-PAGE and underwent in-gel digestion. Extracted protein was injected into LC-MS/MS instrument for measurement

Project description:We performed an in vitro co-culture of allogeneic PBMCs and SC-islet clusters. SC-islet clusters were enriched for β cells (using CD49A magnetic sorting; SC-α still remain at lower numbers) (Veres et al., 2019), dissociated and reaggregated to obtain a more uniform cell count between wells. SC-islets were co-cultured with human allogeneic PBMCs for 24 or 48 hours. As controls [time(t)=0], SC-islets remained in culture without PBMC addition. These samples, in addition to PBMCs alone (t=0), were used for scRNA-seq (Figure 2A).

The effect of simulated night shift work on the circadian regulation of the human transcriptome

Project description:Eight healthy human subjects were enrolled in a 6-day simulated shift work protocol. Blood samples were collected during the two 24-hour measurement periods. Blood samples were collected every 4 hours during both measurement periods. Subjects entered the lab on Day 1. At the start of Day 2, the first 24-hour measurement period was started. Subjects slept according to their habitual sleep/wake schedule, followed by a 16-hour constant posture procedure. On days 3-6, the sleep period was delayed by 10 hours. Following the third night on this schedule, subjects underwent another 24-hour measurement period. During both measurement periods, 7 blood samples were collected and PBMCs were isolated. mRNA was extracted, labelled, and hybridized to microarrays.

Analysis of gene expression in co-culture of PBMCs with lung cancer cell lines by High-Throughput Sequencing

Project description:Objective: The goal of this study is to investigate paracrine effect of Peripheral Blood Mononuclear Cells (PBMCs) on lung cancer cell lines. Method: Transcription profiles of PBMCs (control) and PBMCs co-cultured with lung cancer cell lines (treatment) were generated by deep sequencing,in triplicate, using Illumina HiSeq 4000. Results and Conclusions: With demonstrating of RNA sequencing approach, PBMCs has been shown the gene expression changed when co-cultured with lung cancer cell lines. The result revealed that up-regulated genes were 8,073 and 8,070 genes co-cultured with A549 and H292, respectively. In the other hand, down-regulated genes were 8,341 and 8,342 genes co-cultured with A549 and H292, respectively. Comparing the data to expression array databases from lung cancer patients, up-regulated genes were shown 29 genes co-cultured with A549 and 37 genes with H292 which, among them, there were 2 genes, ANKRD37 and BUB3, found in both treatments. Down-regulated genes were found 24 and 35 genes in the co-culture of A549 an d H292, respectively, that no similar genes found in both treatments.
