LC-MS/MS analysis #1 of pull down of flag antibody against 35S::proBRIpep/35S::HAE:FLAG plants

ABSTRACT: Under the background of 35S::proBRIpep transgenic plants, 35S::BRI1:FLAG, 35S::CEPR:FLAG, and 35S::HAE:FLAG were introduced. After Co-immunoprecipitation by FLAG antibody, LC-MS/MS analysis examined the enrichment of BRIpep.

Pseudomonas aeruginosa PA3904 pull down coupled to LC-MS/MS

Project description:pETDUET His-PAAR4 and pETDUET Flag-PAAR4 were transformed into BL21 E. coli DE3 and expressed as described above. An equal volume of PAO1 retS was grown to an OD600 of 2.0. E. coli expressing His-PAAR4 or Flag-PAAR4 and Pseudomonas cells were harvested by centrifugation and lysed as described above. Lysates of both species were combined and His-PAAR4 or Flag-PAAR4 were purified as before. Eluted sample was mixed with SDS-loading buffer, boiled and run on a 12% SDS-PAGE for 0.6 cm. After coomassie staining and de-staining, the resulting band was excised and used for LC-MS/MS.

Project description:Col-0 transgenic plants expressing MSH1P::FLAG::RPL18 were generated in both msh1 homozygous mutant (SAIL-877-F01) and wild type backgrounds with cloned MSH1 sequence described in Xu et al. (2011) and with transformation by the floral dip method (Clough and Bent, 1998). Positive transgenic lines were screened for FLAG tag by immunoblotting methods. Polysomal mRNA from MSH1-specific cells was obtained by ribosome purification using the TRAP method essentially as reported by (Reynoso et al., 2015) from floral stem tissues.
