Diagnosing and staging epithelial ovarian cancer by serum glycoproteomic profiling

Project description:Background There is a need for diagnostic tests for screening, triaging and staging of epithelial ovarian cancer (EOC). Glycoproteomics of blood samples has shown promise for biomarker discovery. Methods We applied glycoproteomics to serum of people with EOC or benign pelvic masses and healthy controls. A total of 653 analytes were quantified and assessed in multivariable models, which were tested in an independent cohort. Additionally, we analyzed glycosylation patterns in serum markers and in tissues. Results We identified a biomarker panel that distinguished benign lesions from EOC with sensitivity and specificity of 83.5% and 90.1% in the training set, and of 86.7 and 86.7% in the test set, respectively. ROC analysis demonstrated strong performance across a range of cutoffs. Fucosylated multi-antennary glycopeptide markers were higher in late-stage than in early-stage EOC. A comparable pattern was found in late-stage EOC tissues. Conclusions Blood glycopeptide biomarkers have the potential to distinguish benign from malignant pelvic masses, and early- from late-stage EOC. Glycosylation of circulating and tumor tissue proteins may be related. This study supports the hypothesis that blood glycoproteomic profiling can be used for EOC diagnosis and staging and it warrants further clinical evaluation.

Acute and chronic liver injury change the small RNA cargoes carried by serum small extracellular vesicles in mice

Project description:Compared to whole serum miRNAs, miRNAs in serum small extracellular vesicles (sEVs) are well protected form RNA enzymes, thus provide a consistent source of miRNA for disease biomarker detection. Serum sEVs and their miRNA cargos released by injured liver cells could be promising biomarkers for diagnosis of liver diseases. We were very interested to find out the effects of liver injury on serum extracellular vesicles as well as the small RNA components they transported, if there is any difference between acute and chronic injury. Study in this regard will help us to identify new serum biomarkers for liver injury, and to find out if there are specific markers for acute or chronic liver injury. To identify potential biomarker for liver injury based on serum sEVs miRNAs, we established the carbon tetrachloride (CCL4) induced acute and chronic liver injury mice model, and examined the dynamic changes of small RNA components, especially miRNAs, in serum sEVs.

Identification of novel serum biomarkers in early-stage HCC patients.

Project description:Hepatocellular carcinoma (HCC) is one of the most common malignancies worldwide, and is the one of the few cancers in which a continued increase in incidence has been observed over several years. HCC associated with chronic liver disease evolves from precancerous lesion and early HCC to overt cancer, and identifying key molecules contributing to early stage HCC is an urgent need. α-Fetoprotein (AFP) is the best serum biomarker for diagnosis of HCC, but sensitivity is low, particularly in detection of early-stage HCC. Therefore, novel and reliable diagnostic biomarkers to complement AFP are needed to improve HCC diagnosis. We aim to determine transcriptome-based molecular signature of multistep hepatocarcinogenesis, and to identify novel serum biomarkers to diagnose early stage HCC patient.

Project description:In the present study, we employed a proteomics approach to identify parasite-derived proteins in the serum EVs from E. multilocularis-infected mice and assessed their diagnostic values. The study paves the way for the use of EVs to identify of diagnostic candidates and potentiates the applications of TER ATPase and TPx-1 in the early diagnosis and prognostic evaluation of echinococcosis.

Proteomic profiling of serum extracellular vesicles identifies diagnostic markers for echinococcosis.

Project description:Echinococcosis is a parasitic disease caused by the metacestodes of Echinococcus spp. The disease has a long latent period and is largely underdiagnosed, partially because of the lack of effective early diagnostic approaches. Using liquid chromatography-mass spectrometry, we profiled the serum-derived extracellular vesicles (EVs) of E. multilocularis-infected mice and identified three parasite-origin proteins, thioredoxin peroxidase 1 (TPx-1), transitional endoplasmic reticulum ATPase (TER ATPase), and 14-3-3, being continuously released by the parasites into the sera during the infection via EVs. Using ELISA, both TPx-1 and TER ATPase were shown to have a good performance in diagnosis of experimental murine echinococcosis as early as 10 days post infection and of human echinococcosis compared with that of control. Moreover, TER ATPase and TPx-1 were further demonstrated to be suitable for evaluation of the prognosis of patients with treatment. The present study discovers the potential of TER ATPase and TPx-1 as promising diagnostic candidates for echinococcosis.

A multi-omics approach identifies pancreatic cancer cell extracellular vesicles as mediators of the unfolded protein response in normal pancreatic epithelial cells

Project description:The early molecular mechanisms of pancreas cancer development remain elusive. A key driver of poor patient outcomes remains late-stage diagnosis of disease, typically when patients present with pancreatic ductal adenocarcinoma (PDAC). Currently, no diagnostics exist to identify indolent disease. It is crucial to identify the early molecular events that drive the transformation of naïve cells to a neoplastic phenotype. Extracellular vesicles (EVs) are nanometer sized lipid-bilayer particles released from cells of almost all known tissue types. EVs carry an array of biomolecules which are internalized by neighboring and distant cells, critical for facilitating intracellular communication. Although cancer derived EVs impact a myriad of cancer progression events including tumor proliferation, immune escape, and metastasis, their role in mediating early onset of PDAC is unknown. In this study, we report on alterations in the gene expression profiles of normal pancreatic epithelial cells upon internalization of PDAC cell derived EVs (cEVs). Leveraging a layered multi-omics approach, we show that cEVs induce endoplasmic reticulum (ER) stress and the unfolded protein response in recipient normal pancreatic epithelial cells within 24 hours. Consequently, cEV treated cells demonstrated increased proliferation. cEV cargo was enriched for an array of biomolecules which can induce or regulate ER stress and the UPR, including lipid species esterified to palmitic acid, sphingomyelins, metabolites which regulate tRNA charging and protein trafficking and degradation proteins. These results highlight a role for pancreatic cancer cEVs to induce stress in recipient normal pancreas cells; studying these genetic, metabolic and proteomic perturbations may lead to valuable insight to disease etiology and early biomarker identification.
