Notch inhibitor, DAPT, promotes the generation of porcine iPSCs-derived intestinal organoids

ABSTRACT: Three-dimensional (3D) intestinal organoids in pigs have been considered crucial in translational research and drug discovery. This study established a differentiation medium recipe of porcine-induced pluripotent stem cells (piPSCs) to form intestinal organoids.

Quantitative mass spectrometry analysis of intestinal organoids

Project description:Organoids have the potential to bridge 3D cell culture to tissue physiology by providing a model resembling in vivo organs. We propose here a SILAC method to measure protein expression changes in intestinal organoids under different experimental conditions. With the combined use of quantitative mass spectrometry, SILAC and organoid culture, we validated the approach and showed that large-scale proteome variations can be measured in an “organ-like” system under variable conditions.

RNA-seq of small intestinal organoids treated with an inhibitor for the epigenetic modifier LSD1

Project description:To assess the role of LSD1 in mice small intestinal epithelium, small intestinal organoids were treated with an inhibitor for LSD1 (GSK-LSD1) and compared to untreated organoids. Similar to intestinal epithelium from mice with an intestinal epithelium specific LSD1-KO, paneth cells dissappear upon GSK-LSD1 treatment. We used these sequencing data to show that these small intestinal organoids have a similar phenotype as mice epithelium without LSD1.

Epithelium-only cultured stem cells isolated from human pluripotent stem cell derived intestinal organoids grown in matrigel and alginate

Project description:Human intestinal organoids were grown in a typical 3D matrigel culture environment, or in an alginate gel, then a subset from each condition were xenotransplanted to vascularized and mature in vivo. Epithelium was isolated and epithelial only organoids (enteroids) were then grown from each condition prior to sequencing bulk RNA-sequencing.

Transcriptomics analysis of different porcine intestinal models (piglet jejunums, jejunum crypts, and organoids) and conventional cell line models upon infection with the transmissible gastroenteritis virus (TGEV)

Project description:The objective is to study the relevance of porcine intestinal models (incl. piglet jejunums, 2D and 3D organoids derived from piglet jejunums and swine testicular cells) to study intestinal functions and to decipher the host-virus interactions during viral porcine coronavirus infection. This dataset includes RNA-seq profiling of 54 samples covering the four experimental models mentioned above, uninfected or infected with the Purdue P115 strain of the transmissible gastroenteritis virus (TGEV). Details about the experimental models: - For the in vivo model, jejunums of five week old piglets were extracted. - For the organoid model, first jejunum crypts of newborn piglets were extracted and profiled. Then, 3D organoids derived from these crypts were profiled after 5, 14 and 25 passages. Last, 2D organoids derived from 3D organoids at passage 25 were also included. - For the cellular model, we relied on swine testicular cells (ST cells) at passage 53. Samples were either mock-infected or infected with TGEV (Purdue P115 strain) at MOI 0.06 (ST cells), 0.06 and 1 (2D organoids), and 10^4.75 TCID50 (piglets). For ST cells and organoids, the total RNA was recovered at 9hpi (hours post-infection) and 24hpi. For piglets jejunums, only the latest time point was included (24hpi). For each model and each condition (control / infected), three replicates were performed.

Murine intestinal organoids resemble intestinal epithelium in their microRNA profiles

Intestinal organoids largely resembled intestinal epithelial cells in their miRNA profiles.
