Design, synthesis, and biological evaluation of pyridopyrimidine derivatives in gastric cancer cells.

ABSTRACT: This study reports the rational design, synthesis, and in vitro biological evaluation of a novel series of pyridopyrimidine derivatives as potential anticancer agents targeting gastric cancer. The compounds were screened against the AGS gastric adenocarcinoma cell line To investigate the underlying mechanisms of action, a comprehensive label-free quantitative proteomics approach was employed. Differential protein expression analysis revealed key pathways modulated by the lead compound.

To uncover the mode of action of representative compound, label-free quantitative proteomics was performed, revealing key alterations in proteins linked to various spotted pathways as indicated in IPA analysis.

Investigating the multi-mechanistic anticancer effects of 4-bisarylurea thiouracil derivatives in breast cancer cells

Project description:The study reports the synthesis of a series of 4-bisarylureathiouracil derivatives (6a-e) for potential use in breast cancer treatment. In vitro cytotoxicity was assessed in MCF-7 and MDA-MB-231 human breast cancer cell lines, revealing significant anti-cancer activity. Compound 6e exhibited the highest cytotoxicity, with IC50 values of 7.94 μM for MCF-7 and 6.67 μM for MDA-MB-231, although it was also the most toxic to RAW 264.7 macrophage cells. In contrast, compound 6c demonstrated strong efficacy against both cancer cell lines (IC50 = 9.23 ± 0.6 μM for MCF-7 and 7.72 ± 0.6 μM for MDA-MB-231) while maintaining selectivity (SI values >10.8 and > 12.9, respectively). Flow cytometry and caspase-3 assays indicated that compounds 6a-c induced apoptosis in MCF-7 cells. In anti-inflammatory assays, compounds 6a and 6d showed significant effects, while 6c demonstrated the weakest, suggesting its cytotoxicity is not linked to anti-inflammatory properties. Compound 6c was prioritised for further investigation because of its preferential targeting of cancer cells. Proteomic analysis of 6c-treated cells revealed significant dysregulation of apoptosis, angiogenesis and VEGF signalling, Rho signal transduction, and pi3k-akt signalling pathways. These findings highlighted the potential of compounds 6a-e as effective anticancer agents, warranting further investigation and optimization for therapeutic applications.

Design, synthesis, and bio-evaluation of 9-(pyrimidin-2-yl)-9H-carbazole derivatives that disrupt mitochondrial homeostasis in human lung adenocarcinoma

Project description:Purpose:To further explore the possible molecular mechanism by which compound 5n induces A549 cell apoptosis, we analyzed global transcriptome alterations of A549 cells at certain time points (48 h) after compound 5n treatment using RNA-seq analysis. Methods: Differential expression analysis for mRNA was performed using R package edgeR. Differentially expressed RNAs with |log2(FC)| value >1 and q value <0.05, considered as significantly modulated, were retained for further analysis. This choice is motivated by the decision to maximize the sensitivity of this analysis, in order to perform a massive screening and identify candidate genes to be validated with a wider sample population with real-time PCR analysis Results: The quantity of gene expression was calculated by FPKM (Fragments Per Kilobase of transcript per Million fragments mapped). Genes with log2 (Fold change) >1 and P value < 0.05 were considered as differentially expressed genes (DEGs). There were 146 upregulated genes and 158 downregulated genes for a total of 304 DEGs that were results are shown in Fig. 8A. Furthermore, the enriched KEGG pathways were analyzed and the top 30 pathways involved in these 304 DEGs between the two groups are shown in Figure SI. Among the top 30 pathways, 9 pathways related to mitochondrial function had significant effects. 37 DEGs in KEGG pathway associated with mitochondrial function was conducted and analyzed. Conclusions: Metabolism-related pathways was affected by compound 5n according the RNA-seq analysis

Characterization of novel a-Mangostin and Paeonol derivatives with cancer-selective cytotoxicity

Project description:a-Mangostin and Paeonol have shown anti-cancer and anti-inflammatory properties, however these two natural compounds have no clinical value because of their low solubility and low membrane permeability. In this study, we screened chemically synthesized derivatives from these two natural compounds as potential novel chemicals that increase cancer cell cytotoxicity over non-transformed human cells. We found the two derivative compounds, named a-Mangostin-1 (aMan1) and Paeonol-1 (Pae1) more efficiently and specifically induced cytotoxicity in HCT116, HT-29, and SW48 colorectal cancer cell lines than the parental compounds. Both, aMan1 and Pae1 arrested HCT116 cells in G1 and HT-29 and SW48 cells in G2/M phase of the cell cycle. Both aMan1 and Pae1 induced apoptosis in human cancer cells, through a Caspase-dependent mechanism. aMan1 and Pae1 induced selective transcriptional responses in CRC cells involving genes related to metabolic stress and DNA damage response signaling pathways. Finally, experiments on primary colon organoids showed that both the novel derivatives kill organoids derived from cancer without affecting the viability of organoids derived from healthy tissue, where the parental compounds and the currently-used chemotherapeutic drug Irinotecan failed in. In conclusion, our findings increase the knowledge about natural compound derivatives as anticancer compounds and open new research options on the derivation of lead compounds aimed to the development of novel CRC chemotherapeutic drugs that selectively target cancer, but not healthy cells.

PCW-1001, a novel pyrazole compound, modulates the expression of DNA damage response genes

Project description:As pyrazole and its derivatives have a wide range of biological activities, including anticancer activity, the design of novel pyrazole derivatives has emerged as an important research field. This study describes a novel pyrazole derivative that exerts antitumor and radiosensitizing activities in breast cancer both in vitro and in vivo. We synthesized a novel pyrazole compound N,N-dimethyl-N'-(3-(1-(4-(trifluoromethyl)phenyl)-1H-pyrazol-4-yl)phenyl)azanesulfonamide (PCW-1001) and showed that it inhibited several oncogenic properties of breast cancer both in vitro and in vivo. PCW-1001 induced apoptosis in several breast cancer cell lines. Transcriptome analysis of PCW-1001-treated cells showed that it regulated the DNA damage response, suggesting its potential use in radiotherapy. Indeed, PCW-1001 enhanced the radiation sensitivity of breast cancer cells by modulating the expression of DNA damage response genes. Therefore, our data describe a novel pyrazole compound, PCW-1001, with antitumor and radiosensitizer activities in breast cancer.

In silico approach and mass spectrometry-based proteome profiling unravel IGF-1R inhibition potential of 3-(2-furoyl)-indole scaffolds in hepatocellular carcinoma

Project description:Considering the multi-targeting potential of furan-indole hybrids as anticancer agents, this study aimed to unveil the mechanism of new 3-(2-furoyl)-indole derivatives for their cytotoxicy on hepatocellular carcinoma cells. Initially, we conducted an in vitro cytotoxicity assay for thirteen 3-(2-furoyl)-indole derivatives on HepG2 and MCF-7 cells using MTT assay, correlating the results with in silico screening of the selected drug targets. Among them, compounds 4a, 4b, and 4c demonstrated better selectivity towards HepG2 cells than MCF-7 cells. This was in good agreement with docking studies, which showed the high binding affinity of compound 4a to the Chain A of IGF-1R at Ile1160, Glu 1080, Met 1082 and Asp-1086, similar to reference ligand. Molecular dynamic study inferred the stable and high binding affinity of compound 4a to IGF-1R. The RMSD and RMSF values were found to be acceptable, and accordingly, a pharmacophore model was constructed. Hence, we conducted label-free quantitative proteome profiling of HepG2 cells treated with active compounds using an in-solution trypsin digestion procedure followed by high-resolution mass spectrometric analysis. We noticed that compound 4a targets IGF-1R pathway by dysregulation of proteins such as SHC-transforming protein 1(P29353), Mitogen-activated protein kinase kinase 3 isoform B variant (Q53EZ9), dual specificity mitogen-activated protein kinase kinase 4 (P45985), stress-activated protein kinase JNK (A0A286YF97), and mitogen-activated protein kinase (E9PQW4). In addition, the apoptotic activities of 4a, 4b, and 4c were studied against HepG2 cells using FACS. These findings are in good agreement with our in silico results, proteome profiling, and also earlier reports on indole compounds. Furthermore, we predicted the in silico ADMET property of the compounds.

Characterization of novel natural compound derivatives with cancer-selective cytotoxicity (RNA-seq dataset 3)

Project description:Colorectal cancer is one of the most frequent and lethal cancer in the world. Current therapeutic approaches are still not fully successful, yet cause numerous and severe side effects for the patient. We screened chemically synthesized derivatives from two natural compounds (a-Mangostin and Paeonol) as potential novel chemicals that increase cancer cell selectivity over non-transformed human cells. We found two of them that efficiently induced cell cycle arrest and apoptosis in human cancer cells and organoids derived from tumor tissue without affecting the viability of human non-cancer cells and intestinal organoids derived from healthy tissue. Our findings increase the knowledge about cancer-specific compounds and propose two natural compound derivatives as potential novel chemotherapeutics.
