Project description:Purpose: To assess global changes of wheat coleoptiles gene expression in response to fusaoctaxin B which is a linear, C-terminally-reduced and D-amino acid residue-rich octapeptide, as the product of the two non-ribosomal peptide synthetases encoded by Fusarium graminearum fg3_54 cluster. Methods: For RNA seq of wheat coleoptiles, three biological replicates of after 3 hours fusaoctaxin B-treated coleoptiles, after 3 hours control (0.5% Tween-20) treated coleoptiles, after 24 hours fusaoctaxin B-treated coleoptiles and after 24 hours control (0.5% Tween-20) treated coleoptiles were sequenced. Results: For each sample, 33-95 M RNA-seq clean reads were obtained mapped to the Triticum aestivum Chinesespring genome sequence. Conclusions: Fusaoctaxin B represents an unprecedented virulence factor.

Project description:The analysis of gene expression during wheat development: Gene expression measurements were carried out on a developmental tissue series for wild-type wheat (cv. Chinese Spring) using the Affymetrix Wheat GeneChip. Thirteen tissues at defined developmental stages were chosen to match the barley (cv. Morex) tissue series of Druka et al. 2006 that used the Affymetrix Barley1 GeneChip. Three replicates of: root tissue at two different developmental stages, leaf, crown, caryopsis, anther, pistil, inflorescence, bracts, mesocotyl, endosperm, embryo and coleoptiles were hybridised. Comparisons between this wheat data and the barley dataset were performed and are available at http://contigcomp.acpfg.com.au [PLEXdb(http://www.plexdb.org) has submitted this series at GEO on behalf of the original contributor, Tim Sutton. Project description:Waxy starch has an important influence on the qualities of breads. Generally, grain weight and yield in waxy wheat (Triticum aestivum L.) are significantly lower than in bread wheat. In this study, we performed the first proteomic and phosphoproteomic analyses of starch granule-binding proteins by comparing the waxy wheat cultivar Shannong 119 and the bread wheat cultivar Nongda 5181. These results indicate that reduced amylose content does not affect amylopectin synthesis, but it causes significant reduction of total starch biosynthesis, grain size, weight and grain yield. Two-dimensional differential in-gel electrophoresis identified 40 differentially expressed protein (DEP) spots in waxy and non-waxy wheats, which belonged mainly to starch synthase (SS) I, SS IIa and granule-bound SS I. Most DEPs involved in amylopectin synthesis showed a similar expression pattern during grain development, suggesting relatively independent amylose and amylopectin synthesis pathways. Phosphoproteome analysis of starch granule-binding proteins, using TiO2 microcolumns and LC-MS/MS, showed that the total number of phosphoproteins and their phosphorylation levels in ND5181 were significantly higher than in SN119, but proteins controlling amylopectin synthesis had similar phosphorylation levels. Our results revealed the lack of amylose did not affect the expression and phosphorylation of the starch granule-binding proteins involved in amylopectin biosynthesis.

Project description:Drought is one of the most serious abiotic stresses, under which the crop yield is significantly reduced. Elite winter wheat cultivar Henong 341 (Triticum aestivum L.) was grown in experimental fields of China Hebei province (116°37′23″E, 37°41′02″N) Two different treatment conditions were designed: rain-fed (no water irrigation) and well-watered (water irrigation) conditions at jointing and flowering stages. Each treatment included three biological replicates and each plot was 25m2. In this study, a comprehensive phosphoproteomic analysis of developing grains of 26th day after flowering (DAF) in Chinese bread wheat cultivar Henong 341 under well-watered and water-deficit conditions was performed by means of TiO2 enrichment, LC/MS/MS analysis, and Label-free peptide-intensity quantification. Under well-watered conditions, a total of 590 unique phosphopeptides corresponding to 603 nonredundant phosphorylation sites, representing 471 phosphoproteins, were identified. While under water deficit, 63 unique phosphopeptides, corresponding to 61 phosphoproteins, were found to be differently regulated in phosphorylation status compared to well-watered conditions (≥2 fold intensities).
