Project description:We performed a label-free quantitative mass spectrometry-based proteomic analysis comparing the dry and imbibed seed proteome and phosphoproteome of two genotypes with contrasted level of dormancy in three independent biological replicates. This assay aimed to identified protein accumulation pattern and change in phosphopeptide abundances correlating with germination phenotypes.

Project description:Drought is one of the most serious abiotic stresses, under which the crop yield is significantly reduced. Elite winter wheat cultivar Henong 341 (Triticum aestivum L.) was grown in experimental fields of China Hebei province (116°37′23″E, 37°41′02″N) Two different treatment conditions were designed: rain-fed (no water irrigation) and well-watered (water irrigation) conditions at jointing and flowering stages. Each treatment included three biological replicates and each plot was 25m2. In this study, a comprehensive phosphoproteomic analysis of developing grains of 26th day after flowering (DAF) in Chinese bread wheat cultivar Henong 341 under well-watered and water-deficit conditions was performed by means of TiO2 enrichment, LC/MS/MS analysis, and Label-free peptide-intensity quantification. Under well-watered conditions, a total of 590 unique phosphopeptides corresponding to 603 nonredundant phosphorylation sites, representing 471 phosphoproteins, were identified. While under water deficit, 63 unique phosphopeptides, corresponding to 61 phosphoproteins, were found to be differently regulated in phosphorylation status compared to well-watered conditions (≥2 fold intensities).

Dynamic coding and noncoding RNA interactions coordinate gene expression during meiosis in wheat

Project description:We present a high-resolution network of gene expression during meiosis and its regulatory control elements in polyploid wheat. MeioCapture method (Shunmugam et al. 2018 BMC Plant Biology 18: 293) was used for isolating high-purity sporogenous archesporial columns of synchronized male meiocytes from wheat anthers in different meiotic stages. Total RNA extracted from three independent biological replicates per developmental stage of the meiocytes, as well as vegetative flag leaf, young leaves, and reproductive anthers and pollen, was sequenced using Illumina platform, generating 548 million paired-end short mRNA reads. Filtered reads were aligned to the wheat reference genome (IWGSC v1.0) using STAR. Transcript abundance was estimated using the RSEM (v1.3.3) and the IWGSC v1.1 annotation, and read counts and transcripts per million (TPM) values that normalize counts to a consistent number per library (1 million) to facilitate the comparison of relative expression across various samples were generated.

Project description:We used two wheat genotypes, the susceptible wheat cultivar ‘8866 ’(S) and its near isogenic line with single powdery mildew resistance gene ‘pm30’ (R), to investigate gene expression changes in response to powdery mildew infection by using Wheat Genome Array wheat young leveas of near isogenic lines before or 12 hours after powdery mildew infection were selected for RNA extraction and hybridization on Affymetrix microarrays.The leaf samples were harvested from three independent biological replicates, and the leaves without inoculation were regarded as control.
