Organ-Specific Proteomic Responses to Tetrodotoxin Administration in the Liver and Skin of Takifugu rubripes

ABSTRACT: TTX was administered intramuscularly to 8-month-old tiger pufferfish, and their livers　and skin were collected 24 hours later. This is the mass spectrum and result obtained during proteomic analysis of those livers and skins. liver OE_250220_FH_YS1_1~OE_250220_FH_YS1_5: Control group OE_250220_FH_YS1_6~OE_250220_FH_YS1_11: TTX administration group A total of 11 raw files were generated from Takifugu rubripes liver samples. To balance the sample size between groups (n=5 for both control and exposure), one sample (OE_250220_FH_YS1_7, TTX2) was excluded from the final statistical analysis. Characterization of hyperglycemia due to sub-chronic administration of red ginseng extract via comparative global proteomic analysis

Project description:Ginseng (Panax ginseng Meyer) is commonly used as an herbal remedy worldwide. Few studies have explored the possible physiological changes in the liver although patients often self-medicate with ginseng preparations, which may lead to exceeding the recommended dose for long-term administration. Here, we analyzed changes in the hepatic proteins of mouse livers using quantitative proteomics after sub-chronic administration of Korean red ginseng (KRG) extract (control group and 0.5, 1.0, and 2.0 g/kg KRG) using tandem mass tag (TMT) 6‐plex technology. The 1.0 and 2.0 g/kg KRG groups exhibited signs of liver injury, including increased levels of aspartate transaminase (AST) and alanine aminotransferase (ALT) in the serum. Furthermore, serum glucose levels were significantly higher following KRG administration compared with the control group. Based on the upregulated proteins found in the proteomic analysis, we found that increased cystathionin beta-synthase (CBS) and cystathionine gamma-lyase (CSE) levels promoted greater hydrogen sulfide (HRR2RRS) synthesis in the liver. This investigation provides novel evidence that14T sub-chronic administration of 4T14TKRG can elevate HRR2RRS production by increasing protein expression of CBS and CSE in the liver.

Expression data from adult male Sprague Dawley rat liver, investigating how pomegranate skin extract prevents non alcoholic fatty liver diseases

Project description:Pomegranate skin extract could prevent fatty liver due to high fat diet in adult male Sprague Dawley rat. There are 3 groups of rats, feeding chow diet, high fat diet and high fat diet combined with pomegrante skin extract. After 8 weeks feeding, high fat diet group developped fatty liver but the other two groups still have healthy liver. We used microarrays to detail the global programme of gene expression underlying the fatty liver development and preventive effect of pomegranate skin extract on fatty liver. There are 3 treatment groups and 6 replicates for each group. The 3 treatment groups are control group feeding chow diet, high fat group feeding high fat diet and PE group feeding high fat diet and pomegranate skin extract. Rat livers were collecteded for RNA extraction and hybridization on Affymetrix microarrays.

Gene expression profile in the liver of insulin-deficient mice with icv leptin administration

Project description:Analysis of gene expression in the liver of insulin-deficient mice regulated by icv leptin administration. Control group received icv PBS administration. Icv leptin administration ameliorates hyperglycemia in insulin-deficient mice. Our transcriptome data provides important aspects of the leptin’s anti-type 1 diabetes action. Mice were harvested 10 days after icv leptin administration (25ng/0.11uL/hour). Liver samples were quickly removed, frozen in liquid nitrogen and subsequently stored at –80ºC. RNAs were extracted by Qiagen mRNA extract kits (RNeasy plus). gDNA was eliminated in this procedure by gDNA eliminator column, which is included in Qiagen mRNA extract kits. Genomics and Microarray Core Facility at UT-Southwestern (http://microarray.swmed.edu/) checked RNA quality with Bioanalyzer Chip and processed the samples for hybridization with Illumina Mouse-6 V2 BeadChip (Illumina Inc., San Diego, CA).

Aging effects on DNA methylation modules in blood tissue

Project description:Genome wide DNA methylation profiling of whole blood in schizophrenia patients and healthy subjects. The Illumina Infinium 27k Human DNA methylation Beadchip v1.2 was used to obtain DNA methylation profiles across approximately 27,000 CpGs. Raw data included 325 schizophrenia patients (2) and 394 controls (1) from Dutch descent. One sample was excluded since it was a bipolar patient, indicated by 3 in diseaseStatus column. The column used_in_analysis shows the samples that were used in our study. The excluded samples failed quality control. Normalized data are also included in the non_normalized.txt (i.e. AVG_Beta column).

Comparing different administration routes for mesenchymal stromal cell infusion in an in vivo porcine ischemia-reperfusion injury model

Project description:Hepatic ischemia-reperfusion injury is a complex pathophysiological process that occurs following reperfusion of blood supply in previously ischemic tissues. This form of injury plays a major role in cell dysfunction and death following hepatic surgery and liver transplantation. Mesenchymal stromal cells (MSC) seem to be a promising option for mitigation of this type of injury, as they possess anti-inflammatory and immune modulating capacities that may stimulate liver regeneration. The aim of this study was to demonstrate safety, feasibility and effectiveness of direct hepatic administration of MSC for treatment of ischemia-reperfusion injury in a pig model. After total vascular exclusion of the liver for 45 minutes, 3x10^6 human bone-marrow derived (BM)-MSC/kg were directly administrated into livers either via the intraportal route or hepatic artery route, whereas the control group did not receive any BM-MSC. Following reperfusion, hepatic blood flow and pulmonary pressures were continuously measured. Samples of blood and tissue of spleen, lung, and liver were taken among groups at the end of follow-up. Injection of BM-MSC did not result in obstruction of hepatic blood flow or increased pulmonary artery pressures. Blood flow in the portal vein was higher following injection BM-MSC in comparison to baseline blood flow. Arterially infused BM-MSC were more equal distributed over the different areas of the liver in comparison to the portal vein infused group (p=0,04). No BM-MSC were detected in pulmonary circulation nor in biopsies of spleen and lung. These results show that infusing therapeutic quantities of BM-MSC directly in the vascular bed of the liver is safe and feasible. Regarding risks of thrombosis or embolism, no differences between the routes of admission was observed.

Microarray analysis on livers, spleens and hearts of mice fed an atherogenic diet and supplemented with either oil palm phenolics (OPP) or distilled water

Project description:OPP (1500 ppm gallic acid equivalent (GAE)) was supplemented to BALB/c mice given an atherogenic diet for six weeks to observe for possible anti-atherogenic effects. The control group received distilled water instead of OPP. Livers, spleens and hearts were harvested six weeks after the feeding regimen for gene expression studies. Results from the separate microarray data analysis carried out on the different organs show that OPP attenuated the effects of the atherogenic diet in the organs. Total RNA obtained from livers, spleens and hearts of BALB/c mice given OPP (six weeks after administration of an atherogenic diet) were compared to controls given distilled water (liver: three replicates in the treatment group versus four replicates in the control group; spleen and heart: three replicates in the treatment group versus three replicates in the control group for both organs)
