Project description:In this study, we explored the metabolic processes of P. marinus under lipid deprived conditions to elucidate the interchanging flux of lipid and carbohydrate metabolism. Though P. marinus can synthesize their own lipids from available nutrients, they display a slower growth in media not supplemented with lipids as opposed to media with lipids. Under these conditions, using transcriptomics, we surprisingly observed evidence of stimulated lipid degradation through increased transcription of two core β-oxidation pathway enzymes.

Project description:Changes in lipid levels/profiles can reflect health status and diseases. Urinary lipidomics, thus, has a great potential in clinical diagnostics/prognostics. Previously, only chloroform and methanol were used for extracting lipids from the urine. The present study aimed to optimize lipid extraction and examine differential lipid classes obtained by various extraction protocols. Urine samples were collected from eight healthy individuals and then pooled. Lipids were extracted by six solvent protocols, including (i) chloroform/methanol (1:1, v/v), (ii) chloroform/methanol (2:1, v/v), (iii) hexane/isopropanol (3:2, v/v), (iv) chloroform, (v) diethyl ether, and (vi) hexane. Lipid profiles of the six extracts were acquired by MALDI-TOF mass spectrometry (MS) and some lipid classes were verified by LIFT-TOF/TOF MS/MS. The data revealed that phosphatidylglycerol (PG) and phosphatidylinositol (PI) could be detected by all six protocols. However, phosphatidylcholine (PC) and sphingomyelin (SM) were detectable only by protocols (i)-(iv), whereas phosphatidylserine (PS) was detectable only by protocols (iii)-(vi), and phosphatidylethanolamine (PE) was detectable only by protocols (v)-(vi). In summary, we have demonstrated differential lipidome profiles yielded by different extraction protocols. These data can serve as an important source for selection of an appropriate extraction method for further highly focused studies on particular lipid classes in the human urine.

We used lipid antigen microarrays and lipid mass spectrometry to identify bona fide lipid targets of the autoimmune response in MS brain and an animal model of MS to explore the role of the identified lipids in autoimmune demyelination. We found that autoantibodies in MS target a phosphate group in phosphatidylserine and oxidized phosphatidylcholine derivatives. Administration of these lipids ameliorated experimental autoimmune encephalomyelitis by suppressing activation and inducing apoptosis of autoreactive T cells, effects mediated by the lipids' saturated fatty-acid side chains. Thus, phospholipids represent a natural anti-inflammatory class of compounds that have potential as novel therapeutics for MS. Fig. 2A. Project description:Long wavelength Ultraviolet (UVA-1) radiation causes oxidative stress that leads to the formation of noxious substances within the skin. As a defensive mechanism skin cells produce detoxifying enzymes and antioxidants when they detect modified molecules. We have recently shown that UVA-1 irradiation oxidizes the abundant membrane phospholipid 1-palmitoyl-2-arachidonoyl-sn-glycero-3-phosphorylcholine (PAPC), which then induced the synthesis of the stress response protein heme oxygenase 1 (HO-1) in dermal fibroblasts. Here we examined the effects of UVA-1 and (UV-) oxidized phospholipids on the global gene expression in human dermal fibroblasts. We identified a cluster of genes that were co-induced by UVA-1-oxidized PAPC and UVA-1 radiation. The cluster included HO-1, glutamate-cysteine ligase modifier subunit (GCLM), aldo-keto reductases-1-C1 and -C2 (AKR1C1, AKR1C2), and interleukin 8 (IL8). These genes are members of the cellular stress response system termed âantioxidant responseâ or âPhase II detoxificationâ. Accordingly, the regulatory regions of all these genes contain binding sites for NF-E2-related factor 2 (Nrf2), a major regulator of the antioxidant response. Both UVA-1 irradiation and treatment with oxidized lipids led to increased nuclear accumulation of Nrf2. Silencing expression of Nrf2 using siRNA or using cells and tissue from Nrf2-deficient mice, we show that the induction of the co-regulated genes was suppressed. Expression of other canonical UVA-1-induced genes, including cyclooxygenase 2 (Cox2) and interleukin 6 (IL6) was unaltered in the absence of Nrf2. Together, our data show that UVA-1-mediated lipid oxidation induces induction of antioxidant response genes, which is dependent on the redox-regulated transcription factor Nrf2. To activate Nrf2 is a major strategy for novel antioxidant drugs, the skin photo-adaptation (SPA) inducers. Our finding that specific uv-oxidized lipids act similar sheds a new (ultraviolet) light on the usually detrimental âimageâ of UV generated lipid mediators. Experiment Overall Design: we profiled global mRNA expression levels in human dermal fibroblasts that had been treated with either UVA-1 or oxidized lipids. To investigate the effect of oxidized phospholipids on gene regulation, we used two preparations, which differed in their degree of oxidation; the minimally oxidized UV-PAPC resulting from UVA-1 irradiation of PAPC, and air-oxidized PAPC (OxPAPC), which represents the full spectrum of oxidation products (Gruber 07) (Reis et al., 2005). We irradiated dermal fibroblasts with UVA-1 (40J/cm²) or treated them with UV-PAPC, OxPAPC or native PAPC (100µg/ml each). We analyzed global gene expression four hours after stimulation with gene arrays (Affymetrix U133A Plus 2.0 Gene Chips).

Project description:Lipid homeostasis is dysregulated in several forms of neurodegeneration. Here we examined changes in sterols, neutral lipids and transcripts that control their homeostasis during the neuronal death induced in the CA1 region of mouse hippocampus by focal kainic acid (KA) injection. We defined changes in the lipid economy of neurons and glial cells by staining for neutral lipids and for free cholesterol. Lipid droplets filled with neutral lipids were induced in microglia at ~24 hrs after KA-treatment. At 2-4 days after injection, filipin staining revealed punctate deposits of free cholesterol in neuronal somata. These changes were correlated with alterations in cellular organelles observed in electron microscopy and transcriptomic changes obtained with RNA-seq of tissue from the CA1 region. Mitochondria, the innate inflammatory response and lipids. Lipid droplet transcriptome. Cholesterol economy. Lipids and phagocytosis.
