Human Renal Tubular Anti-Brush Border Antigen (ABBA) Discovery

ABSTRACT: Discovery of target antigen resulting in autoimmune form of renal interstitial nephritis using a brush border biotinylation, affinity purification of target antigen using patient sera, and characterization of biotinylated species in the immunopurified complex. Results were confirmed by IF of patient biopsies and IB using recombinant target antigen.

Project description:The aim of the study was to test the hypothesis that SLE patient sera contains IgG reactivity to serum factor antigens concomitant with prototypical autoimmne antigens. We diluted patient sera 1:250 and incubated dilutions on a nitrocellulose-platform array printed with whole protein antigens. In this study, serum from 45 individuals (subsetted into high and low interferon signature as well as healthy control categories) was profiled for IgG autoantibody reactivity to whole protein autoimmune and serum factor antigens. SLE and specifically IFN high patient sera was found using the significance analysis of microarrays (SAM) algorithm to be significantly more reactive with prototypical autoimmune antigens and select serum factor array antigens.

In this study, serum from 14 ndividuals (subsetted into APS-1 patient and healthy control categories) was profiled for IgG autoantibody reactivity to whole protein autoimmune and serum factor antigens. APS-1 patient sera was found using the significance analysis of microarrays (SAM) algorithm to be significantly more reactive with prototypical cytokine antigens previously identified as targets of autoantibodies.

Profiling antibody function on antigen arrays

Project description:Humoral immune responses are traditionally characterized by determining the presence and quality of antibodies specific for certain antigens. Arraying of large numbers of antigens allows the parallel measurement of antibodies, generating patterns called antibody profiles. Functional characterization of these antibodies could help draw an even more informative map of an immune response. To generate functional antibody profiles we simultaneously tested not only IgM, IgG and IgA binding to but also complement activation by a panel of endogenous and exogenous antigens printed as microarrays, using normal and autoimmune human sera. We show that complement activation by a particular antigen in a given individual cannot be predicted by the measurement of antigen specific antibodies, in spite of a general correlation between the amount of antigen-bound antibody and the deposited C3 fragments. This is due to both differences in the isotypes that dominate in the recognition of an antigen and individual variations for a given isotype, resulting in altered complement activation potential. Thus, antigen specific C3 deposition can be used as an additional parameter in immune response monitoring. This is exemplified by comparing the coordinates of antigens, used for the diagnosis of systemic lupus erythematosus, of normal and autoimmune serum samples in a two-dimensional space derived from C3 deposition and antibody binding. Since cleavage fragments of C3 mediate important immunological processes we propose that measurement of their deposition on antigen microarrays, in addition to antibody profiling, can provide useful functional signature about the tested serum. Project description:Autoimmune diseases that affect the kidney proximal tubule (PT) cause proteinuria and are implicated in the pathogenesis of kidney failure. Previous evidence demonstrated that circulating anti-brush border antibodies (ABBA) target megalin (LRP2) but whether other components of the PT drive autoimmunity was unknown. Here we investigate a patient with LRP2-negative ABBA disease, and applied immunoprecipitation, mass spectrometry, and confocal imaging to identify cubilin as a novel antigen associated with the autoimmune disorder.

Project description:The aim of the study was to test wether our microarray platform is capable of detecting previously described IgG autoantibodies to IFN-gamma. We diluted patient sera 1:250 and incubated dilutions on a nitrocellulose-platform array printed with whole protein antigens. In this study, serum from 10 individuals (subsetted into two groups: individuals positive or negative for previously described IFN-gamma-targeted autoantibodies) was profiled for IgG autoantibody reactivity to whole protein autoimmune and serum factor antigens. We found that IFN-gamma-targeted IgG autoantibodies were recognized in the IFNg-pos samples and also identified other array targets significantly associated with anti-IFNg positive individials using the significance analysis of microarrays (SAM) algorithm.

Autoimmune peptide antigen reactivity in BXD2 mice with spontaneous systemic autoimmune disease

Project description:The aim of the study was to determine autoimmune BXD2 mouse sera reactivity to linear peptide epitopes from the Immune Epitope Database. Pooled sera from six 8-10 month old BXD2 mice was diluted at 1:1000 and incubated on a PEPperPRINT peptide microarray platform printed with peptide autoantigens. In this study, pooled sera from six 8-10 month old BXD2 mice was profiled for anti-mouse IgG (H+L) analysis of autoantibody reactivity to peptide auto-epitopes printed in duplicate on a PepperPrint Chip.
