Endophyte-infected diet alters gene expression in heifer luteal tissue

Project description:Gene expression analysis in heifer luteal tissue by interspecies microarray analysis. Post-pubertal heifers were fed diets containing endophyte free fescue seed (EF), endophyte infected fescue seed (EI) or EI seed supplemented with 1.44 mg domperidone/kg body weight (EID). One heifer from each treatment group was slaughtered at a local abattoir 11-14 days after ovulation (EF = 11d, EI = 14d and EID = 14d ) and used for this study. Ovaries were collected and shipped at room temperature to the laboratory. Approximately 3 mm3 samples of luteal tissue were placed in cryovials and frozen at -80°C until processed. One 3 mm3 sample each for the EI and EID treatments and two samples for the EF treatment were ground to a fine powder in liquid N and total RNA was extracted using the RNeasy® Mini Kit (Qiagen, Valencia, CA). Quality and quantity of RNA was determined by gel electrophoresis and spectrophotometry. Five µg of total RNA was converted to cDNA and then biotin-labeled cRNA by linear amplification (CodeLink® Expression Bioarrays, Amersham Biosciences Corp, Piscataway, NJ, USA). Ten micrograms of labeled cRNA were hybridized to CodeLink UniSet Rat I Bioarray (Amersham Biosciences Corp, Piscataway, NJ, USA) by the Amersham Biosciences Facility. Keywords: other

Nocardamine-dependent iron uptake in Pseudomonas aeruginosa : exclusive implication of the FoxA outer membrane transporter

Project description:Iron is a key nutrient for almost all living organisms and paradoxically poorly soluble and consequently poorly bioavailable. To get access to this metal, bacteria have developed many different strategies. One of the most common consists of the use of siderophores, small compounds chelating ferric iron with a very high affinity. Many bacteria are able to produce their own siderophores or use those produced by other microorganisms (exosiderophores) in a piracy strategy. Pseudomonas aeruginosa produces two siderophores, pyoverdine and pyochelin and is able to use a large panel of exosiderophores. We investigated the ability of P. aeruginosa to use nocardamine (NOCA) and ferrioxamine B (DFOB) as exosiderophores under iron-limited planktonic growth conditions. Proteomic and RT-qPCR approaches showed an induction of the transcription and expression of the outer membrane transporter FoxA in the presence of NOCA or DFO in the bacterial environment. Expression of the proteins of the heme or pyoverdine and pyochelin dependent iron uptake pathways were not affected in the presence of these two tris-hydroxamate siderophores. 55Fe uptake assays using foxA mutants demonstrated that ferri-NOCA was exclusively transported by FoxA, while ferri-DFO was transported by FoxA and at least one another unidentified transporter. The crystal structure of FoxA in complex with NOCA revealed very similar siderophore binding sites between NOCA and DFO. Iron uptake by hydroxamate exosiderophores in P. aeruginosa cells is discussed in the light of these results.

The role of FoxA, FiuA, and FpvB in iron acquisition via hydroxamate-type siderophores in Pseudomonas aeruginosa

Project description:Siderophores are specialized molecules with different chemical structures, produced by bacteria and fungi to scavenge iron from the environment, a crucial nutrient for their growth and metabolism. These iron-chelating compounds enable bacteria to overcome iron limitation, a key factor in microbial survival and pathogenesis. Catecholate-type siderophores are primarily produced by bacteria, while hydroxamates are predominantly produced by fungi. The capacity of nine hydroxamate-type siderophores produced by fungi to serve as siderophores for iron acquisition by Pseudomonas aeruginosa, a human pathogen, has been investigated. Growth assays under iron limitation and 55Fe incorporation tests clearly highlighted that all nine siderophores promoted bacterial growth and facilitated iron transport. Additionally, the study aimed to identify the TonB-dependent transporters (TBDTs) responsible for iron import mediated by the tested siderophores. Mutant strains lacking genes encoding TBDTs were employed, revealing that iron is imported into P. aeruginosa cells solely by FpvB for the siderophores coprogen, triacetylfusarinine C, fusigen, ferrirhodin, and ferrirubin siderophores. Iron complexed by desferioxamine G is imported by two TBDTs, FpvB and FoxA. Ferricrocin-Fe and ferrichrycin-Fe complexes are imported by FpvB and FiuA. Lastly, rhodotorulic acid-Fe complexes are imported by FpvB, FiuA, and another unidentified TBDT. In conclusion, the data illustrate the effectiveness of hydroxamate-type siderophores in transporting iron into P. aeruginosa cells and provide insights into the intricate molecular mechanisms involved in iron acquisition, which have implications for understanding bacterial pathogenesis and developing potential therapeutic strategies.
